Enzyme linked immunosorbent assay: determination of anti-adenovirus antibodies in an infant population

Abstract: SUMMARYIn order to define an accurate assay for anti adenovirus antibody detection, a recently developed ELISA was compared with IFA and CF. On 58 sera, the ELISA was more sensitive than both CF and IFA, which showed relative sensitivities of 63% and 94% . respectively. It was not possible to determine the exact specificity of the tests because of the lack of a gold standard. Furthermore, the ELISA was used to define the prevalence of adenovirus antibodies in 116 infants between 1 and 24 months old (mean 7.28)… Show more

“…Weinberg et al 21 examined the prevalence of anti-adenovirus antibodies in 116 infants between 1 and 24 months old and showed that more than 80% of the children had been infected by adenoviruses by the age of 10 months. Rosenecker et al 22 in a noteworthy report demonstrated that IgG antibodies against adenovirus were present in 192 of 199 cystic fibrosis patients (96.5%), and were significantly higher in cystic fibrosis patients older than 7 years than in younger patients and in age matched controls.…”

“…Doubtful or discrepant samples were restained and read at least twice to check for reproducibility. An indirect immunofluorescence test was done for adenovirus and respiratory syncytial virus for the detection of antibody response as described elsewere 31 .…”

“…There have been promising attempts to modify the E4 region by either deleting the entire E4 or introducing alterations within individual E4 coding regions so as to minimize immunogenicity, eliminate the toxicity of adenoviral vectors and improve transgene persistence in vivo. 3,17,18,[27][28][29][30] In this study, we developed a recombinant adenovirus with deleted E1 and E3 regions and an inactivated E4 to minimize expression of viral proteins. We used this vector to express a human tumor suppressor gene p53.…”
