Background: Recent studies have shown that depression is inversely correlated with high protein and low fat intake and positively correlated with vitamin D-binding protein (VDBP). Therefore, the aim of this study was to examine the interaction between protein/fat dietary patterns and VDBP genotypes with regard to the risk of depression in apparently healthy adults who have not been diagnosed with any chronic disease. Methods: In this study, 265 individuals (126 males and 139 females) aged 18–55 years were recruited from the communities of central and west Tehran based on convenience sampling. Body composition was measured with a body composition analyzer and depression symptoms were categorized as normal, moderate depression, or severe depression using the Depression Anxiety Stress Scales 21 (DASS-21) questionnaire. Dietary patterns were determined by a semiquantitative food frequency questionnaire to assess typical food intake during the 12-month period. Blood samples were collected from and biochemical measurements performed on all participants. An analysis of two polymorphisms (rs7041 and rs4588) in the GC gene, which encodes VDBP, was performed by polymerase chain reaction-restriction fragment length polymorphism. Results: A statistically significant association was found between depression and diet (p = 0.03) after having categorized the participants into three groups: a high-protein/low-fat (HP/LF) group, a moderate-protein/moderate-fat (MP/MF) group, and a low-protein/high-fat (LP/HF) group. Moreover, the findings demonstrated that depression was related to both the rs7041 and the rs4588 polymorphism (p = 0.05 and p = 0.02, respectively). We next used multinomial logistic modeling to investigate the risk of depression. A significant interaction was observed between HP/LF diet and the rs7041 polymorphism in the moderate- and severe-depression groups (β = –0.30, p = 0.05, and β = –0.48, p = 0.01, respectively). Conclusion: This study showed that an HP/LF diet interacts with the rs7041 polymorphism, with T allele carriers having a greater prevalence of moderate and severe depression.

Palm oil (PO) and olive oil (OO) have been widely used in animal diets, but the knowledge of metabolic change in animals fed a diet with different oil sources is limited. Here, we compared the metabolic differences in the Chinese mitten crab fed with PO and OO as the main dietary lipid sources by performing GC‐MS‐based serum metabolomics assays and traditional nutritional assessments. The crab fed OO displayed lower lipid accumulation and oxidative stress in the hepatopancreas than those in the crab fed PO. In the metabolomics assay, 68 metabolites with high credibility were identified and five metabolites were significantly different between two feeding groups. In these five metabolites, hydroxylamine, 3‐hydroxypropionic acid and 2‐hydroxypyridine were higher in the OO group, while lysine and citrulline were higher in the PO group. We demonstrate that olive oil can provide comprehensive benefits to the crab by providing more energy, improving cell membrane structure, containing phenols as a natural antioxidant, and improving the composition of intestinal microbiota. On the contrary, palmitic acid‐enriched palm oil tended to increase protein degradation and lipid accumulation‐induced lipotoxicity. This study illustrates that the metabolic mechanism differs between crabs fed olive oil and palm oil, and OA‐enriched olive oil is recommended as an ingredient for crab diet formulation.

There is a constant interest in blood‐based protein biomarkers, which can help to improve diagnosis and treatment outcomes of multifactorial human pathologies. In this regard, proteomic studies usually employ plasma immunoaffinity fractionation to deplete the most abundant plasma proteins, due to the high dynamic concentration range of proteins. The depletion of high abundant proteins allows to obtain less abundant and, oftentimes, more interesting proteins. However, the removal of the fraction of the high abundant plasma proteins ‐ the depletome ‐ may co‐elute many unintended proteins due to protein‐protein interactions. Little data is available about the depletome and potential protein biomarkers may be lost during this process. To visualize and characterize these proteins, we analyzed the depletome of 20 plasma samples by shotgun mass spectrometry‐based proteomics. Thus, using immunoaffinity depletion followed by 2‐D liquid chromatography coupled to an ion mobility‐enhanced mass spectrometer, our analysis identified that over 100 proteins are co‐eluting with the high abundant fraction. These proteins play roles in several biological processes, such as receptor‐mediated endocytosis, complement activation, and regulation of immune response. This study supports that investigating the depletome is important in the quest for biomarkers.
