Mutagenic and genotoxic effects of dicapthon were investigated by using the bacterial reverse mutation assay in Salmonella typhimurium TA97, TA98, TA100 and TA102 strains with or without metabolic activation system (S9 mix), and chromosome aberrations (CAs), sister chromatid exchanges (SCEs), and micronucleus (MN) tests in human peripheral blood lymphocytes in vitro. Dicapthon was dissolved in dimethyl sulfoxide for all test systems. 0.1, 1, 10 and 100 lg/plate doses of dicapthon were found to be weakly mutagenic on S. typhimurium TA 98 without S9 mix. The human peripheral lymphocytes were treated with four experimental concentrations of dicapthon (25, 50, 100, and 200 lg/mL) for 24 and 48 h. Dicapthon increased the frequency of SCE only at the 100 lg/mL concentration for the 24 and 48 h applications. Dicapthon also induced abnormal cell frequency, CA/cell ratio and frequency of MN dose dependently for 24 and 48 h. Dicapthon showed a statistically significant cytotoxic effect by decreasing the mitotic index in all concentrations and a cytostatic effect by decreasing nuclear division index in 100 and 200 lg/mL concentrations for both treatment periods when compared with both untreated and solvent controls. These values decreased also in a dose dependent manner.

The organophosphorous insecticide acephate was tested for its ability to induce in vitro cytogenetic effect in human peripheral lymphocytes by using the chromosomal aberrations (CAs), sister chromatid exchange (SCE) and micronuclei (MN) assay. The level of nuclear DNA damage of acephate was evaluated by using the comet assay. Concentrations of 12.5, 25, 50, 100 and 200 lg mL -1 of acephate were used. All concentrations of acephate induced significant increase in the frequency of CAs and in the formation of MN dose dependently (r = 0.92 at 24 h, r = 0.95 at 48 h for CAs, r = 0.87 for MN). A significant increase was observed in induction of SCE at 50, 100 and 200 lg mL -1 concentrations during 24 h treatment and at all concentrations (except 12.5 lg mL -1 ) during 48 h treatment period in a dose-dependent manner (r = 0.84 at 24 h, r = 0.88 at 48 h). Acephate did not affect the replicative index and cytokinesis-block proliferation index (CBPI). However, it significantly decreased the mitotic index at all three highest concentrations (50, 100, 200 lg mL -1 ) for 24 h treatment and at all concentrations (except 12.5 lg mL -1 ) for 48 h treatment, dose-dependently (r = 0.94 at 24 h, r = 0.92 at 48 h). A significant increase in mean comet tail length was observed at 100 and 200 lg mL -1 concentrations compared with negative control in a concentration-dependent manner (r = 0.94). The mean comet tail intensity was significantly increased at only 200 lg mL -1 concentration. The present results indicate that acephate is a clastogenic, cytotoxic agent and it causes DNA damage at high concentrations in human lymphocytes in culture.

Özet. Spor tarihi boyunca genç yaştaki bir çok sporcunun doping amacıyla anabolik-androjenik steroidleri yoğun olarak kullandığı bilinmektedir. Bu çalışmayla, kullanımı yasaklanmış olsa da yapılan araştırmalarda illegal olarak halen sıkça kullanılmakta olan Nandrolone Decanoate (Deca-durabolin®) ve Stanozolol (Winstrol) doping maddelerinin olası genotoksik, mutajenik ve sitotoksik etkilerinin insan kan kültüründe in vitro mikronükleus testi ile belirlenmesi amaçlanmıştır. Bu amaçla doping maddelerinin farklı konsantrasyonları (1, 10, 25, 50, 75 ve 100 mM) hazırlanmış ve kan kültürüne uygulanmıştır. Genotoksik etki için mikronükleus frekansları, sitotoksik etki için ise Nükleer Bölünme İndeksi (NBİ) hesaplanmıştır. Elde edilen veriler, çözücü olarak kullanılan dimetil sülfoksit (DMSO) ile hazırlanan negatif kontrol grubuyla ve genotoksik etkisi çok iyi bilinen Etil metansülfonat (EMS) ile hazırlanmış pozitif kontrol grubuyla karşılaştırılmıştır. Bu sonuçlara göre; çalışmamızda kullandığımız doping maddelerinden Nandrolone Decanoate özellikle son iki konsantrasyonda (75 ve 100 mM) insan periferal lenfositlerinde mikronükleus frakansını istatistiksel olarak anlamlı (p< 0,05) bir şekilde artırmıştır. Ayrıca NBİ oranlarına bakıldığında da özellikle son iki konsantrasyonda kontrol grubuna göre önemli düşüşler tespit edilmiştir. Bununla birlikte diğer doping maddesi olan Stanozolol ise sadece son konsantrasyonda (100 mM) anlamlı artışlara neden olmuşken NBİ değerini etkilememiştir. Tüm elde ettiğimiz sonuçlardan her iki doping maddesinin de yüksek konsantrasyonlarda genotoksik ve sitotoksik etkiler yaratabileceği bu nedenle kullanımlarının tehlikeli olabileceği sonucuna ulaşılmıştır.Anahtar Kelimeler: Genotoksisite, Lenfosit kültürü, Mikronükleus, Nandrolone Decanoate, Stanozolol The Investigation of the Cytotoxic and Genotoxic Effcts of the Anabolic Steroids in Human Peripheral Lymphocytes Culture by Micronucleus TestAbstract. In this study, the genotoxic, mutagenic and cytotoxic effects of possible doping substances, Nandrolone Decanoate (Deca-durabolin®) and stanozolol (Winstrol) that are currently being illegally widely used, though the use of them are prohibited, are used to determine in human blood culture by in vitro micronucleus test. For this purpose, different concentrations of doping substances (1, 10, 25, 50, 75 and 100 mM) were prepared and applied to the blood culture. For genotoxic effects micronucleus frequency and for to the cytotoxic effects Nuclear Fission Index (NBI) was calculated.
