Depth profiles of hydrogen bound water molecule types and their relation to lipid and protein interaction in the human stratum corneum in vivo

Abstract: Confocal Raman microscopy has been used to measure depth-dependent profiles of human SC in vivo in the high wavenumber (HWN) region. In order to keep the linearity of HWN region boundaries and to not remove an informative signal from Raman spectra, a new baseline subtraction procedure has been introduced. After baseline subtraction, the HWN spectrum was deconvoluted using 10 Gaussian functions with individual chemical meanings. The results show that the hydrogen bound water molecule types contributed different… Show more

“…Figure A shows the ratio of weakly bound/strongly bound water molecule types in the skin, which represent the hydrogen bonding state of water molecules . It can clearly be seen that the application of both OCAs within 30 and 60 minutes did not affect the hydrogen bonding states of water in the depths 0 to 200 μm.…”

“…for a routine investigation of cosmetic problems. This led to an impetuous development of several in vivo methods, which allow for the assessment of skin structure, including high frequency ultrasonography 23 , optical coherence tomography (OCT) 24, 25 , confocal laser scanning microscopy (LSM) 2628 , confocal Raman microscopy (RM) 29, 30 and multiphoton imaging 31 , including multiphoton tomography (MPT) 32 combined with coherent anti-Stokes Raman scattering (CARS) 33, 34 .…”

“…Although all textiles showed highly significant increase in relative moisture determined by a Corneometer, the increase in cotton, having the highest water permeability, is lowest. Alternative approaches to determine the hydration status of the skin could be confocal Raman microscopy, which can also be applied depth dependently and in vivo …”
