It is shown that diethylaminoethyl‐cellulose (DEAE‐cellulose) columns can be used for efficient chromatographic fractionation of polysaccharide mixtures. At about pH 6 neutral polysaccharides are not or only weakly adsorbed by DEAE‐cellulose. They can be readily eluted at the same pH by buffers of increasing strength. Neutral polysaccharides are adsorbed by DEAE‐cellulose in the hydroxyl form and can be fractionated by elution with NaOH of increasing strength. Acidic polysaccharides like pectic acid are adsorbed readily at neutral and weakly acidic pH values, but can only be eluted by increasing the pH. The use of the borate form of DEAE‐cellulose and elution with Na‐borate of increasing strength increases the fractionation effect. Examples are given for the fractionation of wheat starch dextrin, a mixture of sugar beet araban and pectic acid, sugar beet pectic acid, a mixture of wheat pentosan and serum albumin, and of the cold water‐soluble wheat flour polysaccharides.

Wird mit verschiedenen Mitteln zum gleichen Veresterungsgrad partiell verseift, so entstehen Pektine, die sich deutlich voneinander unterscheiden, SO in Bezug auf Loslichkeit, Dissoziation der Carboxylgruppen, Flockbarkeit durch Elektrolyte, Selektivitat fur Kationen, elektrophoretisches Verhalten, Viskositat, Geliervermogen, Stabilitat der restlichen Methylester-Gruppen und Angreifbarkeit durch Pektinenzyme. Man nimmt an, dass diese Unterschiede durch verschiedene interund intramolekulare Verteilung der Estergruppen bedingt sind. Mit rein cheniischen Methoden hat man diese Annahme noch nicht bewiesen.Der Mechanismus der Pektinverseifung hangt stark von dem Verseifungsmittel ab. Nach der partiellen Verseifung durch Suure durften die freigelegten COOH-Gruppen weitgehend statistisch verteilt sein. Durch die Saure wird die Dissoziation der COOH-Gruppen des Pektins zuruckgedrangt, und dadurch werden intramolekulare elektrostatische Wechselwirkungen herabgesetzt. Bei tiefem pH und tiefer Temperatur kann ohne nennenswerte Nebenreaktionen, wie Spaltung der Glykosidbindungen oder Decarboxylierung, verseift werden *). -Bei der partiellen Verseifung durch Alkali durften die freigelegten COO-Na+-Gruppen vorzugsweise einzeln zwischen restlichen COOCH,-Gruppen verteilt sein. Bei einem Pektin mit einem Veresterungsgrad von 50% durften die COO-Na+-und COOCH,-Gruppen alternieren. Dies kann aus der Kinetik der alkalischen Verseifung vermutet werden; die ((Reaktionskonstante o zweiter Ordnung nimmt im Laufe der Reaktion sehr stark ab, da die Makromolekeln immer starker negativ aufgeladen werden und die Hydroxyl-

IX. 61)Cellulose-Anionenaustauscher, die sich zur Fraktionierung von Proteinen und Nucleinsauren bewahrt haben, eignen sich auch zur Auftrennung von Polysaccharidgemischen 2-6). An Diathylaminoathyl-(DEAE)-Cellulose lassen sich auch Pektinpraparate fraktionieren3. Dabei konnte gezeigt werden, dass der grosste Teil der in Pektinstoffen stets vorhandenen neutralen Begleitkohlenhydrate sich von der Polygalakturonsaure (PGS) nicht trennen lassen und wahrscheinlich kovalent als Seitenketten gebunden sind. Diese Seitenketten bestehen vor allem aus Arabinoseund Galaktose-Bausteinen.In dieser Arbeit werden weitere chromatographische Trennungen von Pektinstoffen nach dem Gehalt an Seitenketten, nach dem Veresterungsgrad mit Methanol und nach dem Polymerisationsgrad der PGS-Hauptketten beschrieben. cber den Einfluss der Verteilung der Methylester-Gruppen langs dieser Hauptketten auf das Haftvermogen an DEAE-Cellulose wird in der folgenden Arbeit berichtet 7).Je nach Pflanzenart, Gewebeart, Wachstumsbedingungen, Wachstumsstadium, Extraktionsund Reinigungsmethode variiert die Zusammensetzung von Pektinpraparaten sehr stark. Pektinpraparate haben sich u. a. beim Ultrazentrifugieren, bei der Elektrophorese und bei der fraktionierten Fallung stets als heterogen erwiesens). Es ist kaum anzunehmen, dass in einem Pektinpraparat auch nur zwei Makromolekeln miteinander identisch sindo). Von Makromolekel zu Makromolekel konnen variieren : die Art und Menge der Seitenketten, der Veresterungsgrad der PGS-Hauptkette mit Methanol, die Verteilung der Methylester-Gruppen langs der Hauptkette, der Acetylierungsgrad und der Polymerisationsgrad der Hauptkette. Es sei betont, dass die Existenz kovalent gebundener Seitenketten noch nicht durch direkte chemische Methoden bewiesen, aber wegen verschiedener Beobachtungen angenommen wurde4) 8, lo).
