Artificial Spider Silk Based Programmable Woven Textile for Efficient Wound Management

Abstract: Trauma often causes great damage to the human body's skin and soft tissues, and results in severe fractures. Though the regeneration of wound tissue and the sensing of wound secretions have been studied extensively, wound therapy remains challenging. Herein, an intelligent artificial spider silk protein (spidroin) composite programmable woven textile (i-SPT) with high tensile properties is developed, which possesses biochemical sensing abilities and air permeability as well as motion monitoring capabilities fo… Show more

“…developed an intelligent artificial composite made of spider silk protein (spidroin) and programmable woven textile (i-SPT) with high tensile properties that possesses biochemical sensing abilities and air permeability as well as motion monitoring capabilities for wound management. High sensitivity for motion monitoring is endowed by the highly integrated microelectronic circuit on the i-SPT [ 182 ]. Similarly, Tian et al.…”

“…The synthesis of recombinant spidroin has been reported in detail in the group's previous work. 20 Briefly, chimeric recombinant spidroin was constructed using the N-terminal structural domain (NT) of Eubalaena australis MaSp1 and the C-terminal structural domain (CT) of the highly soluble A. ventricosus MiSp, as well as the short repeat region (Rep) of E. australis. In addition, the fusion tag Nutilizing substance A was utilized to enhance the solubility of the heterologously expressed recombinant spidroin in E. coli.…”

“…Recombinant spidroin is a multifunctional biomaterial secreted by gene-edited Escherichia coli. Due to their excellent mechanical properties and outstanding biocompatibility, spidroin biomaterials have been extensively developed and modified for drug carriers, tissue engineering scaffolds, and other materials requiring strength and elasticity. āˆ’ Compared to conventional microneedle patches, the drug-carrying capacity of the cactus-mimicking microneedle scaffold is enhanced by the inverse opal (IO) photonic crystal (PC) structure of the microneedle portion and the gap filling of the scaffold portion. āˆ’ In addition, in early wound trauma, there is a large amount of exudate around the wound that needs to be removed immediately. The high water content, porous structure, and good biocompatibility of hydrogels allow for high absorption of exudate and provide a moist environment conducive to wound healing. , Meanwhile, microneedles enable painless drug medication without reaching the nerves and blood vessels. āˆ’ By combining materials, preparation techniques, and the structural advantages of the scaffold itself, this microneedle scaffold offers great promise and efficiency in skin wound healing.…”

“…1,2 For example, diabetic wounds due to nerve necrosis lack a motion of perception, and frequently unconscious movement of joints will stretch the granulation tissue and further disturb the normal healing process. 3 Real-time monitoring of joint movement is thus critical as it can transform the external movement into easily identifiable electrical signals, benefiting in a timely way by notifying patients and their caregivers for effective interventions. [4][5][6] Therefore, developing dressings with simultaneous motion-sensing functions and promoted healing effect may be a wise way to treat these kinds of special wounds.…”
