It is known that ram's sperms are sensitive to oxidative stress during cooling storage. The purpose of this study was the evaluation of adding an aqueous prickly pear extract (PPE) in ram semen on sperm parameters during cooling storage. Three levels of PPE were added to the diluted ram semen (PPE1=5mg +Tris extender, PPE2=10 mg +Tris extender, and PPE3=30 mg +Tris extender), and compared with control group (Tris extender without addition of PPE). Motility of sperm, hypo osmotic swelling test (HOS-Test), and dead sperm percentage were assessed after 2 and 48 hour at preservation. The PPE2 group exhibited the significantly (p≤0.05) enhanced sperm motility at 2 and 48 hour of cooling. A non-significant (p>0.05) increase in HOS-test percentage for all groups after 2 hours of cooling was observed. The PPE1 and PPE3 groups recorded the lowest percentage of HOS-test after 48 hours, however, groups PPE2 and the control recorded the significant (p≤0.05) excess in this characteristic. The sperm dead percentage were reduced by adding 10 mg of prickly pear extract (PPE2) in comparison with other groups at 2 hours post-cooling. The aqueous prickly pear extract has the potential to be used as an additive in ram semen to improve sperm motility, plasma membrane integrity, and dead sperm percentage at cooling preservation.

N-Nitroso compounds are suspected to be causative agents for human cancer. N-Methyl-N-nitrosourea (MNU) is a typical direct acting mutagen with alkylation activity of DNA, and has been reported to be formed in vivo. Therefore, it is important for cancer chemoprevention toˆnd some compounds to inhibit mutagenicity induced by MNU. The inhibitory eŠect of plant extracts against the mutagenicity of MNU was evaluated using the Ames assay with Salmonella typhimurium TA1535. Among 43 extracts derived from medicinal and edible plant assayed, Glycyrrhiza aspera ethanolic extract, Glycine max extract with 40% iso‰avone aglycone (ISOMAX AG40) and Zingiber o‹cinale ethanolic extract at the range 0.01-1.0 mg/plate inhibited MNU-induced mutagenicity in S. typhimurium TA1535. No cytotoxicity of the three extracts were observed in Salmonella typhimuirum TA1535, indicating that inhibition of MNU-induced mutagenicity was apparently due to the antimutagenic potency of Glycyrrhiza aspera ethanolic extract, ISOMAX AG40 and Zingiber o‹cinale ethanolic extract. Therefore, the results of relative mutagenicity showed that Glycyrrhiza aspera ethanolic extract and ISOMAX AG40 decreased the mutagenic eŠects to 5.4% and 2.6%, respectively, whereas Zingiber o‹cinale ethanolic extract decreased it to 45%.

This study was conducted to explore the adding effect of Opuntia Ficus Indica extract (OFI) to Tris extender preserved of Awassi rams semen characteristics. This study was carried out at the Animal Farm of the Animal Production Department / College of Agricultural Engineering Sciences/ University of Baghdad. Semen was extended using Tris extender with adding different concentrations of OFI extract which were 0% (control group), 0.5% (T1), 1% (T2), and 3% (T3). The effect of additives was studied during different preservation periods 0, 24, and 48 hours at 5°C. Adding 1% of OFI extract significantly increased (P≤0.05) individual sperm motility at different time post-cooling. While adding 0.5, 1, and 3% of OFI extract decreased sperm's abnormality after 24 hours post-cooling. Also, using 0.5 and 1% of OFI extract improved plasma membrane integrity at 24 and 48 hours post-cooling. The sperm viability significantly increase (P≤0.05) in T2 group at zero time and after 24 hours post-cooling. This study proved that adding 3% of OFI extract to diluted Awassi rams semen may be lead to sperm damage.
