Objective. The aim of the present study was to investigate the presence of inflammatory mediators in rats with only periodontitis and periodontitis in a setting of hyper- and hypo-thyroidism and to analyze the correlative linkages between inflammatory mediators and thyroid hormones. Methods. White male 12–14 weeks old inbred rats (n=48) weighing 180–200 g were employed in the experiment. They were randomly divided into the following groups: Group I – control group, Group II – group with a model of periodontitis, Group III – group with a periodontitis in a setting of hyperthyroidism, and Group IV – group with periodontitis in a setting of hypothyroidism. The presence of tumor-necrosis factor-α (TNF-α) and interleukins IL-1β and IL-10 in the periodontal homogenate supernatant was studied by a solid-phase enzyme-linked immunosorbent assay. Results. It was shown that experimental lipopolysaccharide (LPS)-induced periodontitis is accompanied by hyperproduction of pro-inflammatory cytokines (TNF-α, IL-1β) and reduction of anti-inflammatory cytokines (IL-10), whereas TNF-α underwent to maximum changes. Thyroid dysfunction exacerbates cytokine imbalance and severity of inflammation in experimental LPS-induced periodontitis, especially pronounced at hyperthyroidism, as evidenced by the predominance of TNF-α and IL-1β levels in the periodontal homogenate supernatant by 38.5% (р<0.01) and 75.6% (p<0.001), respectively, hyperthyroid over the euthyroid, and by 20.1% (p<0.05) and 24.1% (p<0.05), respectively, over the hypothyroid rats. Conclusions. Thyroid dysfunction, especially hyperthyroidism, may play an important role in the pro-inflammatory response in periodontitis. Hyperproduction of inflammatory mediators in thyroid dysfunction can induce a noticeable damage in the whole apparatus of the periodontium, thereby causing progression of periodontitis.
L'influence des hormones thyroïdiennes sur les mécanismes mitochondriaux de l'apoptose des neutrophiles sanguins en cas de parodontite expérimentale L'objectif de l'étude. Cette étude vise à étudier le rôle des mitochondries dans l'apoptose des neutrophiles sanguins en cas de parodontite associée à une hyper et une hypothyroïdie chez le rat. Méthodes. Des études expérimentales ont été menées sur des rats blancs, non lignés et blancs, âgés d'environ 4 mois. L'analyse des échantillons de cellules pour déterminer les espèces d'oxygène réactives a été évaluée par la méthode de la cytométrie au laser en flux, en utilisant du diacétate de 2,7-dichlorodihydrofluorescéine. Le pourcentage de neutrophiles à faible potentiel mitochondrial transmembranaire et le pourcentage de neutrophiles apoptotiques ont été évalués par la méthode de la cytométrie au laser en flux, à l'aide de kits spécifiques.
The article presents the results of study effect of quercetin on the indicators of endogenous intoxication syndrome under conditions of experimental periodontitis. One of the key links in the pathogenesis of any inflammatory disease, including maxillofacial area, is endogenous intoxication, an important pathophysiological mechanism of which is the activation of lipid peroxidation. The experimental animals were treated by intramuscular injections with water-soluble quercetin for 7 days. For further research the blood serum was selected in which was determined the content of middle molecular weight molecules and erythrocyte intoxication index. Excessive lipoperoxidation was accompanied by accumulation of peroxidation products and depletion of antioxidant reserves, which led to accumulation of toxic substances. That is why the study of the level of middle molecular weight molecules and erythrocyte intoxication index in simulated bacterial-immune periodontitis is a reliable criterion. Quercetin was able to reduce the expression of endogenous intoxication syndrome in experimental bacterial-immune periodontitis.
In the article was studied the effects of flavonol quercetin on indices of adaptive immune response in experimental animals on the 14th day of the experimental bacterial-immune periodontitis development. Indices of immune protection were determined by the relative number of lymphocytes with CD3+, CD4+, CD8+, CD19+, CD16+ and immunoregulatory index (CD4+ / CD8+) in intact animals and on the 14th day of inflammatory process development in periodontal tissues as well as the therapeutic effects of flavonol quercetin. As a result of the study, characterized changes associated with the activity of both the cell-mediated and humoral-immune response were found, both in the development of experimental periodontitis, and apply of flavonol. In particular, there was an increase in the animal’s blood relative amount of CD8+, CD16+ cells on the 14th day, and content of CD3+, CD4+, CD19+ was decreased. In this case, the immunoregulatory index (CD4+ / CD8+) as an important index of immunological activity was decreased. The apply of flavonol quercetin in the period development of bacterial-immune periodontitis animals functional activity of the T-cell line of the immune system was increased, as evidenced percentage increase of B- and T-cells due to T-helper cells decrease as well as T-killers content during this period of inflammatory reaction in the periodontal complex, in comparison with animals, which were not treated.
The aim: To study the value of C-reactive protein in the experimental animals blood serum with bacterial-immune periodontitis and its correction with quercetin. Materials and methods: Modeling of periodontitis was performed by the following method: after thiopental anesthesia (at a dose of 40 mg / kg intramuscularly) rats were fixed. A subcostal injection of 0.01 ml of egg protein with cultures of Streptococcus hemolytic and Staphylococcus aureus at a dose of 4 CFU was performed in the area of periodontal tissues of the lower incisor as an initiating inflammatory factor. To enhance the immune process, a complete Freund’s adjuvant was introduced into the animal’s hind limb at the same time. Results: Analysis of the results of the study of the content of C-reactive protein in the blood serum of animals with experimental bacteria and immune periodontitis, receiving injections of quercetin, showed a significant decrease by 1.31 times, compared with animals with this simulated pathology on the 14th day of the experiment without the use of flavonol. When comparing this indicator on the 14th day of development of experimental periodontitis with correction, it was found that it remained slightly higher than the indicators of the intact group of rats. Conclusions: The level of C-reactive protein in the blood serum of experimental animals is an important indicator of the immune-inflammatory response, which increases its activation of the inflammatory system. The administration of flavonoid quercetin for 7 days helps to reduce the level of C-reactive protein in the blood serum of animals with experimental bacterial and immune periodontitis.
