RESUMO.-O uso terapêutico de antibióticos ionóforos em medicina veterinária difundiu-se muito nos últimos anos, com conseqüente aumento no risco de intoxicação em animais. Antibióticos ionóforos são usados como coccidiostáticos e como aditivo em alimentos para animais, com o propósito de estimular o desenvolvimento e o ganho de peso. Os ionóforos mais utilizados na alimentação de animais são a monensina, lasalocida, nasarina e salinomicina. Há uma grande variação na susceptibilidade dos efeitos tóxicos dos ionóforos de acordo com a espécie animal. A intoxicação pode ocorrer quando dosagens elevadas de ionóforos são adicionadas aos alimentos, ou quando ionóforos são incluídos inadvertidamente ou acidentalmente em dosagens não corretas para determinada espécie animal. Casos de intoxicação têm sido descritos em bovinos, ovinos, suínos, eqüinos, cães e aves. Para os eqüi-nos os ionóforos são extremamente tóxicos. São considerados seguros quando usados nas espécies-alvo, dentro das dosagens recomendadas pelo fabricante. The therapeutic use of ionophores in veterinary medicine has grown in the last years, with resultant increase in the risk of poisoning in animals. Ionophores are used as food additives as coccidiostacts in several animal species and growth promoter and bloat prevention in ruminants. The most often used ionophores are monensin, lasalocid, narasin and salinomycin. There is a great variation in the susceptibility to the toxic effect of ionophores in different animal species. Poisoning can occur when the dosage is too high or when not correct doses for a certain animal species are given. Cases of poisoning have been described in sheep, swine, horses, dogs and poultry. For horses ionophores are extremely toxic. The use of ionophores is only safe when used accordingly to the instructions of the manufacturer and especially for each animal species. In this paper the most important data regarding clinical-pathological and pathogenic aspects, and also the conditions in which the poisoning may occur are critically reviewed.

Bras. 29(7): 498-508, julho 2009 RESUMO.-Descreve-se a intoxicação natural por Pseudocalymma elegans em pelo menos um bovino em Rio Bonito, RJ e a reprodução experimental dessa intoxicação em três bovinos e em 3 coelhos com exemplares dessa planta colhida no local onde ocorreu a morte. Necropsia e histopatologia do bovino naturalmente intoxicado não revelaram alterações significativas. A administração, por via oral, de 1g/kg da brotação da planta causou a morte do bovino dentro de 5h e 30 minutos após o início da administração, já pela administração de 0,5 g/kg, a morte do Natural poisoning by Pseudocalymma elegans, a plant that causes "sudden death" and only occurs in the State of Rio de Janeiro, is described in a cow. This condition was experimentally reproduced in three calves and three rabbits with the plant collected in the area where the deaths had occurred. Postmortem and histopathological examinations of the natural case in the cow did not reveal significant alterations. Oral administration of 1g/ kg of the sprouts of P. elegans caused death of the calf 5 hours and 30 minutes after beginning of the administration. The dose of 0.5 g/kg caused death of another calf with onset of symptoms after 3 hours and 24 minutes and a course of 73 hours and 12 minutes. Animals that ingest borderline doses may show a longer course with a characteristic clinical picture of heart impairment. The dose of 0.25g/kg did not cause death, but only clinical signs. Heart palpitation and arrhythmia, increase of breathing frequency, reluctance in moving, positive venous pulse, ingurgitated jugular veins, falling down and peddling movements shortly before death, were observed. At postmortem examination the lesions were those of acute heart insufficiency, and histopathological examination revealed the typical kidney lesion (hydropic-vacular degeneration in the distal convoluted tubules) seen in poisoning by plants that cause "sudden death". In the experimentally poisoned rabbits the clinical course was less than 1-2 minutes. In the kidney of two rabbits there was also the characteristic microscopic lesion. The presence of the plant in the county of Rio Bonito had not been known by the veterinarians. It is concluded that the plant could be more toxic than described before.

Bras. 33(8) Inspite that Brachiaria spp. are most important forage grasses for beef cattle raising in Brazil, they can under certain conditions be toxic, cause outbreaks of hepatogenic photosensibilization and determine severe economic losses. Animals grazing toxic Brachiaria spp. commonly show in their liver foamy macrophages, isolated or grouped together, and crystals inside the biliary ducts. Steroidal saponins have been identified in these crystals and are liable for damage the liverleading to accumulation of phylloerytrin. Occasionally negative images of the crystals may be seen in the cytoplasm of foamy macrophages. The pathogenesis of formation and the type of material stored in the foamy cells is still unknown. The lectin histochemistry aims to assist in the detection of foamy macrophages and consequently in the diagnosis of the poisoning. Lectin histochemistry detects what kind of specific sugar residues exists in the cytoplasm of the foamy cells. The purpose of the present study was to identify the most suitable lectins the detection of steroidal saponins in the liver and kidney of sheep with photosensitization caused by Brachiara decumbens. Fragments of liver and kidney of fifteen sheep of both sexes and variable age, from Conceição do Mato Dentro, Minas Gerais, southeastern Brazil, and one sheep kept free grazing Brachiaria spp. were evaluated by lectin histochemistry. Fourteen lectins were used (Con-A, DBA, SBA, PNA, SJA, RCA-I, UEA-I, WGA, SWGA, GSL, PSA, PHA-L, PHA-E and LCA). It was found that the PNA lectin detected in the liver of sheep with photosensitization, caused by ingestion of Brachiaria decumbens, showed specificity and great binding reactivity for foamy macrophages, and also specificity and slight binding reactivity for hepatocytes. The WGA lectin showed specificity and moderate binding reactivity for foamy macrophages of the liver, and also specificity and slight binding reactivity for hepatocytes. The SBA, GSL and LCA lectins showed specificity and moderate binding reactivity for foamy macrophages, but were not specific for hepatocytes. The PNA lectin showed greater binding reactivity in the kidney of the sheep. Lectin histochemistry evaluation had not yet been performed in liver and kidney of sheep with photosensitization caused by Brachiaria spp.
Descreve-se um surto de intoxicação por monensina em ovinos no Estado do Rio de Janeiro, no qual de 180 animais, oito morreram após serem alimentados com ração contendo o ionóforo. A enfermidade, de evolução variável, caracterizou-se clinicamente por apatia, arritmia cardíaca, mioglobinúria, incoordenação, incapacidade de se levantar, decúbito esternal; uma ovelha abortou. As lesões macroscópicas consistiram de áreas pálidas no miocárdio, hidroperitônio, hidrotórax e edema pulmonar. O exame histopatológico evidenciou alterações degenerativo-necróticas no coração e na musculatura esquelética. No miocárdio, as lesões eram mais marcadas e caracterizavam-se por necrose multifocal com substituição das miofibras por tecido conjuntivo fibroso e inflamação intersticial mononuclear. Adicionalmente, verificaram-se proliferação de células satélite e reação inflamatória mononuclear em músculos esqueléticos. Ao que tudo indica, a adição excessiva de monensina sódica, talvez associada à homogeneização inadequada da droga ao alimento, tenha determinado a ingestão de grande quantidade de monensina por parte dos animais.
Monofluoracetato de sódio (MF) foi identificado, por cromatografia, em três das doze plantas que causam morte súbita em bovinos no Brasil, incluindo Palicourea marcgravii, a mais importante desse grupo. Uma lesão considerada típica por alguns autores para intoxicação foi¹ a degeneração hidrópico-vacuolar dos túbulos uriníferos contorcidos distais (DHV). O objetivo deste trabalho foi verificar se a ingestão de MF induz sinais clínicos e lesões similares às observadas nos bovinos intoxicados pelas plantas que causam morte súbita. Seis vacas receberam, por via oral, 0,5 e 1,0mg/kg de MF diluídos em 50mL de água destilada. Clinicamente, os animais apresentaram taquicardia, jugular repleta com pulso venoso positivo, respiração abdominal, ligeira perda de equilíbrio, por vezes cambaleavam, deitavam e apoiavam a cabeça no flanco. Na fase final (agônica), todos os animais caíam em decúbito lateral, esticavam os membros, faziam movimentos de pedalagem, apresentavam opistótono, nistagmo, mugidos e a morte ocorria em 2-14 minutos. À necropsia verificaram-se aurículas, jugulares, ázigos e pulmonares moderadamente ingurgitadas. Observaram-se ainda leve a moderado edema da subserosa nos locais de fixação da vesícula biliar no fígado, além de leve edema entre o duodeno e o pâncreas. O exame histopatológico revelou, em todos os animais, desde leve a acentuada DHV das células epiteliais dos túbulos uriníferos contorcidos distais associada à cariopicnose. Com relação à ultraestrutura, observou-se vacuolização do citoplasma devido ao acúmulo de água. Vacuolização e necrose de coagulação individual ou de grupos de hepatócitos e leve congestão hepática secundários à estase venosa também foram observados. DHV tem sido observada em casos de envenenamento por outras substâncias, porém nestes não está restrita aos túbulos distais e não se observa cariopicnose. Este estudo demonstra que a DHV dos túbulos renais de bovinos pode ser causada pelo envenenamento por MF e, por analogia, essa substância deve ser considerada como um dos fatores importantes, senão o mais significativo, implicado na morte dos animais que ingerem plantas que causam morte súbita no Brasil. Isso indica que estudos, que envolvam metabolização de MF por bactérias ruminais, teriam grande aplicabilidade econômica, uma vez que pelo menos 500.000 bovinos morrem anualmente intoxicados por plantas do grupo das que causam morte súbita no Brasil.
The clinical signs and postmortem features in 2 cows and 2 sheep experimentally poisoned with lethal doses (0.5 and 1.0 mg sodium monofluoroacetate) are described. Poisoned animals showed tachycardia, dyspnoea and slight loss of equilibrium as swaying and reluctance to stand. The cattle developed a prominent jugular pulse. In the final stages before death, cattle and sheep fell into lateral recumbency, with paddling movements of the legs. At postmortem examination both cattle and sheep had dilated and blood-filled auricles and jugular veins. The lungs were congested and oedematous. The cattle also developed slight to moderate subserosal oedema at fixation sites of the gall bladder to the liver and slight oedema of the mesentery between the duodenum and pancreas. The histological examination revealed hydropic-vacuolar degeneration of the distal convoluted uriniferous tubules (HVDDT) associated with nuclear pkynosis in all animals. One cow also developed moderate hepatocellular vacuolation. This is an identical lesion to that observed after the ingestion of Palicourea marcgravii and the other sudden death-causing plants in Brazil.
As folhas recém-colhidas de Jatropha gossypifolia (Euphorbiaceae) foram letais para ovinos em administrações únicas de 40g/kg. A dose de 5g/kg não causou sintomas de intoxicação; as doses intermediárias provocaram a morte de parte dos animais. A evolução da intoxicação foi de 6 a 22 dias. O quadro clínico-patológico nos ovinos experimentais era caracterizado por perturbações digestivas, pulmonares, cardíacas e ainda alterações regressivas leves somente evidenciadas através de exames histológicos, hepáticas e renais. Esses achados são semelhantes aos observados em experimentos com as sementes de Jatropha curcas em caprinos, ovinos e bezerros, e com os com os frutos e as folhas de Jatropha glauca e Jatropha aceroides em caprinos, realizados por outros autores. Estas comparações indicam que, independentemente das espécies, as folhas de Jatropha spp. devem conter compostos semelhantes aos encontrados nas sementes.
