The ''Sedano Nero di Trevi'' (Black Celery from Trevi) is a landrace grown in Umbria (Italy) at risk of extinction. A morpho-physiological and molecular characterisation of the landrace and a detailed investigation of its cultural and cultivation context were carried out with the aim of facilitating a request for a quality mark, implement in situ (on farm) conservation schemes that are being developed in Italy and register the landrace in the European common catalogue of 'conservation varieties'. These actions can help the landrace survive. Six farmer populations of the landrace and four e´lite varieties were characterised for thirteen morpho-physiological traits and nine AFLP primer combinations. The ''Sedano Nero'' farmer populations were better performers than e´lite varieties for important agronomic traits, such as yield, under the local agronomic conditions. Both morphophysiological traits and molecular markers clearly distinguished the landrace from the e´lite cultivars. In addition, a structure was detected in the landrace with farmer populations being characterised by specific alleles. These results are discussed in connection with the local seed system and possible in situ conservation strategies for landraces.

BackgroundCelery is an increasing popular vegetable species, but limited transcriptome and genomic data hinder the research to it. In addition, a lack of celery molecular markers limits the process of molecular genetic breeding. High-throughput transcriptome sequencing is an efficient method to generate a large transcriptome sequence dataset for gene discovery, molecular marker development and marker-assisted selection breeding.Principal FindingsCelery transcriptomes from four tissues were sequenced using Illumina paired-end sequencing technology. De novo assembling was performed to generate a collection of 42,280 unigenes (average length of 502.6 bp) that represent the first transcriptome of the species. 78.43% and 48.93% of the unigenes had significant similarity with proteins in the National Center for Biotechnology Information (NCBI) non-redundant protein database (Nr) and Swiss-Prot database respectively, and 10,473 (24.77%) unigenes were assigned to Clusters of Orthologous Groups (COG). 21,126 (49.97%) unigenes harboring Interpro domains were annotated, in which 15,409 (36.45%) were assigned to Gene Ontology(GO) categories. Additionally, 7,478 unigenes were mapped onto 228 pathways using the Kyoto Encyclopedia of Genes and Genomes Pathway database (KEGG). Large numbers of simple sequence repeats (SSRs) were indentified, and then the rate of successful amplication and polymorphism were investigated among 31 celery accessions.ConclusionsThis study demonstrates the feasibility of generating a large scale of sequence information by Illumina paired-end sequencing and efficient assembling. Our results provide a valuable resource for celery research. The developed molecular markers are the foundation of further genetic linkage analysis and gene localization, and they will be essential to accelerate the process of breeding.

Celery (Apium graveolens L.) has been known as traditional medicine. Apigenin is one of active component in Celery leaf that has fungicidal activity, so celery leaf's extortion suspected has fungicidal activity to Candida albicans. Apigenin can solved in water. The aim of this research was to know antifungal activity of celery leaf's extortion againsts Candida albicans in vitro. This is a laboratory experimental research by using completely randomized design and disc diffusion method. Celery leaf's extortion was divided into 3 concentrations, there were 25%, 50% and 100%. Ketoconazole 2% was used as positive control and aquadest as negative control. The result of this research was based on analysis of variance, there was significant difference between minimal 2 treatment (p=0.000) and confirmed with Post Hoc test, there were significant difference in all treatments. As the conclusion, celery leaf's extortion had antifungal activity againsts Candida albicans in vitro with the best concentration 50% but smaller than ketoconazole 2%.
