Aim. In recent decades, Ukrainian breeders have created a large number of sweet cherry cultivars. Further progress in the breeding of sweet cherry requires a broad involvement of molecular methods. Especially important is the development of methods for the identification of genes / alleles that control economically valuable traits. The goal of the study was to develop a new method for discrimination of alleles of the PavCNR12 gene, which controls the fruit size in sweet cherry, and to reveal the allelic status of PavCNR12 in Ukrainian sweet cherry cultivars. Methods. The SNP-polymorphisms in the promoter regions of the PavCNR12-1, -2 and -3 alleles was detected applying comparison of published sequences. PCR amplification of the region was conducted, the obtained PCR products were cut by TaiI restriction endonuclease and separated by electrophoresis in a polyacrylamide gel. The identity of PCR products was confirmed by direct sequencing. Results. A new convenient method for the identification of allelic variants of the PavCNR12 gene using CAPS-markers is proposed. Using the method the allelic status of PavCNR12 in 56 sweet cherry cultivars of Ukrainian and foreign breeding was elucidated. Conclusions. A significant prevalence of the desirable allele PavCNR12-1 over the alleles PavCNR12-2 and -3 was found among the studied cultivars.Keywords: Ukrainian sweet cherry cultivars, genetic control of fruit size, alleles of PavCNR12 gene, CAPSmarkers, Prunus avium.
Мета. Українськими селекціонерами створено велику кількість сортів черешні, які все ще залишаються майже недослідженими на молекулярно-генетичному рівні. Метою роботи було провести ідентифікацію алелів самонесумісності (S-алелів) в українських сортів і форм черешні та з'ясувати їх приналежність до груп перехресної несумісності. Методи. Для генетичного профілювання сортів черешні було застосовано ПЛР із виродженими праймерами до першого та другого інтронів гена S-РНКази та до єдиного інтрону гена SFB. Електрофоретичний аналіз ПЛР-продуктів другого інтрону S-РНКази проводили в агарозному гелі, а детекцію флуоресцентно мічених фрагментів ДНК першого інтрону S-РНКази та інтрону SFBна генетичному аналізаторі. Результати. Ідентифіковано S-алелі у 25 сортів черешні української селекції та десяти культивованих форм (ландрас). Оцінено частоти зустрічальності S-алелів та розподіл сортів та ландрас по групах перехресної несумісності, що може бути використано в селекційній роботі та при плануванні промислових насаджень. Висновки. У досліджуваній вибірці виявлено 12 різних S-алелів та 79 S-гаплотипів. Алелі S1, S3, S4, S5, S6 та S9 є найбільш поширеними серед українських сортів і форм черешні. Високі частоти зустрічальності алелів S5 та особливо S9 характерні для українських сортів і відрізняють їх від інших європейських. XXXVII (S5S9) група перехресної несумісності є найчисленнішою серед українських сортів черешні.Ключові слова: українські сорти черешні, S-локус, S-генотипи, перехресна несумісність, Prunus avium. ступ. За наявністю та використанням генетичного різноманіття культурних рослин Україна є однією з найбагатших країн світу, входить до десятки країн, що зробили найбільший вклад у розвиток селекції черешні [1] та до десятки найбільших виробників плодів цієї культури [2]. Для подальшої цілеспрямованої селекції рослин вкрай важливим видається вивчення їх походження, генетичної спорідненості та різноманіття з використанням сучасних молекулярних методів [3, 4, 5]. Нещодавно аналогічні дослідження було проведено і для черешні [6, 7, 8]. Зокрема, застосування у селекційній роботі молекулярних маркерів, зчеплених із господарсько-цінними ознаками суттєво полегшує та прискорює отримання нових сортів [9, 10]. До таких ознак у черешні належать маса та забарвлення плодів, самонесумісність, діаметр штамбу тощо [9,[11][12][13].Черешня (Prunus avium L.) є аллогамною рослиною. Як і в інших представників родин Розові (Rosaceae), у черешні запилення контролюється системою гаметофітної самонесумісності (ГСН; gametophytic self-incompatibility, GSI). Ця система генетично контролюється одним мультиалельним локусом (S) з двома генами, які кодують специфічні детермінанти реакції самонесумісності. Aim. To estimate the possibilities of using the promising apple cultivars, created by the Institute of Horticul- ture (IH) NAAS, in the breeding programs according to the availability of valuable allele variants of gene Rf, related to red color of apple fruit. Methods. Polymerase chain reaction, electrophoresis in agarose gel, the evaluation of phenotype manifestation of the color trait of apple skin according to the grading system. Results. 17 genotypes of apple cultivars of different genetic origin, including 12 cultivars, breeded by IH NAAS, were identifi ed by allele variants of gene Rf. The comparison was made between the genetic analysis results and visual observations of the phenotype manifestation of fruit skin color of different apple cultivars. Conclusions. Apple cultivars Amulet, Perlyna Kyeva and Edera, which are homozygous carriers of dominant alleles А 1 and А 2 of gene Rf are recommended as basic forms in selection programs. It was demonstrated that exact prediction of apple skin color was possible only in case of the available homozygous genotype by both alleles of gene Rf.
