Fetal cerebellar tissue contains the largest number of neurogenic progenitors committed on the differentiation into glutamatergic neurons that can be used in the development of promising new treatment for spinal cord injuries.To evaluate the effect of fetal cerebellar tissue transplantation (FСTT) on the restoration of motor function after spinal cord injury in experiment.Materials and methods. Animals: inbred albino Wistar rats (5.5 months males, weighting 300 grams); main experimental groups: 1 – spinal cord injury + transplantation of a fragment of fetal (E18) rat cerebellum (n = 15), 2 – spinal cord injury only (n = 40). Model of an injury – left-side spinal cord hemisection at Т11; monitoring of the ipsilateral hind limb function (IHLF) – the Вasso-Вeattie-Вresnahan (BBB) scale.Results. FСTT normalizes the distribution of IHLF values, distorts the dynamics of the motor function recovery, transforming it from a progressive (in a control group) to the constant with variation within 3-3.6 points BBB during the experiment. FСTT causes early temporary positive effect on the functional state of the motor system, probably provided by mediator-dependent, neuroprotective, proangiogenic effect and remyelination. In our view, the gradual depletion of the FСTT positive effect due to resorption of the graft within the first 2 months is compensated by autoregenerative neoplastic process that is typical for the control group and by autoimmune utilization of myelin-associated inhibitors of axonal growth in the zone of injury that causes stability of the IHLF value during the observation period.Conclusion. Transplantation of fetal cerebellar tissue causes a short-term positive effect on the motor function recovery limited by the 1st month of the traumatic process. Evaluation of such type of neurotransplantation effectiveness requires taking into account the dynamics of the spasticity and chronic pain.

В условиях экспериментальной дислипопротеидемии, которую моделировали путем скармливания кроликам холестерина в дозе 0,5 г / кг массы тела в течение 3 месяцев, с помощью морфометрических методов определяли особенности структурной перестройки коронарных артерий. Установили, что при экспериментальной дислипо-протеидемии отмечаются признаки выраженного ремоделирования артерий мелкого и среднего калибра, которые проявляются в виде утолщения их стенки и сужения просвета. Более выраженные морфологические изменения обнаружены в сосудах мелкого калибра. Так, площадь просвета артерий мелкого калибра уменьшилась на 48,67%, толщина стенки увеличилась на 42,38% по сравнению с группой интактных животных. Также статистически достоверно изменялись сосудистые индексы: индекс Вогенворта вырос в 4,24 раза, сосудистый индекс -в 2,11 раза, а индекс Керногана снизился в 4,02 раза. Указанные морфометрические изменения приводят к снижению функциональной способности сосудов, нарушению их пропускной способности и, как следствие, ухудшение кровоснабжения тканей миокарда с последующим развитием явлений гипоксии и ишемии.Ключевые слова: экспериментальная дислипопротеидемия, сердце, сосуды, морфометрия.The special features of structural transformation of coronary arteries were studied by morphometric methods under conditions of experimental dislipoproteinemia, provided by feeding model rabbits with cholesterol in a dose of 0.5 g/kg of body weight during 3 months. It was established, that the experimental dislipoproteinemia presented the signs of marked remodeling of small and medium arteries, manifested in the form of thickening the walls and narrowing the lumen. Small vessels presented the most pronounced morphological changes. Thus, the lumen of the small arteries decreased by 48.67%, while the wall thickness increased by 42.38% compared with the one in the group of intact animals. Vascular indices also demonstrated statistically significant changes: the Vogenvort index increased by a factor of 4.24, the vascular index -by a factor of 2.11, and the Kernogan index decreased 4.02 times. The above-mentioned morphometric changes lead to a decrease in the functional performance of vessels, a violation of their throughput and, as a consequence, a deterioration of blood supply to the myocardium tissues with subsequent development of hypoxia and ischemia.Key words: experimental dislipoproteinemia, hearth, vessels, morphometry. The urgent problem of long-term storage of multipotent mesenchymal stromal cells (MMSCs) is to improve the protocol of their cryopreservation for further application maintaining the therapeutic properties and minimizing the risks of adverse effects on the health of the recipient. As a standard cryoprotectant, a mixture of 90 % fetal bovine serum (FBS) and 10 % dimethyl sulfoxide (DMSO) is used, which, however, can cause a variety of adverse reactions. Therefore, it is important to study the possibility of reducing the concentration of potentially dangerous DMSO by adding other components to the mixture for cell cryopreservation. Purpose. To determine the efficiency of cryopreservation of human Wharton's jelly MMSCs using cryoprotectants of different composition by studying the proliferative activity, phenotype and features of cell morphology in culture in vitro. Materials and methods. The cryoprotective effect of various combinations of DMSO, ethylene glycol, sucrose and trehalose was studied. The efficacy was assessed by cell viability, their adhesive properties, expansion rate and monolayer formation, as well as the expression of main MMSCs markers. Results. It is shown that the most effective combination is 4 % DMSO with 6 % trehalose which provides the highest level of preservation of cell viability, as well as their adhesive and proliferative properties during thawing. Other combinations of the cryoprotectant components showed a much slower cell division, in some cases, the monolayer was not formed at all. For all investigated variants, the main surface markers of MMSCs were preserved. Conclusions. The obtained results indicate the possibility of reducing the concentration of DMSO to 4 % in the freezing medium for MMSCs cryopreservation while maintaining their viability, proliferative activity and common surface markers.

On the model of experimental analogue of human multiple sclerosis we studied the effects of the mesenchymal stem cells transfected with plasmid vector containing gene IL-10 (MSCs-T) on the functional state of the CNS in rats.Materials and methods. The experimental allergic encephalomyelitis (EАЕ) was induced with spinal cord homogenate of rats with complete Freund’s adjuvant. The mesenchymal stem cells (MSCs) were isolated by the explants technique from Wharton’s jelly of the human umbilical cord and culture up to two passages. Then the MSCs of second passage were transfected of plasmid vector with gene IL-10 and marker gene of green fluorescent protein. Cell transplantation was performed suboccipitally on the 17th day at a dose of 1 million cells in 100 µl of saline per animal.Results. In the open field test we have established that the use of MSCs-T transfected with gene IL-10 suppressed the vertical locomotor activity and elevated the emotional activity as well as partially corrects horizontal locomotor activity indexes which approach the indexes of intact animals.Conclusions. The use of MSCs transfected with plasmid vector with gene IL-10 in the rats with induced EAE is more effective method than treatment using non-transfected MSCs. Combined treatment with IL-10 + MSCs in ЕАЕ rats is more effective than treatment with transfected МSCs.
