ABSTRACT. This study uses ISSR molecular markers to characterize the demographic pattern, and spatial genetic structure (SGS) at different life stages of development (cohorts) in a natural population of Copernicia prunifera in the Rio Grande do Norte State, Brazil. All individuals were sampled and georeferenced in a 0.55-ha plot. The demographic analyses showed a clustered pattern in the first-distance classes and a random or segregated pattern at higher distance classes for all cohorts. Among the three studied life stages, juveniles showed the greatest value for Nei's genetic diversity index (H E = 0.369), while the lowest was found among reproductive adults (H E = 0.341). Greater genetic differentiation was found within life stages (98.61%) than among stages (1.39%). The total population (N = 161) showed positive and significant kinship in the first-distance class (12.3 m). The juveniles showed significant kinship up to 10.5 m. Non-reproductive adults had a positive kinship in the first-distance class (11.0 m) and a random distribution of genotypes in the remaining classes, while reproductive adults showed a random spatial distribution of genotypes. Tests for genetic bottleneck showed that the number of loci with excess observed heterozygosity was greater than expected. The SGS results reflected the restricted seed dispersion of the species, and the genetic bottleneck reflected the reduction of genotypes as a result of the anthropization of C. prunifera natural environments.

ABSTRACT. In order to support further genetic, diversity, and phylogeny studies of Capsicum species, the transferability of a Capsicum annuum L. simple sequence repeat (SSR) microsatellite set was analyzed for C. frutescens L. ("malagueta" and "tabasco" peppers) and C. chinense Jacq. (smell peppers, among other types). A total of 185 SSR primers were evaluated in 12 accessions from 115 C. frutescens L. and 480 C. chinense Jacq, representing different types within each species. Transferability to C. frutescens L. and C. chinense Jacq. occurred for 116 primers (62.7%). Nineteen (16.37%) were polymorphic in C. frutescens L. and 36 (31.03%) in C. chinense Jacq., 17 of which were coincident and could be used to analyze samples obtained for the 2 species.Among these primers, CA49 showed a different amplitude range of alleles between the 2 species (130-132 base pairs for C. frutescens L. and S.I.C. Carvalho et al. 7938©FUNPEC-RP www.funpecrp.com.br Genetics and Molecular Research 14 (3): 7937-7946 (2015) 120-128 base pairs for C. chinense Jacq.), and could differentiate the species. A total of 55 alleles were identified among the 19 polymorphic SSR loci among accessions of C. frutescens L., with the number of alleles per locus ranging from 2 to 5, a mean of 2.89, and the polymorphic information content ranging from 0.30 to 0.65. The number of alleles identified in C. chinense Jacq. was 119, ranging from 2 to 5 alleles per locus, an average of 3.30, and polymorphic information content from 0.19 to 0.68. The C. annuum L. SSR primers were most often transferable and polymorphic for C. frutescens L. and C. chinense Jacq., and we present a set of SSR for each species.

RESUMOO objetivo desse estudo foi selecionar primers ISSR (Inter Simple Sequence Repeat) para estudos da estrutura genética, assim como quantificar a variabilidade genética de uma população natural da C. prunifera. (UBC 841). O número ótimo de locos para se estimar confiavelmente a diversidade genética para esse trabalho foi de 76 locos. Foi encontrada alta diversidade genética, com o número de alelos observados (na = 2,00), alelos efetivos (ne = 1,46), índice de diversidade de Nei (He = 0,28) e índice de Shannon (Ho = 0,44). O valor da coancestria se manteve dentro do intervalo de confiança (P > 0,05), indicando ausência de estrutura genética espacial. Os marcadores ISSR se mostraram eficientes na caracterização genotípica dos indivíduos de carnaúba, servindo como subsídio para planos de manejo e conservação da espécie.Palavras-chave: Arecaceae, carnaúba, nordeste do Brasil, variação genética Genetic diversity of Copernicia prunifera using ISSR molecular markers ABSTRACTThe aim of this study was to select primers ISSR (Inter Simple Sequence Repeat) for studies of the genetic structure, and to quantify the genetic variability in a natural population of C. prunifera. Thirty seven individuals were sampled in the municipality Macaíba, Rio Grande do Norte, Northeast Brazil. Seventeen ISSR primers were tested, of which 12 amplified DNA and of these, seven were selected to characterize the structure and genetic diversity of the population. The primers that had the highest percentage of polymorphic loci were UBC 841 (16.36%), UBC 842 (15.45%), UBC 857 (12.73%), UBC 859 (10.90%), UBC 840 (10.90%), UBC 813 (10%) and UBC 827 (8.18%), totaling 93 of the 110 loci generated. The polymorphic information content (PIC) of each selected primer ranged from 0.057 (UBC 859) and 0.444 (UBC 841). The optimal number of loci to reliably estimate the genetic diversity for this work was 76 loci, ie, when the correlation reached 0.998 and the stress was less than 0.05. High genetic diversity was found, with the number of observed alleles (na = 2.00), effective alleles (ne = 1.46), Nei's diversity index (He = 0.28) and Shannon index (Ho = 0.44). The value of the coancestry remained within the confidence interval (P > 0.05), indicating no spatial genetic structure, regardless of the distance between plants. ISSR markers were efficient in genetic characterization of individuals' carnauba serving with allowance for management and conservation of the species.
