Avaliação de preparações antigênicas de Strongyloides stercoralis para o imunodiagnóstico da estrongiloidíase

Abstract: Aqueous-soluble (AS) antigens from larvae of Strongyloides stercoralis, extracted with phosphate-buffered saline, are traditionally used for serodiagnosis of strongyloidiasis. To identify sources of antigens for use in serodiagnosis, residual particulates from parasite larvae after aqueous extraction were solubilized with Tris-buffered 8M urea, yielding a urea-soluble (US) antigen fraction. Both AS and US antigens from S. stercoralis were evaluated by an enzyme-linked immunosorbent assay. No significative diff… Show more

“…Uma importante vantagem dos testes imunodiagnósticos, em relação aos parasitológicos, é que eles não dependem do nível de excreção larval 15 . Uma das principais limitações em se desenvolver testes sorológicos mais sensíveis e específicos é a dificuldade em obter quantidades suficientes de antígenos, que permitam seu posterior fracionamento e análise 16 …”

“…Strongyloides stercoralis infection is usually asymptomatic or present mild clinical manifestations in most individuals (Grove 1996;Rossi et al 1993). However, it can cause hyperinfection and dissemination in immunosuppressed patients especially under corticoid therapy (Conway et al 1995;Genta 1992;Grove 1996;Kothary et al 1999).…”

“…ELISA assay for antibody detection has been shown the most reliable: the sensibility of the assay ranges from 85% to 95% and the specificity can reach 90%, being superior to the majority of other immunological assays (Liu and Weller 1993;Rossi et al 1993;Schaffel et al 2001;Uparanukraw et al 1999). Nevertheless, cross-reactivity with antigens of other nematodes, especially filarial parasites, can occur.…”

“…As strongyloidiasis immunodiagnosis continues to be a challenge because of the increasing difficulty in obtaining sufficient quantities of S. stercoralis larvae [4,5], alternative antigens, including heterologous antigens from Strongyloides venezuelensis have also been used with satisfactory results to diagnose human strongyloidiasis [6,7].…”
