MicroRNA (miR)-204-5p was previously identified to be downregulated in melanoma compared with melanocytic nevi. This observation prompted a functional study on miR-204-5p and the newly-identified miR-3065-5p, two miRNAs suggested to be tumor-suppressive oncomiRs. Application of miR-204-5p mimics or inhibitors resulted in a decrease or increase, respectively, in melanoma cell proliferation and colony formation. miR-204-5p mimics hindered invasion, whereas miR-204-5p inhibitors stimulated cancer cell migration. Modulation of miR-3065-5p led to a decrease in melanoma cell proliferation, altered cell cycle distribution and increased expression levels of its target genes HIPK1 and ITGA1, possibly due to functional modifications identified in these cells. miR-204-5p and miR-3065-5p demonstrated antitumor capacities that may need to be taken into account in the development of melanoma treatment approaches.
Background Ileus is common after elective colorectal surgery, and is associated with increased adverse events and prolonged hospital stay. The aim was to assess the role of non‐steroidal anti‐inflammatory drugs (NSAIDs) for reducing ileus after surgery. Methods A prospective multicentre cohort study was delivered by an international, student‐ and trainee‐led collaborative group. Adult patients undergoing elective colorectal resection between January and April 2018 were included. The primary outcome was time to gastrointestinal recovery, measured using a composite measure of bowel function and tolerance to oral intake. The impact of NSAIDs was explored using Cox regression analyses, including the results of a centre‐specific survey of compliance to enhanced recovery principles. Secondary safety outcomes included anastomotic leak rate and acute kidney injury. Results A total of 4164 patients were included, with a median age of 68 (i.q.r. 57–75) years (54·9 per cent men). Some 1153 (27·7 per cent) received NSAIDs on postoperative days 1–3, of whom 1061 (92·0 per cent) received non‐selective cyclo‐oxygenase inhibitors. After adjustment for baseline differences, the mean time to gastrointestinal recovery did not differ significantly between patients who received NSAIDs and those who did not (4·6 versus 4·8 days; hazard ratio 1·04, 95 per cent c.i. 0·96 to 1·12; P = 0·360). There were no significant differences in anastomotic leak rate (5·4 versus 4·6 per cent; P = 0·349) or acute kidney injury (14·3 versus 13·8 per cent; P = 0·666) between the groups. Significantly fewer patients receiving NSAIDs required strong opioid analgesia (35·3 versus 56·7 per cent; P < 0·001). Conclusion NSAIDs did not reduce the time for gastrointestinal recovery after colorectal surgery, but they were safe and associated with reduced postoperative opioid requirement.
Abstract. In the article the derivation of the resolving equations for the calculation of a three-layer plate taking into account the creep of the middle layer by the finite element method is given. Rectangular finite elements are used. The problem reduces to a system of linear algebraic equations. An example of calculating a three-layer plate hinged on the contour and loaded with a uniformly distributed load is considered. A comparison of the results with the solution based on the finite difference method is presented.
Background & objectives:Pathogenesis acute lymphoblastic leukaemia (ALL) in adults is not well understood, as it is more common in children. We examined the immunological status and the activity of certain enzymes in blood lymphocytes in adult patients of ALL at different stages.Methods:ALL patients (n=71) admitted during 2000-2005 were included in this study. All patients had decreased T-lymphocytes content. At first attack, they had CD4 +-cells decreasing and increasing IgM and IgG concentration. In complete remission all examined parameters were low. The peculiarities of ALL recurrence were high NK-cells content and disbalances of the main immunoglobulin concentrations.Results:In the first attack and recurrence the anaerobe glucose oxidation intensity and the reactions of macromolecular synthesis were lower in lymphocytes compared to control. In remission all these processes restored to normal. In all stages in lymphocytes GR had decreased activity.Interpretation & Conclusions:Our results showed that most of changes in immune status of ALL patients were in a stage of complete remission when patients arrived on its maintenance through the small period from spent before therapy when the immune system of the patient has not been restored. Thus, probably cytostatic action causes immune failure in the future and starts disease again.
Цель. изучить в динамике особенности гемостаза и функциональной активно-сти нейтрофильных гранулоцитов у пациентов с острым инфарктом миокарда (ОиМ) чувствительных и резистентных к ацетилсалициловой кислоте (АСК). Материал и методы. В исследование включены 53 пациента в первые 24 часа от развития ОиМ. Контрольная группа сформирована из 50 относительно здоровых добровольцев. Все пациенты до начала лечения и реваскуляриза-ции были обследованы на резистентность к АСК и разделены на группы чув-ствительных (ЧкАСК) и резистентных (РкАСК) к ацетилсалициловой кислоте. В комплексной терапии присутствовала двойная антитромбоцитарная тера-пия (АСК+клопидогрель). изучались показатели сосудисто-тромбоцитарного и коагуляционного гемостаза. Методом хемилюминесцентного анализа исследовалась функциональная активность нейтрофилов. Результаты. У РкАСК пациентов ОиМ в 1-е и на 10-е сутки выявляется увеличе-ние уровня АДФ-зависимой агрегации тромбоцитов (при дозе АДФ в 0,1 мкМ) и повышение уровня фактора Виллебранда. На 10-е сутки у ЧкАСК пациентов снижается уровень АДФ-зависимой агрегации тромбоцитов (при дозе АДФ в 5 мкМ), повышается содержание фактора Виллебранда. Независимо от чувстви-тельности к АСК у пациентов ОиМ повышен уровень РФМК, удлиняется тромби-новое время и снижено содержание Д-димеров. Кинетика хемилюминесценции нейтрофилов пациентов с ОиМ отличается от контрольной группы только увели-чением времени выхода на максимум. У резистентных к АСК пациентов в 1-е сутки обследования обнаружено снижение индекса активации нейтрофилов, величина которого на 10-е сутки лечения нормализуется. Заключение. У РкАСК пациентов с ОиМ наблюдается повышение АДФ-инду-цированной агрегации тромбоцитов, снижение их количества и увеличение уровня фактора Виллебранда. Риск тромбообразования у РкАСК на 10-е сутки лечения обусловлен сохраняющимся нарушением сосудисто-тромбоцитар-ного гемостаза и повышенной спонтанной агрегацией тромбоцитов. Состоя-ние коагуляционного гемостаза на 1-е и 10-е сутки ОиМ характеризуется активацией начальной стадии свертывания с увеличением длительности свертывания и значительным снижением содержания Д-димеров. При ОиМ выявляется замедленная скорость развития "респираторного взрыва" в ней-трофильных гранулоцитах, что, по-видимому, связано с компенсаторными процессами в организме и двойной антитромбоцитарной терапией, обладаю-щей противовоспалительным действием.
