This study evaluated the loss of desiccation tolerance in C. langsdorffii seeds during the germination process. Seeds were imbibed for 24, 48, 72, 96, 120 and 144 hours and dried to the initial moisture content, kept in this state for 3 days after which they were submitted to pre-humidification and rehydration. Ultraestructural evaluations were done aiming to observe the cell damage caused by the dry process. Desiccation tolerance was evaluated in terms of the percentage of normal seedlings. Seeds not submitted to the drying process presented 61% of normal seedlings, and after 24 hours of imbibition, followed by drying, the seeds presented the same percentage of survival. However, after 48 hours of imbibition, seeds started to lose the desiccation tolerance. There was twenty six percent of normal seedlings formed from seeds imbibed for 96 hours and later dried and rehydrated. Only 5% of seeds imbibed for 144 hours, dried and rehydrated formed normal seedlings. At 144 hours of imbibition followed the dry process, there was damage into the cell structure, indicating that the seeds were unable to keep the cell structure during the drying process. Sementes foram embebidas por 24, 48, 72, 96, 120 e 144 horas e depois secas até a umidade inicial, sendo mantidas neste estado durante 3 dias, quando então foram submetidas a pré-umidificação e reidratação. Avaliações ultraestruturais foram realizadas objetivando observar danos nas células causados pelo processo de secagem. A tolerância à dessecação foi avaliada pelo percentual de plântulas normais. Sementes não submetidas ao processo de secagem apresentaram 61% de plântulas normais, sendo que após 24 horas de embebição seguida de secagem, houve o mesmo percentual de sobrevivência. Contudo, após 48 horas de embebição, as sementes começaram a perder a tolerância à dessecação. Vinte e seis por cento de sementes formaram plântulas normais após embeberem por 96 horas e secas. Apenas 5% de sementes embebidas por 144 horas e secas formaram plântulas normais. Após 144 horas de embebição, seguida de secagem, verificou-se danos na estrutura celular, o que indica que as sementes não são capazes de manter a estrutura celular durante o processo de secagem. Verificou-se no presente estudo que sementes de C. langsdorffii perdem a tolerância à dessecação no inicio da fase 2 da embebição.Palavras-chave: Copaíba, sensibilidade à dessecação, embebição.
Specific anti-B19 IgM was demonstrated in sera from three children showing transient aplastic crisis. A two years-old boy living in Rio de Janeiro suffering from sickle-cell anaemia showed the crisis during August, 1990. Two siblings living in Santa Maria, RS, developed aplastic crisis during May, 1991, when they were also diagnosed for hereditary spherocytosis. For a third child from this same family, who first developed aplastic crisis no IgM anti-B19 was detected in her sera.
-The objective of this experiment was to evaluate the effect of priming on the germination of Eucalyptus urophylla and of hybrid E. urophylla × E. grandis seeds under salt stress. Two osmotic potentials (-1.0 and -1.5 MPa) were tested, using PEG 6000 for 1 and 3 days. After priming, seeds were germinated under salt stress in a NaCl solution at 0.0 (control), -0.5, -0.75 and -1.0 MPa potentials, at 25 °C. Seed germination and germination speed index decreased as the water potential of the germination medium decreased. However, E. urophylla was more tolerant to salt stress; it showed a higher germination percentage under all tested potentials, when compared to the hybrid. The osmotic conditioning at -1.0 MPa for three days was more effective when E. urophylla x E. grandis was germinated in a salt solution at -1.0 MPa, indicating that this treatment was more effective in inducing tolerance to salt stress.
Resumo -O objetivo deste trabalho foi avaliar diferentes meios de cultura, utilizados sobre o ponto da enxertia, na microenxertia ex vitro para a eliminação do Cowpea aphid-borne mosaic virus (CABMV), em plantas de maracujá-azedo (Passiflora edulis Sims f. flavicarpa Deg.). Ápices caulinares, provenientes de plantas infectadas, foram microenxertados em plântulas obtidas pela germinação de sementes em substrato comercial esterilizado. Foram conduzidos experimentos com a microenxertia realizada no hipocótilo e no epicótilo, e foram utilizados cinco meios de cultura, que diferiam na concentração de fitorreguladores, aplicados no local da enxertia. O índice médio de microenxertos com folha expandida foi de 27,22 e 32,22%, quando a microenxertia foi realizada no hipocótilo e no epicótilo, respectivamente. Na microenxertia realizada no hipocótilo, não houve efeito da aplicação de meios de cultura. Na microenxertia realizada no epicótilo, o meio MS acrescido de 0,1 mg L -1de AIB e 1 mg L -1 de BAP proporcionou 53,3% de microenxertos com folha expandida, número superior aos demais tratamentos e maior desenvolvimento das brotações. A indexação realizada pelo teste ELISA indireto, 80 a 100 dias após a microenxertia, mostrou que 93% das plantas testadas não apresentavam vírus detectável.Termos para indexação: Cowpea aphid-borne mosaic virus, Passiflora edulis, endurecimento dos frutos, local de enxertia. Ex vitro micrografting aiming the CABMV virus elimination in passion fruit plantAbstract -The objective of this work was to evaluate the effect of different culture media, used over grafting spot, in ex vitro micrografting, in order to eliminate the Cowpea aphid-borne mosaic virus (CABMV) in sour passion fruit plants (Passiflora edulis Sims f. flavicarpa Deg.). Cauline apexes, originated from infected plants, were micrografted in seedlings obtained by the germination of seeds in commercial sterilized substrate. Experiments were conducted with the micrografting performed on the hypocotyl and epicotyl using five culture media, which differed on their concentration of phytoregulators applied on the grafting spot. The average rate of micrografting showing expanded leaf was 27.22 and 32.22%, when micrografting was performed on hypocotyl and epicotyl, respectively. In the micrografting performed on the hypocotyl, there was no effect of the culture media applied. In the micrografting performed on epicotyl, the MS medium supplemented with 0.1 mg L -1 3-indolbutiric acid (IBA) and 1 mg L -1 6-benzylaminopurine (BAP) resulted in 53.3% of micrografting showing expanded leaf, a number superior to the other treatments, and in a larger development of shoots. The indexing performed by the indirect ELISA test, within 80 to 100 days after micrografting, showed that 93% of the plants tested presented no detectable virus.
-This study was aimed at evaluating the desiccation sensitivity in seeds of the tree Tapirira obtusa (Benth.) J. D. Mitchell collected from three different environments and subjected to two distinct drying speeds. Seeds were collected from a rocky area, in the "Cerrado", and in a riparian forest area, in the region of municipality of Lavras, State of Minas Gerais. The seeds were subjected to drying with magnesium chloride (slow drying) or silica gel (fast drying), into closed environment, until moisture contents of 40%, 30%, 20% and 10%, considering as control, the percentage of germination at the initial moisture content in each environment, which varied from 47% to 50%. Percentages of germination and normal seedlings as well as germination speed index were assessed. For the three environments studied, there was no effect of slow drying on seed germination. Seeds from area of Cerrado, however, have shown a slight reduction on germination when subjected to fast drying. Oppositely, seeds from rocky area had germination increased when subjected to fast drying. Seeds from riparian forest area had no reduction on germination percentage, independent of drying speed. Results suggest that seeds of T. obtusa are not sensitive to desiccation.
