Improving pineapple micropropagation protocol through explant size and medium composition manipulation.Abstract -Introduction. Although several pineapple micropropagation protocols have already been published, significant improvement could be achieved if the stages of the in vitro culture were better defined. Our work concerned several experiments aiming at the mass production of high quality plantlets. Materials and methods. Axillary buds were inoculated on an MS liquid culture medium added with NAA (2 µM) and BAP (4 µM). Regenerated shoots, divided into three classes of different sizes, were then used in further experiments. First, these shoots were inoculated in flasks containing the same MS culture medium with or without growth regulators. Then, four basic media, containing different salts and free from growth regulators were tested. In a third assay, the MS culture medium was compared with a half-diluted MS culture medium for studying the plantlet elongation and rooting stage. Results. In the MS culture medium supplemented with NAA and BAP, the highest multiplication rate (13.5 shoots) was obtained with the smallest shoots inoculated, while in the MS culture medium free of growth regulators, the highest plantlets (7.7 cm) were the result of the highest shoots inoculated and showed no vitrification. The normal MS culture medium, in comparison with the half-diluted one and the three other salt formulations, revealed a significant increase in the plantlet elongation and best general features. For acclimatization, the highest values of the survival rate (93.8%) and fresh and dry weights were obtained with the transference of higher than 7.0 cm in vitro plantlets. Conclusion. Using the protocol described in this work, it is possible to obtain 1 million in vitro plantlets after 9 months from a single bud, with a 45 day subculture interval and an average multiplication rate of 10 shoots per bud.Brazil / Ananas comosus / plant propagation / micropropagation / in vitro experimentation / culture media / transplanting / plant establishment Amélioration du protocole de micropropagation de l'ananas par modification de la taille de l'explant et de la composition du milieu de culture.Résumé -Introduction. El enraizamiento se realizó ex vitro se plantaron en materas en un medio preparado con tierra de hormiga, arena fina y musgo en proporciones iguales (1:1:1), desinfestadas con formaldehido un mes antes del iniciado el proceso. Resultados: A los 90 días, el número de brotes y tamaño de los brote por explantes fue: cultivar 1 C1 (78,4±05) brotes y longitud del brote (2,5±0,03 cm); cultivar 2 C2: (85,7±0,5) brotes y longitud del brote (2,1±0,11 cm), Cultivar3 C3 (92,8±0,1) brotes y longitud del brote (1,6±0,07 cm). La mejor combinación de los reguladores de crecimiento fue de 1 mg/l de ANA y 3 mg/l de 6-BAP, lo cual indica que es necesaria la presencia de auxina y citoquinia para la brotación. El enraizamiento fue ex vitro y a los 90 días las plantas tenían en promedio de C1 (5,8±0,4) raíces y longitud de raíz (3,2±0,3 cm); C2 (5,4±0,1) raíces y longitud de raíz (3,9±0,8 cm); C3 (5,7±0,7) raíces y longitud de raíz (3,8±0,5 cm).Palabras clave: Cultivares, Cultivo in vitro, Micropropagación, Piña, Propagación masiva. Methodology: in vitro multiplied buds of apices extracted from crown and basal children cultured in semisolid Murashige and Skoog initiation medium (MS, 1962) containing: 4 mg. l-1 thiamine HCl, 0,5 mg/l NAA, 0,5 mg/l IBA and 0,5 mg/l 6-BAP and pH adjusted to 5,8; 10 g/l activated charcoal, and then a multiplication medium with four hormone treatments; (1) 0,3 mg/l of naphthalene acetic acid (NAA)+1,0 mg/l of benzylaminopurine (6-BAP), (2) 0,5 mg/L NAA+1,5 mg/l BAP, (3) 1,0 mg/l NAA+3,0 mg/l 6-BAP, (4) 1,0 mg/l NAA + 3,0 mg/l 6-BAP+2 mg/l of gibberellic acid (GA3). Rooting ex vitro was performed were planted in pots in a medium prepared with soil from ant, fine sand and moss in equal proportions (1:1:1), disinfested with formaldehyde a month before the process starts. Results: At 90 days, the number of shoots and shoots size of explants was: C1 (78,4±0,5) shoots and shoots length (2,5±0,03 cm); C2: (85,7±0,5) shoots and shoots length (2,1±0,11 cm), C3 (92,8±0,1) shoots and shoots length (1,6±0,07 cm). The best combination of growth regulators was 1 mg/l NAA and 3 mg/l de 6-BAP.lo indicating that the presence of auxin and citoquinia necessary for sprouting.
