Detecção de genes de virulência e grupos filogenéticos de amostrasEscherichia coli isoladas de cães no Brasil INTRODUCTIONEscherichia coli are a component of normal intestinal microbiota in humans and other animals. Phenotypic and genotypic characteristics allow the identification of E. coli pathogenic strains or pathovars (COURA et al., 2014). Dogs are colonized by E. coli during the first days of their life and some strains can cause enteric or extra-intestinal infections (BEUTIN, 1999 (BEUTIN, 1999). Other pathovars are associated with diarrhea in other animals, namely enteroaggregative E. coli (EAEC) and enteroinvasive E. coli (EIEC) (MAINIL, 2013). Because of the close contact with humans, dogs could increase the risk of transmitting potentially zoonotic microorganisms, such as EPEC and STEC (BEUTIN, 1999).In addition to the pathotype and pathovar classification, E. coli strains can be assigned to one of the seven phylogenetic groups, including A, B1, B2, C, D, E, and F (CLERMONT et al., 2013). E. coli genomic structure has shown that the strains belonging to different phylogroups are associated with the disease state and source of isolation (CLERMONT et al., 2013). The extraintestinal pathogenic E. coli (ExPEC) strains are clustered mostly in groups B2 and D and intestinal pathogenic E. coli are mostly in phylogroups A, B1, and E (ESCOBAR- PÁRAMO et al., 2004; CLERMONT et al., 2011). Although phylogenetic characterization is an important tool to improve the understanding of E. coli populations and the relationship between strains and disease (CLERMONT et al., 2011;COURA et al., 2015), few studies have determined phylogenetic groups of E. coli isolated from dogs worldwide, and of those that did, the most frequently used method was a triplex PCR method developed by CLERMONT in 2000. This method is only capable of determining phylogroups A, B1, B2, and D (HARADA et al., 2012;SALVARANI et al., 2012;SCHMIDT et al., 2015) instead of the seven phylogroups that were detected in this study. In Brazil, only two studies identified E. coli pathovars associated with diarrhea in dogs (ALMEIDA et al., 2012;PUÑO-SARMIENTO et al., 2013), but none determined the phylogenetic groups of E. coli. Therefore, this study aimed to detect the virulence genes, pathovars, and phylogenetic groups of E. coli strains isolated from the feces of dogs with and without diarrhea. MATERIALS AND METHODS Stool samplesStool samples were collected from 154 dogs, of which 92 were diarrheic and 62 were without diarrhea. Samples from diarrheic dogs were obtained directly from the rectum, at the Veterinary Hospital of Universidade Federal de Minas Gerais (Belo Horizonte city), upon admission. Collections were performed from dogs that were undergoing a consultation for the occurrence of diarrhea. Um total de 127 cepas de Escherichia coli foi isolado de suínos no Distrito Federal, testado para a presença de genes de enterotoxinas (STa, LT-I, LT-II, Stx1 e Stx2) e para resistência antimicrobiana. Das cepas isoladas, oito (6,3%) possuíam genes para enterotoxinas, sendo quatro (3,2%) positivas somente para LT-I, três (2,4%) somente para STa e uma (0,8%) positiva para STa e LT-I. Nenhuma das cepas isoladas apresentou genes para LT-II, Stx1 ou Stx2. Quanto ao perfil de resistência antimicrobiano, os antibióticos com maiores porcentagens de resistência foram lincomicina (100%), sulfonamidas (74,8%) e tetraciclina (70,1%), enquanto os maiores índices de sensibilidade foram observados na norfloxacina (82,7%), gentamicina (75,6%) e sulfametoxazol + trimetoprim (63%). Esses resultados demonstraram a presença de genes de enterotoxinas e altas taxas de resistência antimicrobiana em E. coli isoladas de suínos hígidos no DF.

In spite of the substantial role of Clostridium difficile in causing diarrhea in piglets, there have been few reports of the disease caused by this bacterium in Rio Grande do Sul, Brazil. In this paper, we describe an outbreak of C. difficile-associated diarrhea in a pig farm in Brazil. The diarrhea rate increased in piglets 1-to-7 days old from an average of 2% to approximately 20%. Necropsied piglets showed mesocolon edema, and in a histological evaluation, severe necrotizing neutrophilic colitis was observed. The intestinal contents were positive for the A/B toxins of C. difficile and negative for other tested enteropathogens. The association between the clinical signs, post mortem findings and laboratory exams confirmed the diagnosis of C. difficile-associated diarrhea. The present report confirms C. difficile as a pathogen in swine in Brazil and highlights the need for up to date routine laboratory protocols for the diagnosis of this disease in swine. Key words: Neonatal diarrhea, colitis, enteritis ResumoApesar da crescente importância de Clostridium difficile como causador de diarréia em leitões, são escassos os estudos e relatos da doença no Brasil. O objetivo do presente trabalho é descrever um surto de diarréia por C. difficile em uma granja localizada no Rio Grande do Sul, Brasil. A frequência de diarréia em leitões com até 7 dias de vida aumentou de uma média de 2% para aproximadamente 20%. Leitões necropsiados apresentavam edema de mesocólon e, na avaliação histopatológica, uma colite neutrofílica necrotizante severa foi observada. O conteúdo intestinal foi positivo para as toxinas A e B e negativo para outros patógenos pesquisados. A associação dos sinais clínicos, achados post mortem e resultados dos exames laboratoriais permite confirmar o diagnóstico de diarréia por C. difficile. O presente relato confirma C. difficile como um patógeno de suínos no Brasil e destaca a necessidade de inclusão desse enteropatógeno no diagnóstico laboratorial para as diarréias em leitões.
