Polyherbal formulation (POL-4) is popularly used by traditional practitioners to treat diabetes in district Badin, Sindh, Pakistan. This study validates the folk claim of POL-4 in diabetes and associated dysfunctions (hyperlipidaemia, endothelial dysfunction, hypertension) using OGTT and alloxan-challenged diabetic assays. Administration of POL-4 extract (300 mg/kg bw) markedly (p < 0.01) lowered blood glucose levels compared to only glucose loaded (2 g/kg bw) rats. In alloxan (135 mg/kg bw) fed rats, chronic (21 days) administration of POL-4 powder (3% and 6% w/w in diet) significantly (p < 0.001) prevented the rise in systolic blood pressure, heart rate, serum blood glucose and lipid parameters, while improved serum high density lipoprotein-cholesterol and endothelium-dependent aortic acetylcholine-mediated relaxation. POL-4 administration showed no signs of toxicity or lethality in mice at 1-5 g/kg bw doses. These findings attest antihyperglycaemic, antihyperlipidaemic, endothelium modulatory and antihypertensive effects of POL-4, thus providing evidence to the folk use of POL-4 in diabetes and related complications. Rezumat În districtul Badin, Sindh, Pakistan, pentru tratamentul diabetului, este utilizată tradițional o formulare ce conține părți aeriene provenite de la mai multe plante (POL-4). Acest studiu validează folosirea POL-4 în diabet și disfuncții asociate (hiperlipidemie, disfuncție endotelială, hipertensiune), pe un model de diabet zaharat indus cu aloxan. Administrarea extractului POL-4 (300 mg/kgc) a scăzut semnificativ (p < 0,01) nivelul glicemiei față de lotul control tratat cu glucoză (2 g/kgc). La șobolanii tratați cu aloxan (135 mg/kgc), administrarea cronică (21 zile) a POL-4 (3% și 6% m/m în dietă) a prevenit creșterea tensiunii arteriale sistolice, a ritmului cardiac, modificarea parametrilor serici glicemici și lipidici. POL-4 la dozele de 1-5 g/kgc nu a prezentat semne de toxicitate la șoareci.

Context: Antioxidants play a major role in protecting the body against oxidative stress that is associated with many chronic diseases and disorders including chronic wounds. Plants are the richest source for antioxidant and are effective in the management of oxidative stress, caused by free radical damage. Wound healing and antimicrobial potential are also attributed to the antioxidant potential of drugs. Aim: The aim of this study is to carry out the antioxidant and antimicrobial activity of given polyherbal formulation (PHF) to correlate with the wound healing potential of the formulation. Materials and Methods: Antioxidant potential of the PHF was evaluated by the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method. Agar well diffusion method was used to determine its antimicrobial activity against the Escherichia coli, Pseudomonas aeruginosa, Proteus vulgaris, Klebsiella aerogenes. Results: Results of the study demonstrated that PHF exhibited significant antioxidant activity. Antibacterial activity of polyherbal formula was evaluated against the four pathogenic microorganisms in which it showed mild-to-moderate antimicrobial activity against the E. coli and K. aerogenes, while mild antimicrobial activity against the P. aeruginosa and P. vulgaris. Conclusion: Results of this study suggested that PHF can be used for the treatment of wound infections due to its marked antioxidant and antimicrobial activity.

Herbs are an effective source of Ayurvedic medicine and other traditional medicines as well as modern medicine. Faced with the challenges of modern medicine, the world is shifting toward Ayurvedic theories of health management. In the treatment of the disease, the use of drugs is not limited. This increases the demand for raw materials exponentially. However, production growth is still linear. Rapid depletion of resources creates major problems. To fill the deficit, advances in adulteration and substitution are becoming more prevalent. This adulteration and substitution are a burning problem in an industry that threatens the integrity of Ayurvedic system of medicine. Imprecise knowledge, misidentification, deforestation, and personal gain have led to adulteration. Therefore, an understanding of all ways of adulteration and substitutions is needed to amend this illegal act and maximize consumer safety. This article focuses on adulteration, its types and various methods of adulteration, as well as the concept of substitution, reasoning, and its types. Attempts have also been made to differentiate these two related terms.
