Os insetos constituem uma das principais causas de danos à produção agrícola no mundo. O controle de insetos tem sido realizado por meio de agroquímicos e, em muito menor escala, pelo emprego de inseticidas biológicos. As plantas transgênicas resistentes a insetos representam uma nova alternativa no combate aos insetos-praga das lavouras. A bactéria entomopatogênica Bacillus thuringiensis Berlinier (Bt) é a fonte dos genes de resistência nas chamadas plantas-Bt, produzidas comercialmente. No presente trabalho de revisão, são abordados os aspectos relacionados à bactéria Bt como fonte de genes de resistência a insetos-pragas, plantas geneticamente modificadas, vantagens do uso de plantas-Bt, bem como perspectivas dessa ferramenta biotecnológica.
ABSTRACT. Ocimum basilicum L. (sweet basil), of the family Lamiaceae, is rich in aromatic essential oils and valuable for its pharmaceutical, aromatic and culinary properties. The present study describes the procedure for micropropagation of O. basilicum using cotiledonary leafs from in vitro geminated plants. Cotyledons from in vitro germinated seeds were used as initial explants, put in MS (Murashige and Skoog, 1962) ). Higher BAP concentrations induced an increase in the number of explants with shoots and a higher number of shoots/cotyledon.
Conservation of castor bean pollen at low temperaturesThe objective of storing pollen is to preserve material for future use, providing optimal conditions to maintain their original germination, vigor and genetic integrity. The aim of this study was to investigate the germination of castor bean (Ricinus communis L.) pollen after storage at low temperatures. For this purpose, pollen of the cultivars IAC 80 and AL Guarany 2002 were used and stored at four different temperatures, -196; -72; -18; and 4°C, for a period of 60 days. The pollen viability was assessed weekly for five weeks, with a last evaluation performed after eight weeks of storage using the in vitro germination test. Culture medium containing 10 gL -1 agar, 100 gL -1 sucrose, 0.004 g L -1 boric acid, pH 6.0 was prepared and poored into cavity slides, where the pollen was evenly distributed over the surface of the medium. The plates were incubated at 20°C in BOD. The experiment was arranged in a 2x4 factorial completely randomized design (2 cultivars x 4 temperatures) for each storage time, with 100 pollen grains analyzed per repetition, totaling six of each treatment. There was significant difference between treatments for the cultivar x temperature interaction in the fifth week of observation (p<0.05). Other cultivar x temperature interactions were highly significant at all times of observation (p<0.01). It is therefore concluded that the use of ultra low temperatures allows the maintenance of castor bean pollen viability until the fifth week of storage.
RESUMO Agentes químicos produzidos em algumas plantas podem provocar alterações no desenvolvimento de outras plantas ou até mesmo de outros organismos. Ensaios laboratoriais podem demonstrar os resultados destas interações, porém, sem considerar a ação do ambiente. Com este trabalho objetivou-se identificar a influência de extratos vegetais de 3 espécies conhecidas no Brasil popularmente como boldo sobre o desenvolvimento de Allium cepa. Os bioensaios foram conduzidos em germinador a 25° C, com extrato aquoso obtido por infusão de folhas frescas na concentração de 30 g.L-1 e água destilada como controle. Os testes de primeira contagem e germinação, que avaliam efeito alelopático foram realizados com 4 repetições de 50 sementes. Para determinação da citotoxicidade, através do índice mitótico (IM) foram contadas pela técnica de varredura 2.400 células por extrato. Os bioensaios realizados revelaram que os extratos vegetais interferiram sobre a germinação e sobre a divisão celular em células meristemáticas radiculares de A. cepa. Sementes expostas ao extrato de Vernonia condensata apresentaram as maiores divergências em relação ao grupo controle.
O objetivo deste trabalho foi avaliar os efeitos da radiação gama (Co60) na qualidade fisiológica de sementes de arroz. O estudo foi realizado em três ensaios. No primeiro ensaio, sementes de arroz foram irradiadas com doses de 0; 1; 2,5 e 5Gy e, para o segundo e o terceiro ensaio, as sementes foram submetidas a envelhecimento acelerado, antes da irradiação. No segundo ensaio, as sementes foram dividas em úmidas e secas após envelhecimento acelerado e, então, foram irradiadas nas doses 0; 1; 2,5 e 5Gy. No terceiro ensaio, as sementes foram secas após o envelhecimento acelerado e irradiadas com doses de 0, 10, 25 e 50Gy. Para avaliação dos efeitos fisiológicos da radiação gama, foram realizados teste de germinação - TG, índice de velocidade de germinação - IVG e crescimento de plantas (comprimento de parte aérea e sistema radicular e massa seca total), em todos os ensaios. Foram determinadas as atividades enzimáticas da fosfatase ácida e alfa amilase nas sementes secas, no segundo ensaio. Os resultados obtidos nos ensaios indicam que as doses de radiação gama estudadas não afetam a qualidade fisiológica das sementes de arroz.
Pereskia aculeata is a Cactaceae plant with valuable nutritional properties, including terrific amounts of protein, minerals, vitamins, and fiber. However, P. aculeata is reported to contain antinutrients and alkaloids in its leaves. In addition, in a study on growth and development, Wistar rats fed with P. aculeata and casein as protein source grew less than the control group (fed with casein only). Therefore, in this study, we evaluated, for the first time, the oral acute toxicity of P. aculeata in rats and also the cytotoxicity behavior of the plant on lettuce seeds. The acute toxicity research was carried out using dried P. aculeata ethanolic extract, in three different doses, administered by gavage to 24 female Wistar rats. The rats were then examined for signs of toxicity, food intake, body weight, and fecal excretion fluctuations, as well as histopathological alterations, using eight different body tissues. The acute toxicity study did not show any difference among the groups in either clinical evaluation or histopathological analyses. For the cytotoxicity study, dried P. aculeata ethanolic extract was applied on lettuce seeds in five different concentrations. These seeds were evaluated for germination, root and shoot length, and mitotic index. The results show that P. aculeata extract affects lettuce root and shoot growth, but not germination or mitotic index. In conclusion, the acute toxicity on rats and the cytogenotoxicity on lettuce of P. aculeata are neglectable, validating the potential of this plant to be used as a functional food.
