Spontaneous and experimental poisoning with the swainsonine-containing and calystegine-containing plant Ipomoea carnea subsp fistulosa is described. Three of 8 goats presenting with emaciation, weakness, symmetrical ataxia, posterior paresis, proprioceptive deficits, abnormal posture, abnormal postural reaction, and muscle hypertonia were necropsied. I fistulosa was suspected to be the cause of the neurologic disease in all cases. An experiment was conducted to confirm the diagnosis using 12 goats and diets containing 3 different concentrations of the plant. All goats fed I fistulosa developed neurological signs that were similar to those observed in the spontaneous intoxication. Muscle atrophy and pallor were the only macroscopic changes observed in spontaneous and in experimental intoxication. Histological lesions of spontaneous and experimental animals were similar. The most prominent lesion was cytoplasmic vacuolation in neurons of the central and the autonomous nervous system, pancreatic acinar cells, hepatocytes, Kupffer cells, follicular epithelial cells of the thyroid gland, and macrophages of the lymphatic tissues. Neuronal necrosis, axonal spheroids formation, and astrogliosis were additionally observed in the brain. Ultrastructurally, the cytoplasmic vacuoles consisted of distended lysosomes surrounded by a single-layered membrane. Nonreduced end-rests or sequence of α-Man, α-Glc, β(1–4)-GlcNAc, and NeuNAc on lysosomal membrane were revealed by lectin histochemistry. Samples of plants used in the experimental trial contained swainsonine and calystegine and their intermediary derivate. We conclude that I fistulosa induces a glycoprotein storage disease primarily based on the inhibition of the lysosomal α-mannosidase by the alkaloid swainsonine.
Although there is solid knowledge on mineral deficiencies and their consequences on health and productivity of cattle in Brazil, paradoxically, those informations, most of the time, are not used by professionals of the agricultural sciences, who persist in stimulating the widespread use of the so-called complete mineral mixtures. The deficiency of many minerals included in these supplements, as iron, chrome, sulfur, and others, do not occur under natural conditions or only in rare and very particular situations. The present paper deals with the misunderstandings and problems related to traditional mineral supplementation and discusses the main aspects of the so-called selective mineral supplementation that is based in supplying exclusively the deficient mineral(s) and these only in necessary amounts. This procedure can result in expressive economy (sometimes up to 700%) when compared with the commercial mineral supplementation usually employed. According to several estimates, mineral supplementation can account for 20 to 30% of the total cost of beef cattle production on tropical pastures. A reversion assay, in which a group of cattle receives the commercial mineral mixture routinely used on the farm, and another group which receives the selective supplement, is the best option for the elucidation of positive or negative effects between the two possibilities of mineral supplementation on a specific farm. Trough this assay, the effects of the two compositions of mineral supplements can be investigated rationally over a sufficient period of time, with no risks and minimal possibilities of economic losses.
RESUMO.-[Aspectos histopatológicos da Hematúria Enzoótica Bovina no Brasil.] Com o objetivo de descrever alterações neoplásicas e não-neoplásicas ainda não relatadas e, paralelamente, reclassificá-las de acordo com nomenclatura mais completa e atual utilizada em medicina humana, foram estudadas, histologicamente, lesões da bexiga de 59 bovinos com Hematúria Enzoótica (HEB), oriundos dos Estados do Rio de Janeiro, São Paulo, Minas Gerais, Espírito Santo, Rio Grande do Sul, Santa Catarina, Paraná e Amazonas.Verificou-se, em termos qualitativos, quase uma perfeita identidade com as lesões de bexiga observadas em seres humanos. Comparações mais exatas com relação à freqüência dessas alterações ficaram prejudicadas, dadas a ocorrência de duas ou mais neoplasias em um mesmo animal e as diferenças da metodologia empregada ou do conceito de classificação. Coexistência entre neoplasias diversas, epiteliais e/ ou mesenquimais, foi vista com freqüência. Neoplasias ou diferenciações raras, ainda não descritas na bexiga de bovinos, como carcinoma trabecular com diferenciação em célu-las de Paneth, adenoma e adenocarcinoma mesonefróides, , were studied histologically. The objective of this study was to describe and reclassify neoplastic and non-neoplastic alterations not yet reported, according to the more complete current nomenclature used in human medicine. There was an almost complete identity with alterations observed in the bladder of man. Due to the occurrence of two or more neoplasms in the same animal, differences in the methodology and in the concept of classification, a more precise comparison was not possible. Coexistence of different types of epithelial and/or mesenchymal tumour growth was frequently seen. Rare neoplasms or differentiations not previously described were found in the bladder of some animals affected by BEH. These were trabecular carcinoma with Paneth cells differentiation, mesonephroid adenoma, mesonephroid adenocarcinoma, "signet ring" cell carcinoma, plasmocytoid carcinoma, chromophobe cell carcinoma and nested type of transitional cell carcinoma. Haemangiosarcomas originating from haemangiomas were also observed. This study also revealed the occurrence of many tumors with anaplasia and pronounced infiltrative features, but which did not metastasize. The elucidation of the cause of this "barrier against metastases" and its relationship with chemical carcinogenesis induced by the ptaquiloside, the active principle of bracken fern (Pteridium aquilinum), could be of interest to future research on the control of neoplasia in man and animals.INDEX TERMS: Histopathology, Bovine Enzootic Hematuria, bladder tumors, Pteridium aquilinum, Brazil.
A comparative study between cytology and histopathology of various neoplasms from 150 dogs was carried out. Several staining methods including Wright, May-Grünwald-Giemsa, NewMethylene Blue and Papanicolau, and histological staining methods such as Hematoxilin & Eosin, van Gieson, Sudan, Toluidine Blue and Periodic Acid Schiff were used. The data obtained indicated an 85.3% accuracy for the cytological diagnosis when the histopathological diagnosis was considered as the correct parameter. In 4.0% of the cases only the embrionary origin of the neoplasms was determined, and in 1.3% only the prognosis was established. The cytological diagnosis differed from the histopathological diagnosis in 8.1% of the cases. In two cases (1.3%), although the cytological and histopathological diagnoses differed, a reexamination determined that the former was correct which increased its accuracy up to 86.6%. Fine needle aspiration biopsy proved to be the best technique to obtain samples for the study. Cytology was found not suitable for the diagnosis of mammary neoplasms due to the wide variation of the cytological aspects from different areas. The imprint on slides is a technique not recommended to evaluate mesenchymal tumors and should be replaced by exfoliative cytology. The Wright method proved to be the more efficient staining method. Adapted staining methods from histopathology such as van Gieson for leiomyomas and leiomyosarcomas, Sudan Black B for lipomas and liposarcomas, and Periodic Acid of Schiff and Toluidine Blue for mastocytomas, were especially useful in visualizing the fine morphological details of various epithelial and mesenchymal neoplasms examined.
Foram examinadas amostras de fígado, rins, baço e linfonodos hepáticos, mesentéricos, retro-mandibulares, pré-escapulares e mediastínicos de 12 lotes em um total de 120 animais, envolvendo diferentes faixas etárias, todos oriundos do Estado de Mato Grosso. Os animais haviam sido mantidos em pastos onde Brachiaria decumbens e Brachiaria brizantha eram as forrageiras predominantes. Macroscopicamente o fígado desses animais mostrava coloração amarelada, mais evidente após 24 horas de fixação em formol a 10%. Nos linfonodos hepáticos e mesentéricos foram evidenciadas, na superfície de corte, estriações esbranquiçadas de forma radiada na cortical e medular com pequenas áreas brancas nodulares multifocais principalmente na medular. Em muitos casos, associado com essas áreas foram vistos focos de aspecto hemorrágico. Microscopicamente foram encontradas, no fígado, linfonodos hepáticos e mesentéricos, células com citoplasma espumoso, muitas das quais multinucleadas. Nos linfonodos hepáticos e mesentéricos estes infiltrados estavam associados a áreas de necrose e hemorragia. No fígado, as células de citoplasma espumoso estavam presentes em todo parênquima, de forma irregular, geralmente formando nódulos ao redor da veia centrolobular. Estas células não se coravam pelo ácido periódico de Schiff (PAS) e apenas fracamente na coloração de gordura pelo Oil Red O. Na ultra-estrutura as células de citoplasma espumoso apresentavam fendas, parcial ou totalmente delimitadas por membrana, que representam a imagem negativa de cristais, presentes também no citoplasma dos hepatócitos.
