Objectives The aim of this study was to evaluate the addition of chitosan nanoparticle with concentrations of 0, 10, 20, and 30% to the epoxy resin-based (ERB) sealer on its antibacterial and cytotoxicity effect. Methods and Materials This research was divided into two studies, the first study was the addition of chitosan with a concentration of 0% (as control), 10, 20, and 30% to an ERB sealer on its antibacterial effect, and the second study was on its cytotoxicity. An agar diffusion test was employed to determine the antibacterial effect on Enterococcus faecalis. An MTT (3-{4,5-dimethylthiazol-2-yl}-2,5-diphenyl tetrazolium bromide) assay was utilized to test the cytotoxicity by evaluating cell viability. Statistical Analysis One-way analysis of variance and Tukey's test (α = 0.05) were used to analyze data obtained from each evaluation with a significance level of 95%. Results The addition of chitosan nanoparticles at concentrations 10, 20, and 30% produced a greater inhibition zone of E. faecalis (p < 0.05), however, had less cytotoxicity compared with no addition of chitosan (0%) (p < 0.05). Conclusion The addition of chitosan nanoparticles at concentrations 10, 20, and 30% to the ERB sealer produced greater antibacterial and less cytotoxicity compared with no addition of chitosan (0%).
ABSTRAKIn office bleaching menggunakan hidrogen peroksida 40% sering memberikan efek samping berupa linu baik selama maupun setelah perawatan tersebut dilakukan. Penelitian ini dilakukan dengan tujuan untuk mengetahui pengaruh waktu aplikasi dan kandungan fluor desensitizing agent pada metode in office bleaching terhadap kandungan mineral gigi. Delapan gigi masing-masing dipotong menjadi 4 bagian, 6 potong digunakan untuk pemeriksaan XRD awal. Gigi dikeringkan dalam oven suhu 50° selama 30 menit diserbuk, diambil 1 mg untuk diperiksa kandungan mineralnya dengan goniometer. Dua puluh empat potong yang lain dibagi dalam 4 kelompok perlakuan. Kelompok I: gigi diaplikasi bahan bleaching 0,5-1 mm hidrogen peroksida 40% selama 1 jam, dicuci, dikeringkan, diikuti aplikasi 0,1 ml desensitizing agent tanpa fluor (CPP-ACP) selama 30 menit, dicuci, dikeringkan. Kelompok II : gigi diaplikasi CPP-ACP 30 menit, dicuci, dikeringkan, dibleaching menggunakan hidrogen peroksida 40% selama 1 jam. Selanjutnya gigi diaplikasi CPP-ACP 30 menit lagi, dicuci, dikeringkan. Kelompok III gigi dibleaching menggunakan hidrogen peroksida 40% selama 1 jam, dicuci, dikeringkan, diikuti aplikasi desensitizing agent yang mengandung fluor (CPP-ACFP) selama 30 menit, dicuci, dikeringkan. Kelompok IV: gigi diaplikasi CPP-ACFP 30 menit, dicuci , dikeringkan, dibleaching menggunakan hidrogen peroksida 40% selama 1 jam. Selanjutnya gigi diaplikasi CPP-ACFP selama 30 menit, dicuci dikeringkan. Sesudah perlakuan, semua gigi dilakukan pemeriksaan kandungan mineral gigi dengan prosedur yang sama. Uji Mann Whitney menunjukkan penurunan mineral paling sedikit terjadi pada kelompok IV (4500 Gi. Desember 2014; 21(2): 136 -140
Latar Belakang. Penelitian seputar ozonazed water akhir-akhir ini cukup banyak diminati karena bahan ini mempunyai potensi antibakteria yang menjanjikan. Tujuan Penelitian ini adalah untuk mengetahui pengaruh ozonated water pada pertumbuhan bakteri Staphylococcus Aureus. Metode Penelitian. Penelitian ini dilakukan dengan cara mencampurkan 1 ml larutan bakteri 106 CFU/ml dengan 10 ml ozonated water konsentrasi 4 ppm selama 10 detik (kelompok I), 20 detik (kelompok II), 30 detik (kelompok III), dan 40 detik (kelompok IV), akuades (kontrol negatif) kemudian semua diencerkan dengan akuabides 10ml. selanjutnya dari masing-masing kelompok diambil 0,01 ml untuk ditanam pada MHA, inkubasi 24 jam pada suhu 370 C dalam anaerobic jar kemudian dilakikan perhitungan koloni. Data dianalisis dengan Avana satu jalur dilanjutkan dengan uji LSD. Hasil. Avana satu jalur memperlihatkan adanya perbedaan yang bermakna antar kelompok perlakuan. Uji LSD menunjukkan perbedaan rerarta yang bermakna (p<0,05) antar kelompok I, II, III, IV dengan kelompok V (akuedes) terdapat perbedaan rerata yang bermakna antara kelompok I dan kelompok VI (iod). Tidak terdapat perbedaan bermakna antara kelompok II, III, IV terdapat kelompok VI (oid). Uji korelasi Produk Momen Pearson menunjukkan adanya korelasi negatif yang sangat kuat. Kesimpulan. Ozonated Water 4 ppm memiliki daya antibakreri dalam menghambat pertumbuhan Staphylococcus Aureus. Semakin lama waktu kontak, daya anti bakterinya semakain besar. Background. Some previous research show that the antibacterial effect of ozonated water is very promi-sing. The aim of this study was to know the antibacterial effect of ozonated water on staphylococcus aureus. Method. One ml of 106 CFU/ ml S. aureus suspension was mixed with 10 ml ozonated water for 10 second (group I), 20 second (group II), 30 second (group III), 40 second (group IV)As negative control S. aureus was mixed with aquadest (group V), and as positive control S. aureus was mixed with iod (groupVI). Furthermore, 0,01 ml from each group was cultivated on MHA, incubated for 24 hours-37C followed by colony caunting. Data was analyzed using one way anova followed by LSD. Result. One way anova show significant difference among the group and LSD test show significant mean different between group I, II, III, IV, and V. Significant difference can also be seen between group I and V. No significant difference between group II, III, IV and VI. Product momen test show strong negative correlation. Conclusion. Ozonated water 4 ppm has antibacterial effect on S. aureus. The longer the contact time, the stronger the antibacterial effect.
Root canal configuration influence the succeed of root canal treatment, because the morphological complexity of the root canal makes the procedure for preparation and obturation of the root canal more challenging. The anatomy and morphology of the root canal can be classified according to the Vertucci root canal classification system. In the Vertucci type V classification at the apical third of root the canal is divided into two separate root canals with two different apical foramen. This condition requires appropriate obturation techniques in order to obtain a hermetic seal. Case report: A 19 year old female patient wanted her upper left posterior tooth to be treated. Post dental history revealed there was spontaneous pain in the tooth, particularly happened at night. There was no pain detected during examination. The objective examination showed there was secondary caries in mesioincisal area of maxillary left first premolar tooth. Percussion, palpation and thermal vitality test was negative. The periapical radiograph revealed a radiolucent area at the apex of the tooth. The diagnosis established for this tooth was partial pulp necrosis with asymptomatic apical periodontitis. The configuration of the root canal was established as Vertucci type V. Root canal treatment using rotary crown down preparation technique and obturation with warm vertical condensation were performed, followed by class II composite resin restoration reinforced by short fiber reinforced composite. Conclusion: Root canal treatment with warm vertical condensation obturation technique is the appropriate treatment of choice for Vertucci type V root canal configuration.
