BACKGROUND: Pesticides are extensively applied globally. Pesticide residues induce calamitous effects on the environment and untargeted organisms. Public concerns for the safety of freshwater organisms and the challenges posed by aquatic contaminants remain high. In the present study, the acute toxicity of avermectins (AVMs) to the crayfish, Procambarus clarkii was evaluated. We also evaluated the potential effects of AVM on the biochemical and transcriptomic status of the hepatopancreas and gastrointestinal tract in P. clarkii. RESULTS:The 24, 48, 72, 96 h median lethal concentrations (LC 50 ) of AVM on crayfish were 2.626, 1.162, 0.723, 0.566 mg L −1 , respectively. The crayfish were then exposed to 0.65 mg L −1 of AVM for 96 h. AVM significantly altered biochemical parameters including AChE and CAT activities in the hepatopancreas, and AChE, SOD and Na + -K + -ATPase activities in the gastrointestinal tract at several time points. Furthermore, transcriptomic analysis identified 953 and 1851 differentially-expressed genes (DEGs) in the hepatopancreas and gastrointestinal tract, respectively. KEGG enrichment showed that the gene expression profiles of the hepatopancreas and gastrointestinal tract were distinct from each other. The DEGs in the hepatopancreas were mostly enriched with stress-response pathways, while the majority of the DEGs in the gastrointestinal tract belonged to metabolism-related pathways.CONCLUSION: We demonstrated that the AVM induced acute toxicity, oxidative stress, osmoregulation disturbance, neurotoxicity and transcriptome imbalance in crayfish. These findings unraveled the detrimental effects of AVMs exposure on crayfish.

Introdução: Arboviroses transmitidas pelo Aedes aegypti têm grande relevância para a saúde pública, sendo o controle químico do vetor uma importante estratégia de mitigação. Entretanto, o uso intensivo de pesticidas está associado a seleção de insetos resistentes e impacto ambiental. Por isso, é essencial desenvolver e regulamentar novos produtos, eficazes e sustentáveis. Objetivo: Descrever os requisitos para regulação de inseticidas e repelentes no Brasil, à luz de orientações internacionais. Comparar os ativos regulamentados no país com os disponíveis nos Estados Unidos (EUA) e União Europeia (UE). Por fim, realizar um levantamento de estudos ecotoxicológicos dos ativos empregados no controle vetorial. Método: Revisão narrativa de instrumentos regulatórios nacionais e internacionais, literatura científica, consulta a agências regulatórias e ao ECOTOX Knowledgebase. Resultados: As normas brasileiras trazem exigências consoantes com as internacionais. Todos os ativos disponíveis nos EUA têm monografia aprovada pela Anvisa; já a UE mostrou-se mais restritiva. Há menos repelentes tópicos disponíveis no Brasil e na UE, comparando-se com EUA. Muitos dos inseticidas aprovados para controle vetorial representam risco para organismos não alvo. Ainda assim, não identificamos quaisquer estudos para alguns dos produtos no banco de dados pesquisado. Conclusões: A UE mostrou-se a agência mais restritiva com relação ao registro de inseticidas. Existe a necessidade de guias com orientações específicas sobre o desenvolvimento de produtos inovadores para o controle do mosquito, bem como de ferramentas de busca com maior transparência na página da agência brasileira. Finalmente, esta revisão reitera a importância de análises ecotoxicológicas mais abrangentes para os produtos em questão, considerando seu potencial dano ambiental.

BACKGROUND: Balancing the safety and efficiency of chiral pesticides can help protect pollinators. We evaluated the stereoselective behavior, bioactivity, toxicity and exposure risk of the chiral insecticide pyriproxyfen in a citrus nectar system. RESULTS: Density functional theory (DFT) and ultra-performance liquid chromatography tandem mass spectroscopy (UPLC-MS/ MS) were applied for absolute configuration appraisal and chiral analysis validation, respectively. The recoveries ranged from 72.3% to 100.5% with an relative standard deviation (RSD) ranging from 1.2% to 9.7%. In a field trial, we determined insecticide half-lives in citrus leaves and flowers, which were 7.0 and 8.6 days for R-(+)-pyriproxyfen, and 11.7 and 14.7 days for S-(−)-pyriproxyfen, respectively. We found that the bioactivity of R-(+)-pyriproxyfen was 3.39 and 2.37 times higher than S-(−)-pyriproxyfen against Unaspis yanonensis and Diaphorina citri nymphs, respectively. S-(−)-pyriproxyfen had 3.8 times higher acute toxicity than R-(+)-pyriproxyfen on Apis mellifera L., and its exposure risk was moderate based on the hazard quotient.CONCLUSION: The phenomenon of stereoselective degradation and biological effect demonstrated that the high-risk stereoisomer of S-(−)-pyriproxyfen degraded more slowly than R-(+)-pyriproxyfen, but R-(+)-pyriproxyfen with better efficiency for target. Therefore, an increased duration of R-(+)-pyriproxyfen activity on citrus was beneficial for efficacy. Our results could guide the scientific application and evaluation of chiral pesticides on nectar plants.
