In the species Olea europaea L. the tissue culture techniques have not been sufficiently studied by the lack of efficient establishment of protocols for various cultivars. The objective of this work was to evaluate the effect of different culture media and sampling times of explants on in vitro establishment of six cultivars of olive (Ascolano 315, Leccino, Maria da Fé, Coratina, Arbequina and Frantoio). The work was divided into two experiments conducted in the Fruit Tree Propagation Laboratory, Crop Science Department, Faculty of Agronomy Eliseu Maciel, Federal University of Pelotas, Brazil. Stock plants of the olive cultivars maintained in semi- hydroponic system were used. In the first experiment, nodal segments were used to establish in vitro culture medium MO and WPM, consisting of salts and vitamins. In the second experiment four seasons to obtain the explants were evaluate (autumn, winter, spring and summer). The WPM medium promoted greater establishment of olive explants. There is a higher phenolic oxidation in olive explants collected in winter. Spring is indicated for collecting explants, therefore favors the in vitro cultivar Maria da Fé, while Ascolano 315 and Arbequina cultivars have a higher rate of in vitro establishment by collecting explants over the year

No entanto, Brondani et al (2010) observaram altas taxas de contaminação na introdução de brotações de Liquidambar styraciflua, mesmo utilizando soluções desinfestantes, o que indicou, segundo os autores, necessidade de maior prudência no controle asséptico utilizado. Resumo -Objetivou-se avaliar procedimentos para introdução in vitro de brotações retiradas de mudas de candeia mantidas em viveiro. Para a desinfestação das brotações, testou-se tempos de imersão (5, 10, 15 e 20 min) em hipoclorito de sódio a 2,5% e a influência do uso de fungicida antes (7 e 15 dias) e após a coleta das brotações, nas concentrações de 0,5 e 1,0 g L -1 respectivamente. Para o controle da oxidação dos explantes in vitro, avaliou-se a adição de polivinilpirrolidona (PVP) e carvão ativado ao meio de cultura, ambos nas concentrações 1 e 2 g L -1 . O uso de hipoclorito de sódio permitiu a desinfestação das brotações introduzidas, porém não houve diferença significativa entre os tempos testados. A coleta de brotações 15 dias após a aplicação de fungicida nas mudas fornecedoras das brotações resultou em ausência de contaminação. A oxidação dos explantes atingiu altos níveis, mesmo na presença de PVP e carvão ativado no meio de cultura.Abstract -The objective of this study was to evaluate the procedures for introduction of shoots in vitro taken from seedlings kept in nursery. For the disinfestation of the shoots, immersion times (5, 10, 15 and 20 min) in 2.5% sodium hypochlorite were tested. We also analysed the influence of using fungicide before (7 and 15 days) and after the shoots collection, in the concentrations of 0.5 and 1.0 g L -1 respectively. In order to control the oxidation of explants in vitro, the addition of poly vinyl pyrrolidone (PVP) and activated charcoal to the culture medium, both at concentrations 1 and 2 g L -1 , were evaluated. The use of sodium hypochlorite allowed disinfestation of the introduced shoots, but there was no significant difference between the times tested. The collection of shoots 15 days after the application of fungicide in the seedlings suppliers of the shoots, allowed the absence of contamination. The oxidation of the explants reached high levels even in the presence of PVP and activated charcoal in the culture medium.

Este estudo teve por objetivo desenvolver uma metodologia de multiplicação in vitro para Xylopia aromatica, visando a produção de mudas, avaliando-se meios de cultura e concentrações de reguladores de crescimento mais adequados. Foram utilizados, como explantes, segmentos nodais, provindos de brotações de mudas produzidas via seminal e mantidas em viveiro. Foram testados os meios de cultura Murashige Skoog (MS) e Wood Plant Medium (WPM) suplementados com 800 mg L-1 de polivinilpirrolidona (PVP), 30 g L-1 de sacarose, 0,054 µM de ácido naftalenoacético (ANA) e 6 g L-1 de ágar e concentrações de 2,22 e 3,55 µM de benzilaminopurina (BAP) para a multiplicação in vitro. Aos trinta dias, avaliou-se o número de brotações laterais emitidas e o percentual de explantes formando calos na base. As maiores taxas de multiplicação foram obtidas com o meio MS acrescido de 3,55 µM de BAP. Em relação à formação de calos na base dos explantes, o meio MS promoveu maior formação quando comparado ao WPM, no entanto não comprometeu a formação de brotos. Foi possível estabelecer as fases de introdução e multiplicação, definindo assim o meio de cultura e concentrações de reguladores de crescimento mais adequados ao cultivo in vitro de Xylopia aromatica.
