BackgroundCornus officinalis, is a deciduous tree native to the eastern Asia, distributes mainly in (e.g. Korea, as well as China, and Japan). It is used as folk medicine to backache, polyuria, hypertension and nervous breakdown. Pharmacological studies have demonstrated that C. officinalis possess anti-oxidant, anti-hyperglycemic, and immune regulatory effects. However, reports on the antioxidant activity of C. officinalis have been limited to in vitro radical scavenging studies. Its mechanism of action within the cell at the genetic level especially has not yet been clearly defined. Therefore, we investigated the anti-antioxidant activities of C. officinalis in RAW 264.7 cells.MethodsThe antioxidant activities and protective effects of C. officinalis ethanol extract on cell damage and the antioxidant enzyme system in lipopolysaccharide (LPS)-induced oxidative stress-damaged RAW 264.7 cells were assessed. To measure the effects of C. officinalis on antioxidant activities, we used the following methods: Total phenol and flavonoid contents, DPPH scavenging activity assay, ABTS scavenging activity assay, FRAP value measurement, xanthine oxidase activity assay, ROS generation measurement and real time PCR.ResultsThe total phenol and flavonoid contents of C. officinalis extracts were 27.04 mg GAE/g and 3.70 mg QE/g, respectively. The antioxidant activities of C. officinalis extracts increased in a dose-dependent manner: the IC50 values for DPPH and ABTS radical scavenging activities of C. officinalis extracts were 99.32 μg/mL and 138.51 μg/mL, respectively. C. officinalis extracts inhibited xanthine oxidase activity and reactive oxygen species generation. The expression of antioxidant enzymes, Cu/ZnSOD, MnSOD, catalase, and glutathione peroxidase increased upon treatment with C. officinalis extracts at 100 μg/mL, compared to that in the LPS-treated group.ConclusionsThese results suggest the therapeutic potential of C. officinalis extract as an anti-oxidant agent.Electronic supplementary materialThe online version of this article (doi:10.1186/s12906-016-1172-3) contains supplementary material, which is available to authorized users.

Cornus officinalis is widely distributed in Korea, and its fruit has been used to make as herbal drug for traditional medicine in Korea, Japan, and China because of its tonic, analgesic, and diuretic properties. However, the effects of C. officinalis methanol extract (COME) on melanogenesis remain poorly understood. We evaluated the melanogenic capability of COME in melan-a cells, which are immortalized mouse melanocytes. COME increased melanin synthesis in a dose-dependent manner. Treatment with 12.5 μg/mL of COME significantly increased melanin content by 36.1% (p < 0.001) to a level even higher than that (31.6%) of 3-isobutyl-1-methyl-xanthine, a well-known pigmentation agent. COME also upregulated tyrosinase activity and its messenger RNA and protein expression. In addition, COME upregulated the expression of tyrosinase-related proteins 1 and 2 and microphthalmia-associated transcription factor-M messenger RNA expression. These results imply that COME may be appropriate for development as a natural product to treat hair graying.

Methods: The Rubus coreanus Miquel was extracted with 70% methanol and concentrated with a rotary evaporator. Antifungal effect of Rubus coreanus Miquel extract on C. albicans was determined by paper disc diffusion method and standard plate count method. Seven different concentrations (2,4,8,15, 30, 60, 120 mg/ml) of the extract were tested by paper disc diffusion method. Two kinds of concentration (8, 60 mg/ml) of the extract were tested using standard plate count method on C. albicans with different incubating time (for 6, 12, 24 hours immediately after the cultivation). Morphological changes of C. albicans cells after exposure to the extract were observed with scanning electron microscopy (SEM) images. Results: The Rubus coreanus Miquel extract showed an antifungal effect on C. albicans in 8, 15, 30, 60, 120 mg/ml of concentrations (P<0.05). The extract with 8 mg/ml of concentration showed about 30% of growth inhibition at 6 h and with 60 mg/ml it showed about 90% of growth inhibition at 24 h. SEM analysis showed damaged surfaces of C. albicans cells when treated with Rubus coreanus Miquel extract. Conclusions: The Rubus coreanus Miquel might have the potential as a nobel growth inhibitory agent against C. albicans that causes oral infection.
