A Focused Multiple Reaction Monitoring (MRM) Quantitative Method for Bioactive Grapevine Stilbenes by Ultra-High-Performance Liquid Chromatography Coupled to Triple-Quadrupole Mass Spectrometry (UHPLC-QqQ)

Abstract: Grapevine stilbenes are a family of polyphenols which derive from trans-resveratrol having antifungal and antimicrobial properties, thus being considered as phytoalexins. In addition to their diverse bioactive properties in animal models, they highlight a strong potential in human health maintenance and promotion. Due to this relevance, highly-specific qualitative and quantitative methods of analysis are necessary to accurately analyze stilbenes in different matrices derived from grapevine. Here, we developed … Show more

“…The final steps involve the stepwise reactions that promote the decarboxylation, dehydration, and enolization of stilbene-2-carboxylic acid to yield the resveratrol molecule [ 52 ]. Resveratrol can undergo other biochemical reactions to produce new stilbenes, including ε -viniferin, t-piceid, t-piceatannol, and t-pterostilbene [ 53 ].…”

“…LC–MS/MS is a rapidly developing technique and has raised bioanalysis to a new level. Multiple reaction monitoring (MRM) based on LC–MS/MS (LC–MRM) is a “gold standard” for the bioanalysis of small molecules [168–181]. MRM assay is a target MS technique based on ESI–MS [182–191].…”

“…To prepare a sample, an extraction is carried out using different solvents and concentrates. The best option for extracting from grapes is alcohol mixed with water, and in the case of wine, it is a concentrated using Amberlite resin (Hurtado-Gaitán et al, 2017;López-Hernández et al, 2016;Decendit, 2017). The interest in studying stilbenes is great due to its antioxidant properties.…”
