Nod-like receptor pyrin domain-containing-3 (NLRP3) has been implicated in the pathogenesis of experimental renal injury, yet its characterization in human kidney disease remains largely unexplored. NLRP3 expression was evaluated in human kidney biopsies, primary renal tubular cells (HPTC) and correlated to disease outcomes in patients with IgA nephropathy (IgAN). NLRP3 localized to renal tubules in normal human kidney tissue and to mitochondria within HPTC by immunohistochemistry and immunofluorescence microscopy. Compared to control kidneys, NLRP3 gene expression was increased in biopsies of patients with IgAN. While NLRP3 expression in IgAN was detected in glomeruli, it remained largely confined to the tubular epithelial compartment. In vitro NLRP3 mRNA and protein expression were transiently induced in HPTC by TGF-β1 but subsequently diminished over time as cells lost their epithelial phenotype in a process regulated by transcription and ubiquitin-mediated degradation. Consistent with the in vitro data, low NLRP3 mRNA expression in kidney biopsies was associated with a linear trend of higher risk of composite endpoint of doubling serum creatinine and end stage renal disease in patients with IgAN. Taken together, these data show that NLRP3 is primarily a kidney tubule-expressed protein that decreases in abundance in progressive IgAN.

Thirteen species of Pratylenchidae: Pratylenchus coffeae, P. delattrei, P. loosi, P. neglectus, P. penetrans, P. pseudopratensis, P. thomei, P. vulnus, Pratylenchus sp., Pratylenchoides alkani, P. ritteri, Hirschmanniella sp. and Zygotylenchus guevarai were collected from different crops and plants throughout Iran. The specimens were identified using morphological and molecular methods. Morphometrics and morphology are given for Pratylenchus sp., P. delattrei, Pratylenchoides alkani and Hirschmanniella sp. The D2-D3 expansion segments of the 28S rRNA gene were amplified and sequenced for all 13 species studied. Diagnostic PCR-ITS-RFLP profiles are given for Pratylenchus delattrei, P. penetrans, P. pseudopratensis, Pratylenchus sp., Pratylenchoides alkani and P. ritteri. Pratylenchus neglectus and P. thomei, collected from cereal fields, P. loosi from tea plantations, P. coffeae from banana, P. penetrans from ornamental plants, P. vulnus from pines and Z. guevarai from almonds showed a high level of similarity in the D2-D3 sequences with corresponding GenBank sequences. Nucleotide differences between Iranian populations and reference species were in the intraspecific range. Pratylenchus delattrei, found in vegetable fields, and Pratylenchus sp. from palm rhizosphere, formed a highly supported clade with P. zeae, the two former species being morphologically very close to the latter except in tail shape. Pratylenchus pseudopratensis, from cereal fields, clustered with P. vulnus with low support. Phylogenetic relationships within Pratylenchus species were mainly congruent with those obtained in previous studies. Despite the morphological similarities between P. ritteri and P. alkani, the D2-D3 of 28S rRNA gene sequences differed by 5 bp. Hirschmanniella sp., from a rice field, formed a clade with H. loofi and H. kwazuna.

Abstract.Objective: Both adaptive and innate immune systems are involved in coronary artery disease (CAD). The aim of this study was to evaluate TH17 cytokines expression profiles in un-stimulated peripheral blood lymphocytes (PBMCs) of patients with coronary artery disease. Methods: Expression profiles of IL-17, IL-23, and TGF-β1 were determined in individuals with and without CAD using Real-time PCR.Results: A significant decrease in IL-23 gene expression in un-stimulated PBMCs of patients with CAD compared to those without CAD was found (p = 0.003, OR = 0.045, 95% CI: 0.006-0.355). Conclusion: Our data reinforce the potential role of the IL-23 as a critical regulatory molecule that bridges the innate and adaptive arms of the immune system in the complex mechanisms associated with the development of atherosclerosis.

BackgroundGrowth factors are generally believed to have a perpetuating role in the development of diabetic complications, However there is ample of evidence of a protective or therapeutic potential for some of them. IGF-I, according to some reports, may contribute to complication development, although a protective role for IGF-I has been claimed for all late diabetic complications, making it an exception among growth factors. Transforming growth factor (TGF)-β1 as a pleiotropic cytokine is a key player in immunoregulation. Dysregulation of TGF-β1 in diabetes has been addressed as a leading event of kidney pathologies, while there is no similar pivotal role for TGF-β1 in diabetic retinopathy or neuropathy. An association study was conducted to evaluate the distinctive roles of TGF-β1 and IGF-I in T1DM microvascular complications by gene variation-based regulatory mechanisms that are operational in modulation of both in situ and systemic levels of the gene product.MethodsTwo polymorphisms of the IGF-I gene at positions −383*C/T and −1089*C/T and two functional TGF-β1 gene polymorphisms, including codons 10 (+869*C/T) and 25 (+915*G/C) were examined in 248 British Caucasian T1DM patients and 113 healthy controls.ResultsThe distribution of IGF-1 gene polymorphisms did not reflect any significant association with different endpoints among the cases or different subgroups (complication triad) and controls. For TGF-β1 gene codon 25 polymorphism the low producer variant (allele C) were more frequent in cases than controls, which is compatible with the anti-inflammatory role of TGF-β1 and for codon 10 polymorphism the frequency of allele C was highest in retinopaths and, on the contrary and expectedly, nephropathy was more frequently accompanied by allele T (high producer). The frequency of allele G (high producer) of codon 25 polymorphism was slightly higher in the complication free group than in other subgroups.ConclusionAlthough there were some differences in distribution of allele and genotype frequencies of TGF-β1 gene polymorphism in diabetes microvascular complications the differences were not statistically significant. Regarding IGF-1 our result firstly questions the functionality of the employed polymorphic marker and secondly may entail that the main regulator of IGF-I functionality resides elsewhere rather than the IGF-I gene itself, such as post-transcriptional regulation.

Solitary extramedullary plasmacytomas are plasma cell tumors that tend to develop in mucosa-associated lymphoid tissues including the sinonasal or nasopharyngeal regions. Primary plasmacytoma of the lung is exceedingly rare and often presents as a solitary mass or nodule in mid-lung or hilar areas and diagnosed after resection. Herein, we report a case of primary pulmonary plasmacytoma that presented with diffuse alveolar consolidation and diagnosed by transbronchial lung biopsy.
