Chondroitin Sulfate-Degrading Enzymes as Tools for the Development of New Pharmaceuticals

Abstract: Chondroitin sulfates are linear anionic sulfated polysaccharides found in biological tissues, mainly within the extracellular matrix, which are degraded and altered by specific lyases depending on specific time points. These polysaccharides have recently acquired relevance in the pharmaceutical industry due to their interesting therapeutic applications. As a consequence, chondroitin sulfate (CS) lyases have been widely investigated as tools for the development of new pharmaceuticals based on these polysacchari… Show more

“…Unlike HSPGs, CSPG and DSPGs are a group of mostly secreted proteins forming an integral part of the ECM (Figure 1). They are involved in many essential physiological functions such as morphogenesis, inflammation and neuronal plasticity and play central roles in pathological processes such as cancer, osteoarthritis and thrombosis making them potential therapeutic targets (119,120). Several secreted matrix CS/DS and KS proteoglycans contain leucine-rich repeats (LRRs) in their protein core (around 42 kDa), categorizing them as small leucinerich proteoglycans (SLRPs).…”

“…To control biodegradability CS can be mixed with other polymers to make scaffolds. CS also has pharmacological applications such as coating material for implants and hydrogel in controlled drug release (Benito‐Arenas et al, 2019).…”

“…24 CS chains having a low and ultra-low M W , about 1500 to 2500 Da showed better antioxidant properties, resulting in more bioavailability via administering it orally, this has led to the development of bio-catalytic techniques using enzymes that degrade GAG, to procure chains that are size specific. 25 Sulfation patterns and specific sizes result in precise action, such as highly purified CS, having a size ranging from 15 to 16 kDa obtained from bovine tissues exhibited antiapoptotic and neuroprotective effects. 26 In contrast to HA, CS furthers cell adhesion, and thus can be utilized to develop non-adhesive polymers that show cell adhesion.…”
