Melia azedarach is commonly used in traditional and folk medicine in Korea and China to treat a variety of diseases including diarrheal, diabetic, rheumatic, and hypertensive disease. The aim of this study was to determine the potential prophylactic and therapeutic effects of Melia azedarach against a broad spectrum of viruses in in vitro cell culture model and the protective effect against different influenza A subtypes in BALB/c mice model. An effective dose of pre-treatment, co-treatment, and post-treatment of Melia azedarach significantly reduced the replication of coxsackievirus, herpes simplex virus, influenza A virus, enterovirus, and bovine rhinovirus in both epithelial and macrophage cell lines. Melia azedarach treatment remarkably promoted the phosphorylation of the key molecules associated with the type-1 interferon and NF-κB signaling pathways. Furthermore, it induced the secretion of type-1 interferon and pro-inflammatory cytokines and the subsequent stimulation of the antiviral state in both epithelial and macrophage cells. Interestingly, oral inoculation of an effective dose of herb extract significantly improved viral clearance in the lungs of BALB/c mice, thus exhibiting protection against several subtypes of influenza A virus. Together with our results indicate that an extracts of Melia azedarach and its components could exhibit a potential natural source of an antiviral drug candidate for a broad spectrum of viruses in animal and humans.

Dodder (Cuscuta spp.) is a parasitic weed damaging many plants and agricultural production. The native obligate parasite Cuscuta japonica Choisy (Japanese dodder) parasitizes Dimocarpus longans Lour., Ficus septica Burm. F., Ficus microcarpa L.f., Mikania micrantha H.B.K. and Melia azedarach Linn, respectively. Five Japanese dodders growing on different plants exhibit slightly different metabolites and amounts which present different pharmacological effects. Among these plants, a significant antiviral activity against influenza A virus (IAV) was found in Japanese dodder parasitizing on D. longans Lour. (CL). To further explore methanol extract components in Japanese dodder (CL), four undescribed aromatic glycosides, cuscutasides A–D (compounds 1–4) were isolated, together with twenty-six known compounds 5–30. The chemical structures of 1–4 were elucidated using a combination of spectroscopic techniques. The eighteen isolated compounds were evaluated for antiviral activity against IAV activity. Among them, 1-monopalmitin (29) displayed potent activity against influenza A virus (A/WSN/1933(H1N1)) with EC50 2.28 ± 0.04 μM and without noteworthy cytotoxicity in MDCK cells. The interrupt step of 29 on the IAV life cycle was determined. These data provide invaluable information for new applications for this otherwise harmful weed.

Abstract In the present study, we investigated natural compounds contained in Moroccan medicinal plants and that might be used as natural inhibitors of the novel coronavirus, SARS-CoV-2, that causes coronavirus disease 2019 (COVID-19). We first performed a literature search for natural inhibitors of SARS or MERS coronaviruses. We then selected natural compounds that have been biologically tested and confirmed to possess anti-coronavirus activity. Subsequently, we used a molecular docking to determine whether the selected molecules could interact with the virus proteins. The compounds selected from virtual screening were then subjected to an in-silico analysis of absorption, distribution, metabolism and excretion (ADME) properties to select only natural compounds that could be orally bioavailable. Thereafter, a second search has been launched to select Moroccan medicinal plants that contain at least 3 molecules from those natural compounds. As results, among 41 natural inhibitors of SARS or MERS coronaviruses, only 13 have been successfully passed the ADME filtering. These molecules, showed abilities to interact with the novel coronavirus as it was predicted. Using these molecules and based on the data extracted from literature, 29 Moroccan medicinal plants have been found to contain at least 3 of these coronavirus inhibitors. Therefore, the medicinal plants selected in this study might contain direct anti-SARS-CoV-2 compounds.
