Two classes of nucleic acids, bacterial DNA containing unmethylated CpG motifs and dsRNA in viruses, induce the production of type I IFN that contributes to the immunostimulatory effects of these microbial molecules. Thus, it is important to determine which cells produce type I IFN in response to CpG DNA and dsRNA. CD4+CD11c− type 2 dendritic cell precursors (pre-DC2) were identified as the main producers of type I IFN in human blood in response to viruses. Here we asked whether pre-DC2 also produce type I IFN in response to CpG DNA and dsRNA. Oligodeoxynucleotides containing particular palindromic CpG motifs induced pre-DC2, but not CD11c+ blood DC or monocytes, to produce IFN-α. In contrast, a synthetic dsRNA, polyinosinic polycytidylic-acid, induced CD11c+ DC, but not pre-DC2 or monocytes, to produce IFN-αβ. These data indicate that CpG DNA and polyinosinic-polycytidylic acid stimulate different types of cells to produce type I IFN and that it is important to select oligodeoxynucleotides containing particular CpG motifs to induce pre-DC2 to produce type I IFN, which may play a key role in the strong adjuvant effects of CpG DNA.
We introduce a novel parallel-aligned liquid-crystal (LC) spatial light modulator (SLM) that has been designed to operate in a phase-only mode for wave-front correction. We measured and analyzed theoretically the electro-optic characteristics of the LC SLM and obtained a peak-to-valley value of 0.07049lambda(lambda= 0.6328microm) after correction. A Strehl ratio of 0.989 indicates the approximate upper limit of an aberrated wave front that the LC SLM can correct when it is used in an adaptive optical system.
We demonstrate efficient coupling to the optical whispering gallery modes (WGMs) of nematic liquid crystal (NLC) microdroplets immersed in an immiscible aqueous environment. An individual NLC microdroplet, confined at the tip of a microcapillary, was coupled via a tapered optical fiber waveguide positioned correctly within its vicinity. Critical coupling of the taper-microdroplet system was facilitated by adjusting the gap between the taper and the microdroplet to change the overlap of the evanescent electromagnetic fields; efficient and controlled power transfer from the taper waveguide to the NLC microdroplet is indeed possible via the proposed technique. We also found that NLC microdroplets can function as highly sensitive thermal sensors: A maximum temperature sensitivity of 267.6 pm/°C and resolution of 7.5 × 10-2 °C were achieved in a 78-μm-diameter NLC microdroplet.
