Foi estabelecido um método cromatográfico rápido e simples para a determinação de riboflavina em leite bovino, na presença de tetraciclinas. Para tanto, as proteínas foram precipitadas com ácido tricloroacético e a separação realizada por CLAE em fase reversa com detector de fluorescência. Foi utilizada uma coluna C 8 com eluição isocrática e fase móvel acetato de sódio 0,1 mol L-1 , cloreto de cálcio 35 mmol L-1 , EDTA 25 mmol L-1 : metanol (65:35 v/v). O método foi validado mediante avaliação dos seguintes parâmetros: linearidade (0,9980), faixa linear (0,50 a 1,30 μg mL-1), precisão intra e inter-ensaio (RSD ≤ 0,5%), exatidão (recuperação 92%) e limites de detecção e de quantificação (0,75 μg mL-1 e 0,22 μg mL-1 , respectivamente). O método foi aplicado na análise de leite integral e desnatado, mostrando-se robusto na presença de substâncias fluorescentes como as tetraciclinas, e quando comparado com o método espectrofluorimétrico, não revelou diferença significativa (p≤ 0,05) entre os resultados obtidos. An easy and simple chromatographic method for the determination of riboflavin in bovine milk in the presence of tetracyclines was established. For this, the proteins were precipitated with trichloroacetic acid and the separation was carried out by reversed-phase HPLC with fluorescence detection. A C 8 column was used with isocratic flow using a mobile phase consisting of 0.1 mol L-1 sodium acetate, 35 mmol L-1 calcium chloride and 25 mmol L-1 EDTA : methanol (65:35 v/v). The method was validated by means of the following parameters: linearity (0.9980), linear range (0.50-1.30 µg mL-1), intra-and inter-assay precision (RSD ≤ 0.5%), accuracy (recovery 92%), and detection and quantitation limits (0.75 μg mL-1 and 0.22 μg mL-1 , respectively). The method was applied for analysis of full cream and skimmed milk, showing robustness in the presence of tetracyclines, and when compared with a spectrofluorimetric method showed no significant difference (p≤ 0.05) between the results obtained.

Although royal jelly constitutes one of the richest natural sources of pantothenic acid (vitamin B 5 ), a reliable and validated chromatographic method to determine this analyte in this matrix has yet to be described in the literature. In this work we present an original RP-HPLC procedure to measure the concentration of pantothenic acid in royal jelly. A sample pre-treatment is needed to prevent the interference of high protein concentration in the matrix. The method has been validated in terms of LoD, LoQ, linearity, precision (repeatability and reproducibility) and bias. Finally, the whole procedure was tested on a number of samples of royal jelly from different origins, providing concentration values ranging from 120 AE 30 to 565 AE 40 mg kg À1 of pantothenic acid.

Combination of sugar analysis and stable isotope ratio mass spectrometry to detect the use of artificial sugars in royal jelly production

The effects of feeding bees artificial sugars and/or proteins on the sugar compositions and (13)C isotopic measurements of royal jellies (RJs) were evaluated. The sugars fed to the bees were two C4 sugars (cane sugar and maize hydrolysate), two C3 sugars (sugar beet, cereal starch hydrolysate), and honey. The proteins fed to them were pollen, soybean, and yeast powder proteins. To evaluate the influence of the sugar and/or protein feeding over time, samples were collected during six consecutive harvests. (13)C isotopic ratio measurements of natural RJs gave values of around -25 ‰, which were also seen for RJs obtained when the bees were fed honey or C3 sugars. However, the RJs obtained when the bees were fed cane sugar or corn hydrolysate (regardless of whether they were also fed proteins) gave values of up to -17 ‰. Sugar content analysis revealed that the composition of maltose, maltotriose, sucrose, and erlose varied significantly over time in accordance with the composition of the syrup fed to the bees. When corn and cereal starch hydrolysates were fed to the bees, the maltose and maltotriose contents of the RJs increased up to 5.0 and 1.3 %, respectively, compared to the levels seen in authentic samples (i.e., samples obtained when the bees were fed natural food: honey and pollen) that were inferior to 0.2% and not detected, respectively. The sucrose and erlose contents of natural RJs were around 0.2 %, whereas those in RJs obtained when the bees were fed cane or beet sugar were as much as 4.0 and 1.3 %, respectively. The combination of sugar analysis and (13)C isotopic ratio measurements represents a very efficient analytical methodology for detecting (from early harvests onward) the use of C4 and C3 artificial sugars in the production of RJ.
