While to date investigations provided convincing evidence on the role of aristolochic acids (AAs) in the etiology of Balkan endemic nephropathy (BEN) and upper urothelial cancer (UUC), the exposure pathways by which AAs enter human bodies to cause BEN and UUC remain obscure. The goal of this study is to test the hypothesis that environmental pollution by AAs and root uptake of AAs in the polluted soil may be one of the pathways by which AAs enter the human food chain. The hypothesis driving this study was that the decay of Aristolochia clematitis L., a AA-containing herbaceous plant that is found growing widespread in the endemic regions, could release free AAs to the soil, which could be taken up by food crops growing nearby, thereby transferring this potent human nephrotoxin and carcinogen into their edible parts. Using the highly sensitive and selective high-performance liquid chromatography coupled with fluorescence detection method, we identified and quantitated in this study for the first time AAs in corn, wheat grain, and soil samples collected from the endemic village Kutles in Serbia. Our results provide the first direct evidence that food crops and soil in the Balkans are contaminated with AAs. It is possible that the presence of AAs in edible parts of crops originating from the AA-contaminated soil could be one of the major pathways by which humans become exposed to AAs.

Prolonged exposure to aristolochic acid (AA) was shown to pose rapid progressive renal fibrosis in Belgian women in a slimming regime in the early 1990s. AA was also demonstrated to be strong carcinogen in rats. The carcinogenicity of AA is generally believed to be related to the nitro-reduction of AA, in which the aristolactam-nitriumion ion with a delocalized positive charge is the ultimate carcinogen. In this study, the phase I and phase II metabolism of AA was investigated by using an in vitro system with rat liver S9 and an in vivo animal study with Sprague-Dawley rats. AA was found to have been undergone hydroxylation, lactam formation, and desnitro and desmethyl transformations. Three conjugated metabolites of AA, namely the N- and O-glucuronides of aristolactams, were detected directly in pre-concentrated urine sample, with no acid hydrolysis or enzymatic digestion. Structural elucidation of the metabolites was performed by using liquid chromatography/tandem mass spectrometry (LC/MS/MS). The results indicated that N-glucuronidation was the major phase II metabolic pathway for the aristolactams formed by AA after their nitro-reduction.

Exposure to aristolochic acids (AAs) from Aristolochia plants is one of the major global causes of nephropathy, renal failure and urothelial cancer, including Balkan endemic nephropathy (BEN). The high incidence of BEN on the Balkan Peninsula is assumed to result from consumption of Aristolochia clematitis L. seeds co-harvested with crops. Here we show that AAs are long-lived soil contaminants that enter wheat and maize plants by root uptake, with strong pH-dependence. Soil and crops from Serbian farms in areas endemic for A. clematitis were found to be extensively contaminated with AAs, with contamination strongly correlated with local incidence of BEN. The persistence of AAs as soil contaminants suggests that weed control for A. clematitis plants is needed to reduce the incidence of BEN and aristolochic acid nephropathy, and that systematic surveys of soil and crop AA levels would identify high-risk regions and it is imperative to research soil remediation methods.

We present the first proteomic analysis on the cellular responses to avian influenza virus (H9N2) infection in a human cell line in different time courses in order to search for target proteins for viral pathogenesis/adaptation studies. By using 2-DE coupled with MALDI-TOF MS and nano-ESI-MS/MS, we identified a set of differentially expressed cellular proteins, including cytoplasmic actin, cytokeratin, prohibitin, enoyl-CoA hydratase, peptide-prolyl cis-trans isomerase A (PPIase A), chloride intracellular channel protein 1, pyruvate dehydrogenase E1 component subunit beta, adenine phosphoribosyltransferase, guanine nucleotide-binding protein subunit beta, nucleoside diphosphate kinase A, elongation factor 1-beta and splicing factor, arginine/serine rich 1. The most significant changes in different time courses were found in cytoplasmic actin and cytokeratin, both of which constituted the major components of cytoskeleton network in the cells. The obtained data suggested a possible role of the cytoskeleton during avian influenza virus infection of mammalian cells, which might help for better understanding of the dynamics of avian influenza virus and host interaction in mammalian cell setting.

Fragmentation reactions of protonated α-amino acids (AAs) were studied previously using tandem mass spectrometry (MS/MS) of unit mass resolution. Isobaric fragmentation products and minor fragmentation products could have been overlooked or misannotated. In the present study, we examined the fragmentation patterns of 19 AAs using high-resolution electrospray ionization MS/MS (HR-ESI-MS/MS) with collision-induced dissociation (CID). Isobaric fragmentation products from protonated Met and Trp were resolved and identified for the first time. Previously unreported fragmentation products from protonated Met, Cys, Gln, Arg, and Lys were observed. Additionally, the chemical identity of a fragmentation product from protonated Trp that was incorrectly annotated in previous investigations was corrected. All previously unreported fragmentation products and reactions were verified by pseudo MS 3 experiments and/or MS/MS analyses of deuterated AAs. Clearer pictures of the fragmentation reactions for Met, Cys, Trp, Gln, Arg and Lys were obtained in the present study.

Chemical isotope labeling (CIL) liquid chromatography mass spectrometry (LC-MS) is a powerful technique for in-depth metabolome analysis with high quantification accuracy. Unlike conventional LC-MS, it analyzes chemical-group-based submetabolomes and uses the combined results to represent the whole metabolome. Due to analysis time and cost constraint, not all submetabolomes can be profiled and thus knowledge of chemical group classification is important in guiding submetabolome selection. Herein we report a study of determining the distribution of functional groups of compounds in a database and then examine how well we can experimentally analyze the major chemical groups in two representative samples (i.e., human plasma and yeast). We developed a computer algorithm to classify chemical structures according to their functional groups. After removing lipids which are targeted molecules in lipidomic analysis, inorganic species and other molecules that are unique to drug, food, plant, and environmental origins, five groups (i.e., amine, phenol, hydroxyl, carboxyl, and carbonyl) are found to be the dominant classes. In the databases of MCID (2683 filtered metabolites), HMDB (5506), KEGG (11598), YMDB (1107), and ECMDB (1462), 94.7%, 85.7%, 86.4%, 85.7%, and 95.8% of the filtered metabolites belong to one or more of the five groups, respectively. These groups can be analyzed in four-channel CIL LC-MS where hydroxyls (H), amines and phenols (A), carboxyls (C), and carbonyls or ketones/aldehydes (K) are separately profiled as individual channels using dansyl and DmPA labeling reagents. A total of 7431 peak pairs were detected with 6109 unique-mass pairs from plasma, while 5629 pairs with 4955 unique-mass pairs were detected in yeast. Compared to group distributions of database compounds, hydroxylcontaining metabolites were severely underdetected, which might indicate that the current method is less than optimal for analyzing this group of metabolites. As a result, the overall experimental coverage is likely significantly lower than the databasederived coverage. In short, this study has shown that high metabolome coverage is theoretically attainable by analyzing only the H, A, C, and K submetabolomes and the group classification information should be helpful in guiding future analytical method development and choices of submetabolomes to be analyzed.

ABSTRACT:The metabolic activation of aristolochic acids (AAs) that have been demonstrated to be mutagenic and carcinogenic was investigated. In vitro metabolism study indicated that AAs were metabolized to N-hydroxyaristolactam, which could be either reduced to aristolactams or rearranged to 7-hydroxyaristolactams via the Bamberger rearrangement. In vivo metabolism study is important because the intermediates (aristolactam-nitriumion) of the nitroreduction process are thought to be responsible for the carcinogenicity of AAs. Liquid chromatography-mass spectrometry and liquid chromatography-tandem mass spectrometry (MS/MS) were applied to the analyses of a series of positional isomers of hydroxyaristolactams in rat urine samples after the in vivo study of AAs. Three hydroxylated metabolites of aristolactam II and two hydroxylated metabolites of aristolactam I were identified. The structures of the positional isomers were elucidated from the interpretation of MS/MS spectra and theoretical calculations. In addition, several new metabolites were detected in the rat urine by high-resolution mass spectrometry and MS/MS, including those from the decarboxylation of AAs and the conjugations of acetylation, glucuronidation, and sulfation of aristolochic acid Ia.
