Dexketoprofen trometamol loaded solid lipid nanoparticles (SLNs): Formulation, in vitro and in vivo evaluation

Abstract: Development, in vitro and in vivo evaluation of dexketoprofen trometamol (DT)-loaded nanosized drug delivery system was aimed in this study. DT-loaded solid lipid nanoparticles (SLNs) were prepared using probe sonication. DT release from SLNs prepared and their kinetics were investigated. Structures of SLNs were elucidated by particle size and zeta potential measurements, shape and surface imaging, thermal analysis, X-ray diffraction, FT-IR and 1 H-NMR determinations. DT-loaded particles demonstrated character… Show more

“…Polydispersity index (PDI) indicates the range of particle size distribution, and PDI values in the range of 0 -1 demonstrate homogeneous distribution when it approaches zero. Very low PDI values of prepared formulation indicate homogeneous particle size distribution [13,20]. Zeta potential (ZP) values of PLGA NPs were found to be negative most probably due to the total negative charges of the functional groups on PLGA.…”

“…Higher CH 2 , CH 3 and CH signals in the copolymer caused those peaks to expand. Specific peaks of DT in 1 H-NMR spectrum were observed at 1.45 (d, 3H) ppm, 3.64 (s, 6H) ppm, 3.66 (q, 1IH) ppm and 7.41-7.80 (m, 9H) ppm [13].…”

“…Zeta potential value was found to be around +40 mV, indicating that the NPs have good stability. Higher Zeta potential is expected for the stable colloidal system as it overcomes the particle aggregation due to repulsion forces [24]. Positive surface charge of NPs were attributed to the cationic nature of Eudragit ® polymers [23,24].…”
