Multiparametric flow cytometry was used to analyze the development of the endosperm in Zea mays L. during the period from 8 to 20 days after pollination (dap). Nuclear size, DNA content per nucleus, and frequencies of nuclei with varying properties were measured in preparations that included all of the endosperm nuclei of single kernels of the inbred strain A188. Characteristics of nuclear populations from different kernels on the same ear showed minimal variation. The dynamic changes of non-mitotic cells involved in endosperm development consisted of alternating periods of DNA replication with nonreplication. Seven rounds of DNA replication had occurred in some nuclei in the later developmental stages with the rate averaging approximately one round per 24-hour period. Analysis of the DNA levels in the nuclei showed an exact doubling pattern indicating an endoreduplication process, that is, replication of the entire genome during each round. The loosely organized polytenization of the chromatin occurred to varying extents among the nuclei within an endosperm. A weak positive correlation existed between DNA content and size of nuclei suggesting that DNA increases and nuclear growth may not be highly coordinated in this tissue. Increased proportions of the larger nuclei occurred in the later stages of endosperm development. Considering the entire endosperm, the average DNA content per nucleus at the 15-dap peak level was approximately 12.8 C constituting a 2.7-fold overall increase from 8 dap.

<div>Fueron analizadas citológicamente cuarenta y un especies pertenecientes a ocho de las nueve secciones del género Arachis. Se analizaron en detalle los cariotipos veinticuatro de las cuarenta y un especies. Todas las especies tienen un par de cromosomas con satélite, excepto A. valida, que tiene dos pares. Los satélites se clasifican en diez tipos. Las secciones Procumbentes, Erectoides, Extranervosae, Caulorrhizae, Rhizomatosae, Heteranthae y Trierectoides tienen sólo uno o dos tipos de cromosomas con satélite, mientras que la sect. Arachis tiene seis tipos. Un par de cromosomas pequeños se encuentran en la sect. Arachis, excepto en A.batizocoi, A. ipaensis, A. glandulifera, A. magna y A. benensis. A. glandulifera (sect. Arachis) tiene el cariotipo más asimétrico, con seis pares de cromosomas subtelocéntricos y A.guaranitica (sect. Trierectoides) tiene el cariotipo más simétrico. Esto apoya la hipótesis de que la sect. Trierectoides es la más antigua en el género y la sect. Arachis tiene un origen más reciente. Teniendo en cuenta los cariotipos, morfología externa y la distribución geográfica, A. duranensis y A. ipaensis son probablemente los donantes del genoma de A.hypogaea.</div>

Variation in Karyotype and 4C DNA Content in Six Species of Melocactus of the Family Cactaceae.

Analysis of karyotype including somatic chromosome number determination, total chromosome length and volume, estimation of 4C DNA content and Interphase Nuclear Volume (INV) were carried out in 6 species of Melocactus of the family Cactaceae. Somatic chromosome number recorded 2n=22 in Melocactus bahiensis, M curvispinus , M robustispinus, M. townsendii and 2n=44 in M longispinus, M. matanzanus for the first time. Significant interspecific variation in nuclear DNA amount was recorded. The 4C DNA content varied from 20 .55 pg in M. townsendii to 40.80 pg in M. longispinus. The correlation coefficient studies showed that the genomic chromosome length, chromosome volume and INV were possitively correlated. No interdependent was found between 4C DNA content and chromosome length or volume and INV.
