METHODS: The research method used in this research was experimental laboratory with pre-and posttest only control group design. RESULTS: The result shows that the extradiol level which has range of 26.30–31.03 from 28 experimental animals measured, this showed more measurement diameter which has not had measurement addition compare with the wide percentage of measurement. The result shows strong correlation between digitalized measured wide follicles to the changing of estradiol level with value of 0.453. The result of comparation between estradiol level and measured diameter shows weak correlation. This shows that manual measurement of follicle diameter still weak to the changing of estradiol level. CONCLUSION: There is strong correlation between measured wide area follicle used ImageJ applications to the changing of estradiol level compare to the measurement of follicle diameter.

BACKGROUND: Menstrual disorders to women are generally caused by the disturbance of estrogen production level. This can cause problems in the female reproductive organs. Herbal medications at this moment are demanding to prevent the risk of chemical substances, one of them is grass jelly leaves (Cyclea barbata) which has the potency to induce estrogen levels to women and some people use it to overcome fertility problems. One of the active compounds content is coclaurine. AIM: The study aimed to know the main compound leaves of C. barbata which has an important role in estrogen induction, protein target, and pathway through to activate estrogen production. METHODS: There are 18 active compounds from C. barbata gained from database and had been confirmed the presence of coclaurine compound used liquid chromatography-mass spectrometry. Then, it predicted that the potency of molecular interaction between active compounds of ESRα had been done by the molecular docking and in vitro approach. RESULTS: Coclaurine has the highest binding affinity to activate ESRα. Even though the network analysis prediction did not show the direct interaction, we predict the coclaurine might first interact with other steroid proteins showing by Cytoscape analysis. In vitro results showing the maximum dosage of C. barbata extract is in 100 ppm, while coclaurine dosage is optimum at 100 and 200 μM/L to induce the maturation of oocytes. CONCLUSION: Coclaurine compounds give stimulation of hormonal induction through several proteins such as SRC, ADRB2, and ADRB3 which involve CYP1A1 and CYP19A1, then activate estrone and estradiol to help the increase of estrogen. C. barbata ethanol extracts and coclaurine had been proved having ability to enhance development follicles pre-ovulation.

The aim of this study is to determine the potency of EO canary therapy towards level of malondialdehyde (MDA) and lung histopathology of asthmatic rats. The asthmatic rats were prepared by sensitization of allergen conducted by intraperitoneal injection and nebulized of ovalbumin (OVA) also intrasulcular injection of lipopolysaccharide from Phorphyromonas gingivalis bacteria. Five groups of rats (Rattus norvegicus) were used in this research; i.e. the control group, the asthmatic group, and three other groups with therapy of EO canary with the dose of 25, 50, and 100 mg/kg body weight for 7 days. The EO canary contents were analyzed by gas chromatography-mass spectrometry (GC-MS). The MDA levels were measured using the thiobarbituric acid (TBA) reagent and histopathology of bronchial stained using Hematoxilin-Eosin (HE) and observed microscopically. The results showed that EO canary significantly (p < 0.01) decrease MDA levels and repair lung histopathology of asthmatic rats. The analysis of EO canary performed by GC-MS composed of thujene, α-pinene, camphene, sabinene, 2-β-pinene, βmyrcene, l-phellandrene, p-cymene, γ-terpinene, cis-Sabinene hydrate, α-terpinolene, camphor, limonene dioxide, sabinyl acetate, piperitone oxide, ascaridole, α-cubene, αcopaene, trans-caryophyllene, and 1,8-cineol. It can be concluded that EO canary has potency as anti-inflammatory agent of asthmatic condition. The most effective dose therapy was obtained as high as 100 mg/kg body weight that decreased 46.56 % level of MDA.
