The angiogenesis is an important process in the pathogenesis of malignancies. It is regulated by various growth factors, with the vascular endothelial growth factor (VEGF) playing the central role. The aim of the present study was to evaluate possible associations of functional VEGF −2578C>A, −634G>C, and +936C>T polymorphisms with the risk for occurrence and progression of non-small cell lung cancer (NSCLC) in patients living in Republic of Belarus. Materials and Methods: A total of 202 patients (147 males and 55 females) diagnosed as having the NSCLC. The control group consisted of 336 individuals (245 males and 91 females) without an oncopathology. The total DNA was isolated from peripheral blood. We investigated the single nucleotide polymorphisms of VEGF (rs 2010963), (rs 699947), (rs 3025039). The genotyping was performed by PCR-RFLP analysis. Results: Our results revealed a marginally significant association of the –2578CC genotype (p=0.002) with a greater degree of tumor spread (Т2–Т4). Heterozygous genotypes –2578СА and +936СT carriers were included into the follow-up group significantly more often (р=0.021 and р=0.012, respectively). Our study demonstrate that VEGF –2578A/C and +936C/T polymorphisms are among the factors determining the individual peculiarities of NSCLC course in this population and can be used for clarifying the prognosis of the disease.
РОЛЬ ГЕНЕТИЧЕСКИХ НАРУШЕНИЙ В ФОРМИРОВАНИИ ИНВАЛИДИЗИРУЮЩИХ ПОСЛЕДСТВИЙ У НЕДОНОШЕННОГО НОВОРОЖДЕННОГО Аннотация. Представлен клинический случай проявления первичной врожденной глаукомы, синдрома дыхательных расстройств, бронхолегочной дисплазии, ретинопатии и перивентрикулярной лейкомаляции у недоношенного новорожденного. Для поиска патогенных вариантов использован метод высокопроизводительного полноэкзомного секвенирования. Учитывая данные о многофакторной этиологии исследуемых осложнений, проанализированы гены белков сурфактанта и гены, ассоциированные с ангиогенезом, ренин-ангиотензиновой и антиоксидантной системами. Выявлены молекулярно-генетические факторы риска развития врожденной глаукомы у недоношенного новорожденного: p.E229K CYP1B1 и p.R76K MYOC, а также полиморфные варианты в генах белков сурфактанта SFTPB (rs1130866, VNTR интрона 4) и SFTPC (rs4715, rs1124), фактора роста эндотелия сосудов VEGFA (rs2010963), синтазы оксида азота 3 NOS3 (rs1799983), белка связывания инсулиноподобного фактора роста 3 IGFBP3 (rs2854746), каталазы CAT (rs7943316), ассоциированных с развитием исследуемых инвалидизирующих осложнений. ГЕНЕТИЧЕСКИЙ ПОЛИМОРФИЗМ ВНУТРИКЛЕТОЧНЫХ СИГНАЛЬНЫХ ПУТЕЙ У ПАЦИЕНТОВ С НЕМЕЛКОКЛЕТОЧНЫМ РАКОМ ЛЕГКОГО Аннотация. Ключевым процессом в патогенезе любых злокачественных новообразований, в том числе и немелкоклеточного рака легкого (НМРЛ), является ангиогенез, который активируется двумя тирозинкиназными каскадами (RAS/ RAF/MAPK и PI3K/AKT/mTOR). Основными генами, контролирующими эти пути, являются EGFR, KRAS, PIK3CA и PTEN. В исследовании проанализированы мутации в 18-21 экзонах гена EGFR, 2 экзоне гена KRAS, 9 и 20 экзонах гена PIK3CA и 7 экзоне гена PTEN у пациентов с НМРЛ, проживающих на территории Беларуси, и изучена их связь с клиникоморфологическими характеристиками опухоли. Полученные результаты показали, что мутации в гене EGFR достоверно чаще в 5 раз встречаются у пациентов с НМРЛ, чем в контрольной группе. Классические мутации в гене EGFR обнаружены только у пациентов с аденокарциномой легкого, преимущественно у женщин. Мутации гена KRAS встречаются только у мужчин, причем у пациентов с аденокарциномой в 3 раза чаще, чем у пациентов с плоскоклеточным раком легкого. В данном исследовании не выявлено соматических мутаций в генах PIK3CA и PTEN у пациентов с НМРЛ. Objective. Analysis of a clinical picture and functional dysfunctions with ADRB2 gene molymorphism in patients with PHCM. Materials and methods. 89 patients with PHCM were included in the study (69 males and 20 females, mean age 46,7±15,7), among which 56 had been diagnosed with the obstructive form, 33 patients had a non-obstructive form of the disease. The control set included 104 healthy donors adequate in age, and gender with PHCM. The clinical picture of the disease was assessed, echocardiographic indicators and daily ECG monitoring parameters were specified. The ADRB2 gene polymorphism analysis was made in all patients. Results. The association of the clinical picture, structural and functional LV myocardial peculiarities with ADRB2 gene polymorphism in patients with PHCM. It was revealed that the combination of Arg16rg/Gln27Gln genotypes is a risk genotype and is associated with the appearance of adverse clinical manifestations of the disease. The combination of Gly16Gly/Glu27Glu genotypes is protective and is Conclusion. The results obtained have revealed the association of the ADRB2 gene polymorphism with clinical, structural and functional dysfunctions in patients with PHCM which will allow making prognoses of adverse clinical manifestations risk and improving β-adrenoblocker treatment of PHCM patients.
