Transdifferentiation of human gingival mesenchymal stem cells into functional keratinocytes by Acalypha indica in three‐dimensional microenvironment

Abstract: Gingival tissue is reportedly a promising, easily accessible, abundant resource of mesenchymal stem cells (MSC) for use in various tissue engineering strategies. Human gingival MSC (HGMSCs) were successfully isolated from gingival tissue and characterized. To analyze in a two-dimensional form, HGMSCs were cultured with basal medium and induced with 25 µg/ml of Acalypha indica. Quantitative real-time polymerase chain reaction (qPCR) and western blot analysis showed the presence of keratinocyte-specific markers,… Show more

“…However, compared to PDLSCs, GMSCs are less susceptible to replicative senescence and proinflammatory cytokine-induced impairment in osteogenic potentials in vitro and ectopic bone formation in vivo (27). In term of their multipotency, GMSCs not only possess the mesodermal trilineage differentiation potentials (osteocytes, adipocytes, and chondrocytes), but also the potential to transdifferentiate into ectodermal and endodermal cell lineages, such as neural cells (15,34), keratinocytes (35), and endothelial cells (15,36), and odontogenic cells (28), under defined induction conditions, respectively. Due to these compelling characteristics, GMSCs represent a promising source of adult MSCs with potent regenerative potentials (Figure 1).…”

“…Moreover, both PDLSCs and GMSCs differentiated into tendon-like cells using an injectable and biodegradable arginine-glycine-aspartic acid tripeptide-coupled alginate hydrogel scaffold[ 59 ]. The GMSCs could also be induced to differentiate into functional keratinocytes when treated with Acalypha indica in a three-dimensional microenvironment[ 60 ].…”
