Discrimination Between Celery Cultivars with the Use of Rapd Markers

Abstract: Discrimination Between Celery Cultivars with the Use of Rapd MarkersScreening of celery cultivars with random amplified primers is a way of avoiding mistakes in distinguishing similar genotypes. Six primers showed distinct polymorphism between the studied cultivars. The number of bands varied from 7 to 13. Suitable primers generated 52 markers of which 22 were polymorphic. A similarity matrix was created using Jaccard's coefficient. The group average method was employed to construct a dendrogram. Based on RAPD… Show more

“…However, the mean genetic similarity of the celery e´lite variety group detected in this study was similar to that of the celeriac group of varieties examined by Muminović et al (2004) (J = 0.92 vs J = 0.90). These high genetic similarity values support the opinion (Muminović et al 2004;Domblides et al 2008;Wang et al 2011) that recent breeding activities have restricted the genetic base of the crop.…”

“…Among the available molecular techniques already used in celery (Quiros et al 1987;Huestis et al 1993;Yang and Quiros 1993;Muminović et al 2004;Acquadro et al 2006;Domblides et al 2008;Wang et al 2011), we chose the amplified fragment length polymorphism (AFLP) technique which was proved to be effective in distinguishing celery from celeriac and related species (Muminović et al 2004). Since our analysis was aimed at characterizing a local population, we decided to couple AFLP with bulk analysis.…”

“…Yang and Quiros [8] first used random amplified polymorphic DNA (RAPD) markers in celery to distinguish different cultivars. Domblides [9] did the similar research using RAPD markers. Then amplified fragment length polymorphism (AFLP) markers were used in celery for cultivar classification and genetic diversity analysis [10], [11].…”
