Acteoside, a Component of Stachys Sieboldii MIQ, May Be a Promising Antinephritic Agent (2): Effect of Acteoside on Leukocyte Accumulation in the Glomeruli of Nephritic Rats

Abstract: ABSTRACT-We investigated the effect of acteoside in comparison with that of cyclosporin A on leukocyte accumulation in the glomeruli of rats with crescentic-type anti-glomerular basement membrane (GBM) nephritis. Acteoside given p.o. at a dose of 30 mg/kg once a day for 15 consecutive days after treatment with anti-GBM serum markedly suppressed the urinary protein as well as glomerular histological changes. Acteoside given p.o. for 5 or 15 consecutive days markedly suppressed the accumulation of total leukocyt… Show more

“…In a comparison of compounds 1 and 3 with 2 and 4, the antioxidative effects of these compounds was potentiated by an increase in the number of phenolic hydroxyl groups in the molecule. The free radical scavenging effects of these phenylethanoids proven here may play an important role in the actions of the Veronica species, and partly explain the mechanisms of the activities of phenylethanoids against neoplasm, 19,20) inflammation 21) and nephritis, 22) in which free radicals are seriously involved.…”

“…In addition, Hajhasemi et al (2006) determined analgesic and anti-inflammatory effects of Stachys lavandulifolia extracts on mice. Many studies have confirmed that extracts or components of Stachys species display significant antitoxic (Zinchenko et al, 1981), antinephritic (Hayashi et al, 1994;Savchenko & Khvorostinka, 1978), anti-anoxia (Yamahara et al, 1990), hypotensive (Takeda et al, 1997), and anti-anxious (Rabbani et al, 2005) effects.…”

“…Crescentic-type anti-GBM nephritis was induced by immunizing rats (150-170g), which had received a nephritogenic dose of rabbit anti-rat GBM serum (0.6 ml/animal), with rabbit f-globulin (6.5 mg/animal) according to the previous method, with a slight modifi cation (20). In the experiments, 24-hr urine samples were collected, and the rats were then divided into 5 groups of 8 rats, so that the average protein content in the 24-hr urine in each group was the similar level.…”
