Determination of 1-Deoxynojirimycin in Mulberry Leaves Using Hydrophilic Interaction Chromatography with Evaporative Light Scattering Detection

Abstract: A simple and rapid method for determining 1-deoxynojirimycin (DNJ), a potent glucosidase inihibitor present in mulberry leaves (Morus alba and Morus bombysis), by high-performance liquid chromatography coupled to an evaporative light scattering detector (ELSD) has been developed. DNJ was separated from an extract of mulberry leaves on a TSKgel Amide-80 column, which is a representative column for hydrophilic interaction chromatography. During postcolumn detection, DNJ was detected by ELSD and concurrently iden… Show more

“…[1][2][3] The most important substance in mulberry leaves, 1-deoxynojirimycin (1-DNJ), is known to be a strong intestinal -glucosidase inhibitor. Mulberry leaves contain 1-DNJ, 4) so that mulberry leaf products have been commercialized as health foods.…”

“…However, even these columns do not achieve reasonable retention of DNJ, and for this reason, several other methods have been developed for determining DNJ in mulberry. Kimura et al reported a method that used hydrophilic interaction liquid chromatography (HILIC) coupled with an evaporative light-scattering detector (ELSD) (Kimura et al, 2004); This method is known as the HILIC-ELSD method. HILIC has been developed as an efficient tool for analyzing highly hydrophilic compounds, and its analytical application to carbohydrates (Alpert et al, 1994) and peptides (Alpert et al, 1990) has been reported.…”

The residue was accurately weighted, dissolved in 5 ml of ethanol, and a portion (5 ml) of the solution was subjected to hydrophilic interaction chromatography (HILIC) 6) coupled with tandem mass spectrometry (MS/MS) 14) for DNJ determination. The HILIC-MS/MS apparatus consisted of a liquid chromatograph (Shimadzu, Kyoto, Japan) and an API 3200 MS/MS (Applied Biosystems, CA, USA).…”

“…3,4) The future use of mulberry DNJ can therefore be anticipated, but the amount of the key compound in mulberry leaves is as low as only 0.1%. [5][6][7] Hence, more effective extraction of mulberry DNJ has become one of the important food-processing challenges.…”
