At the present time, more complex analyses of apolar compounds with similar chemical structures or of polar compounds, especially basic ones, having diverse properties require more selective stationary phases having better stabilities. This paper describes several new stationary phases with directed selectivities that were prepared by immobilizing two different phenyl group-containing siloxanes and a trifluoropropyl-containing siloxane onto chromatographic silica and, in the case of the fluorinated siloxane, onto zirconized silica, using thermal treatment or microware radiation. The chromatographic properties and stabilities of these new phases were determined and several applications were evaluated. The phenyl-containing phases showed excellent characteristics related to the separation of several different types of aromatic compounds while the fluorinated phases, which present a more polar character, revealed selectivity for the separation of positional isomers as well as for a mixture of basic pharmaceuticals. Stability tests indicate that immobilization of the polysiloxanes increases column lifetimes by making the stationary phases less susceptible to dissolution, while the phases immobilized with microwave radiation were somewhat more stable than those immobilized by thermal treatments.

The present work reviews recent advances in the preparation of new reversed phase packing materials such as sterically protected, bidentate, hybrid organic-inorganic and monolithic phases and phases containing embedded polar groups. The bonding chemistry involved in the preparation of these phases as well as their advantages over conventional C 8 and C 18 reversed phases are discussed. Understanding the reasons behind the development of these newer column packings helps analysts select the best stationary phase for a given application.Keywords: stationary phases; embedded phases; HPLC. INTRODUÇÃOApesar de existirem colunas recheadas com materiais poliméricos para Cromatografia Líquida de Alta Eficiência (CLAE), a sílica é o material mais utilizado como suporte devido à sua alta estabilidade mecânica e térmica, rigidez e a grande eficiência obtida nas separações cromatográficas 1 . Na década de 70, quando a CLAE começou a ser desenvolvida, a sílica utilizada 2 apresentava forma irregular e tamanho de partícula de 40 µm. Por volta de 1980, foi substituída por partículas menores e de morfologia esférica. O próximo avanço 3 foi o uso de partículas com diâmetros cada vez menores, sendo que aquelas de 10 µm foram preferencialmente trocadas pelas de 5 e 3 µm. Atualmente, já existem colunas recheadas com partículas porosas de 1,6 micrômetros 4 . Também ocorreram mudanças significativas quanto à pureza das sílicas, principalmente no método de obtenção destes materiais porosos. A sílica, que antes era obtida diretamente do silicato de sódio, agora passa a ser preparada através do processo sol-gel, empregando o tetraetoxissilano como fonte de SiO 2 . Com a utilização de solventes e agentes surfactantes apropriados, sílicas de diferentes tamanhos e porosidades são obtidas 5 . Hoje é possível adquirir sílicas altamente puras, livres de contaminantes como ferro e alumínio. A presença desses metais M 3+ e outros, como contaminantes na matriz de sílica, aumenta consideravelmente a acidez dos grupos silanóis (≡Si-OH), dificultando a análise de compostos básicos em condições de fase reversa 6 . Essas melhorias no suporte cromatográfico permitiram ganhos significativos na eficiência, estabilidade e reprodutibilidade das colunas 7 . Alguns pesquisadores consideram que a modificação química da superfície da sílica parece não ter evoluído muito quando comparada com os métodos de obtenção da sílica, por ainda utilizar os mesmos organossilanos empregados há mais de trinta anos 8 . Entretanto, os pesquisadores engajados no desenvolvimento de novas colunas para CLAE conseguiram alguns melhoramentos para as fases quimicamente ligadas do tipo C 8 e C 18 , através da reação de capeamento ou bloqueio dos grupos silanóis 7 .
