Development of an Improved Reverse-Phase High-Performance Liquid Chromatography Method for the Simultaneous Analyses of trans-/cis-Resveratrol, Quercetin, and Emodin in Commercial Resveratrol Supplements

Abstract: The objectives of the study were to develop a reverse-phase high-performance liquid chromatography method for the simultaneous analyses of trans-/cis-resveratrol, emodin, and quercetin and to determine the concentrations of these polyphenols in 28 resveratrol supplements. Samples were separated within 15 min in a C18 reversed-phase column using mobile phases containing 0.1% formic acid and methanol/0.1% formic acid. The calibration graphs for all four compounds were linear from 0.1 to 410 μg/mL (r2=0.99). The … Show more

“…The use of methanol as solvent in the mobile phase has been reported to increase considerably the response when the instrument is operating in positive ionization mode (ESI+) [ 47 ]. In the most of methods to determine and quantify polyphenols based on LC-MS or MS/MS [ 34 , 37 , 42 , 48 , 49 , 50 ], solvent B consisted of acetonitrile or methanol, but here we achieved the best chromatographic resolution using a mixture 70:30 acetonitrile:methanol as solvent B.…”

“…According to different chromatographic retention behavior, compound 7 was confirmed as trans ‐polydatin, while compound 16 corresponded to cis ‐polydatin. Similarly, compounds 14 and 25 were respectively identified as trans ‐resveratrol and cis ‐resveratrol . Compared with the standards and literature data, compounds 10, 12 and 24 were attributed to forsythoside A, pinoresinol‐β‐ d ‐glucoside and phillyrin, respectively .…”

“…One of the best known examples is the laxative emodin that is often copurified with and contaminates resveratrol extracts from Japanese knotweed (Polygonum cuspidatum Siebold & Zucc.) [74]. Furthermore, despite all the progress made in the field of metabolic engineering of plants and plant cell factories for increased production of native secondary metabolites, their application as production hosts for polyphenols is still limited [75][76][77][78][79].…”
