De novo assembly and characterization of leaf and floral transcriptomes of the hybridizing bromeliad species (Pitcairnia spp.) adapted to Neotropical Inselbergs

Abstract: We present the leaf and floral transcriptomes of two hybridizing bromeliad species that differ in their major pollinator systems. Here we identified candidate genes responsible for pollinator attraction and reproductive isolation in these two species. We searched for candidate genes involved in floral traits, such as colour. Approximately 34 Gbp of cDNA sequence data were produced from both tissues and species, resulting in a total of 424 506 914 raw reads. The de novo-assembled transcriptomes consisted of a t… Show more

“…ABA can independently improve anthocyanin accumulation and also has a synergic effect with sugar [ 56 , 57 ]. ABA regulates anthocyanin biosynthesis via MYB, bHLH and bZIP [ 58 60 ]. In this work, one DEG with the function of ABA receptor PYL4 in ABA signal pathway was down-regulated.…”

“…Although they retrieved a great number of transcripts, and opened new opportunities of transcriptome inference for non-model organisms, they also found potential issues, namely: sensitivity of alignment error due to paralogs and multigene families; production of artefactual chimeras; problems reconstructing transcript length, and potentially misestimating allelic diversity. These issues were further confirmed in several subsequent articles dealing with the de novo strategy [ 17 19 ], although the extent of sensibility to each varies slightly depending on the software and analytical solutions used.…”
