Five different morphological types of Maytenus ilicifolia of the same age and harvested under the same conditions showed distinct accumulations of some friedo-nor-oleananes. A rapid, sensitive and reliable reverse-phase HPLC method (employing an external standard) was used for the determination of the cytotoxic triterpenoids, 20 alpha-hydroxymaytenin, 22 beta-hydroxymaytenin, maytenin, celastrol and pristimerin in each of the five types. Well resolved peaks with good detection response and linearity in the range 1.0-100 micrograms/mL were obtained.
Visando a obtenção de triterpenos quinonametídeos em concentração apreciável protocolo para o desenvolvimento de culturas de células de M. ilicifolia foi estabelecido. Estudos visando observar a influência da luminosidade sobre o crescimento das culturas mantidas nas mesmas condições de temperatura e umidade, além da avaliação da influência de diferentes tipos de frascos, que variavam em altura, sobre o crescimento e acúmulo desses metabólitos, foram realizados. O acúmulo desses metabólitos com comprovada atividade biológica se mostrou 100 e 3 vezes maior para a 22beta-hidroximaitenina e maitenina, respectivamente, no sistema in vitro quando comparada à planta in natura. Isto demonstra que o sistema in vitro pode se constituir em uma excelente ferramenta para a produção, em maior escala, destes metabólitos bioativos.
Cell culture of Maytenus ilicifolia were established in order to produce and to quantify the antitumoral and antioxidant quinonemethide triterpenes. In vitro calli were induced from leaf explants of native plants and cultured in semi-solid medium under controlled conditions of humidity, temperature and photoperiod. The quinonemethide triterpenes showed maximum accumulation in the logarithmic phase growth of the cell culture. A rapid, sensitive and reliable reverse-phase HPLC method was used for quantitative determination of the antitumoral and antioxidant quinonemethide triterpenes, 22β-hydroxymaytenin and maytenin in callus of Maytenus ilicifolia. Well resolved peaks with good detection response and linearity in the range 1.0 -100 µg/mL were obtained. This quantitative work was performed by an external standard method.
