Aim. To study the skin microbiota of pregnant women suffering from atopic dermatitis.Methods. 53 women of reproductive age suffering from atopic dermatitis (28 pregnant and 25 non-pregnant) were examined. The control group included dermatologically healthy women (25 pregnant and 25 non-pregnant). Prior to treatment initiation and on 15-day of study pathological process spread, the SCORAD index (scoring of atopic dermatitis - atopic dermatitis severity assessment), dermatology life quality index determination were conducted. In addition, microbiological study of material taken from the forehead, elbow bend skin and visually unaltered forearm skin was performed.Results. In women (pregnant and non-pregnant), suffering from atopic dermatitis skin total bacterial load is increased. In all groups, the skin microbiota is presented mainly by staphylococci: in dermatologically healthy people - coagulase-negative, in atopic dermatitis - Staphylococcusя aureus. In atopic dermatitis Staphylococcus aureus is isolated from both lesions and visually unaltered skin. In pregnant women with atopic dermatitis skin bacterial load was higher, Staphylococcus aureus was found more commonly. The skin microbiota in dermatologically healthy women was more diverse in respect of species comparing with patients with atopic dermatitis. Basic care remedies use leads to clinical improvement and a decrease in the skin total bacterial load and Staphylococcus aureus load. Daily use of emollients has no effect on saprophytic microorganisms.Conclusion. In pregnant patients with atopic dermatitis higher skin total bacterial load and higher rate of skin colonization by Staphylococcus aureus are observed.
Резюме. Работа посвящена исследованию взаимодействия между иммуноглобулином класса А и рекомбинантными полипептидами Р6, Р7, Р8, созданными на основе поверхностного белка Bac стрептококков группы В, обладающего IgA-рецепторной активностью. В настоящее время существует потребность в иммунохимических реагентах, строго специфичных в отношении IgA как для создания диагностикумов по детекции уровня IgA в биологических жидкостях, так и для аффинного выделения IgA и его фрагментов. С целью изучения возможности реализации указанных выше направлений применения протеинов в ходе исследования была определена способность рекомбинантных полипептидов Р6, Р7, Р8, связывающих Fc-область IgA, вступать во взаимодействие с различными формами IgA (сывороточный IgA, секреторный IgA, подклассы сывороточного IgA-IgA1, IgA2) и подтверждена избирательность связывания лиганда. Выяснено, что строение рекомбинантного деривата Р6, среди трех представленных полипептидов, является оптимальным для осуществления IgA-рецепторной способности. Определены структурные особенности связывания иммуноглобулина А фрагментами Вас белка: положение сайта связывания на молекуле IgA (приближен к эпитопам трех МкАт), изменчивость структуры данного сайта, а также устойчивость сайта связывания полипептидов Р6, Р7, Р8 к частичному восстановлению дисульфидных связей на молекуле IgA.
Цель работы - исследование изменений качественного и количественного состава кишечной микробиоты на модели синдрома системного воспалительного ответа (СCВО), вызванного химически индуцированным колитом и осложненного антибиотик -индуцированным дисбиозом, у крыс стока Wistar под влиянием пробиотической терапии. Моделирование ССВО с использованием антимикробных препаратов широкого спектра действия характеризуется снижением титра потенциально полезных микроорганизмов при увеличении этого показателя для условно патогенных представителей, таких как Proteus spp., K. oxytoca. Отмечено снижение общего бактериального числа, представительства лактобацилл, бифидобактерий, Akkermansia muciniphila и F. Prausnitzii и увеличение относительного количества грамотрицательных бактерий. Для здоровых крыс в возрасте 12-14 недель характерно отсутствие в фекалиях таких условно патогенных представителей нормобиоты, как Proteus spp., Klebsiella oxytoca и Enterobacter spp. с их манифестацией после моделирования ССВО. При введении пробиотических штаммов крысам в возрасте 8-9 недель отмечается увеличение общего бактериального числа, F. prausnitzii с повышением колонизационной резистентности за счет роста популяции Bacteroides spp. Исследование причин данных различий важно для понимания перспектив применения пробиотических препаратов в различных периодах онтогенеза. The aim of this work was to study qualitative and quantitative changes in gut microbiota in an experimental model of systemic inflammatory response syndrome (SIRS) caused by chemically induced colitis and complicated by antibiotic -induced dysbiosis in Wistar rats under the influence of probiotic therapy. Modeling of SIRS using broad -spectrum antimicrobials was characterized by a decrease in titer of potentially beneficial microorganisms with an increase in titer of opportunistic representatives, such as Proteus spp., K. oxytoca. There was a decrease in the total bacterial number, suppressed representation of lactobacilli, bifidobacteria, Akkermansia muciniphila and F. prausnitzii and an increase in the relative number of gram -negative bacteria. Healthy rats aged 12-14 weeks were marked by the absence of such conditionally pathogenic representatives of normobiota as Proteus spp., Klebsiella oxytoca and Enterobacter spp. in the faeces with their manifestation after the SIRS simulation. When probiotic strains were administered to rats aged 8-9 weeks, there was an increase in the total bacterial number and F. prausnitzii with increased colonization resistance due to the growth of the Bacteroides spp. population. The study of the causes of these differences is important for understanding the prospects for the use of probiotic drugs in different periods of ontogenesis.
