Comparative study of the speed of acid‐suppressing effects of oral administration of cimetidine and famotidine

Abstract: Background and Aim: H2 histamine receptor antagonists (H2RAs) are widely used in patients with acid-related diseases, and the onset of antisecretory activity of H2RAs is reported to be faster than that of proton pump inhibitors (PPIs). The aim of this study was to compare the rapidity of onset of antisecretory activity of cimetidine and famotidine when orally administered. Methods: Fifteen healthy male Japanese volunteers (five H. pylori-positive and 10 H. pylori-negative) participated in a randomized cross-ov… Show more

“…Both drugs have potent acid‐suppressing effects, but they have different clinical characteristics. H 2 RA have a rapid onset of acid‐suppressing effects and are effective for acid control, particularly at night 1,2 . However, it has been reported that the long‐term administration of H 2 RA causes tolerance and attenuation of its acid‐suppressing effect 3,4 .…”

“…Furthermore, placebo effect could be higher because we set the baseline symptoms of heartburn only once or more in the observation period in our study. H 2 RAs have been reported to inhibit gastric acid secretion more promptly than PPIs and are therefore expected to rapidly improve symptoms of heartburn [18][19][20][21].…”

“…An intra-gastric pH monitoring sensor catheter (Zenetics Medical, Salt Lake City, UT) was introduced into the gastric body and monitoring was started at 17:00, as previously reported. (16,17) A single oral dose of the PPI was administered at 15 min before supper, at 18:45. The subjects started eating their supper (carbohydrates 112.8 g, protein 16.3 g, fat 27.3 g, calories 762 kcal) at 19:00 and were asked to finish within 30 min.…”
