Nach 2stdg. Stehen verdünnt man mit 10 ml Chloroform und gibt bei 5-10' 12,O g I (0,05 Mol), in 60 ml Chloroform gelöst, langsam zu. Nach einstündigem Stchen kocht man noch 1-2 Std. im Wasserbad. Das erkaltete Gemisch wird in Wasser gegeben und rnit Chloroform ausgeschüttelt. Nicht umgesetztes Ausgangsmaterial n-ird mit n NaOH entfernt. AIS dem Chloroform werden 12,l g (75%) kristalher Riickstand erhalten. Kleine ii'iirfel aus Bt'hanol. I n der Kälte saugt man das kristalline Natriumsalz ab, löst wieder in wiiBBgem Alltohol und säuert mit 2 n HCl an. Ausheute 7,9 g. Gut umkristallisierbar aiis Äthanol. Schmp. 117,5O.2. %u 7,3 g gcpulvertcm AlCl, (0,055 Mol) in 25 ml Chloroform läBt, man 4,4 g Aeetylchlorid (0,056Mol) zutropfen. Danach werden 12,6g I (0,05 Mol) in GO ml Chloroform langsam ziigcgcbcn und anschlieBend P 5 Std. auf dem Wasserhad gekocht. Zur Aufarbeitung destillicrt man das Chloroform ab iind bringtrdas Rohprodukt in 2 n NaOH, der zur hesseren L6sung ctwas Alkohol zugefügt wird. Man erhitzt, bis sich der griiBte Teil gelöst hat, filtriert iinil IaBt im Eisschrank das Natriumsalz auskristallisieren. 13,3 g Ausbeute = 84%. F&llun$ der freien Verhindung aus wiiBrigem Alkohol. Uber 7-Halogenbuttersauren aus r-Butyrolacton und einige neue w -Amino -3 -alkyl-6 -halogen-ac ylanilide * ) Mitt.: Synthesen neuer Verbindungen mit lokalanasthetischer WirkungDie eindrucksvollen pharmakologischen Eigenschaften des lokalanasthesierenden a-Diathylamino-2,6-dimethyl-acetanilids (Xylocainm) haben zu Synthesen vieler basischer Saureamide angeregt, bei denen vornehmlich die aromatische Aminokomponente und der basische Rest des Leitmodells variiert worden sind, wiihrend die 2 C-Atom-Briicke zwischen den beiden N-Atomen iiberwiegend erhalten blieb. /3-Amino-propionamide wurden in wesentlich geringerem Umfang gepriift, Unter-suchungen uber r-Amino-butyramide, spez. y-Amino-butyranilide, sind kaum bekannt gewordenl). Diese Tatsache verwundert besonders, weil in der Reihe der isomeren 0-Amino-butyranilide, die z. B. durch Anlagerung von sekundaren Aminen an Crotonsaureanilide gewonnen werden konnen, zahlreiche intensiv anasthesierende Verbindungen mit guter Vertraglichkeit aufgefunden wurden2).Fur die Synthese der y-Amino-butyranilide benotigten wir y -Chlorbuttersaure, die entweder in freier Form oder als funktionelles Derivat (Halogenid **), Anhydrid, Ester) zm Kniipfung der -NH-CO-Bindung mit der aromatischen Aminkomponente eingesetzt wird. Lebensjahres in herzlicher Dankbarkeit ! * *) Das aus y-Butyrolacton und SOCI, in Gegenwart von ZoC1, unter Umgehung der Carbonsaurestufe direkt erhaltliche y-Chlorbutyrylchlorid vom Sdp.,,72-80° und n; ' 1,4539 war fur unsere Synthesen wegen merklicher Verunreinigungen nicht geeignet. Die argentometrische Cblorbestimmung dieses Produktes ergab nur 88-90~0 des theoretischen Chlorgehalts. Durch Fraktionierung lassen sich nach unseren Erfahrungen die Verunreinigungen nur unvollkommen und unter erheblichen Verlusten abtrennen. Der etwas geringere Gehalt erkliirt sich dadurch, da13 auch bei der Fraktionierung unter 1 Torr durch HC1-Abspaltung in Spuren y-Butyrolacton entsteht, das als dunkelroter Eisen-Hydroxamsiiurekomplex nachgewiesen wurde. Selbst hei der Destillation i. Hochvak. (0,02 Torr) erhiilt man nur cine 99proz. Siiure, ng 1,4509.12,3 g (0,l Mol) I und 11 g (0,l Mol + 10%) Acetanhydrid in 50 ml Benzol wurden 6 Std.unter RiickfluB erhitzt. Nach dem Abdestillieren des Benzols fraktionierte man den Ruckstand: 71% d. Th. I-anhydrid vom Sdp.,,, 129-130', farbloses 01, n y 1,4679. o-Amino-3-lthyl-6-halogen-acylanilideDie Synthese der in Tab. 3 aufgefuhrten o-Amino-acylanilide erfolgte im Prinzip nach dem bereits friiher veroffentlichten Verfahren18), also ohne Isolierung der als Zwischenprodukte auftretenden w-Chloracylanilide.
