ИССЛЕДОВАНИЕ СЕРИЛ-тРНК СИНТЕТАЗЫ ИЗ ПЕЧЕНИ БЫКА ИММУНОХИМИЧЕСКИМИ МЕТОДАМИ Получены поликлональные аффинно очищенные антитела на колонке с серил-тРНК синтетазой 95 %-ной чистоты, иммобилизованной в качестве лиганда на BrCN-активированной агарозе. Исследована специфичность полученных антител, их влияние на ферментативную активность серил-тРНК синтетазы, а также межвидовой перекрест серил-тРНК синтетаз из различных организмов. Установлено, что межвидовой перекрест имеет место между серил-тРНК синтетазами различных представителей класса эукариот и отсутствует с прокариотическими ферментами, что свидетельствует в пользу предположения об отсутствии гомологии у антигенных детерминант серил-тРНК шнтетаз эукариот и прокариот.
Monospecific polyclonal antibodies (pAbs) against highly purified bovine seryl-tRNA synthetase (SerRS, EC 6.1.1.1) were prepared and their specificity tested. The interactions of pAbs with SerRS from different organisms were investigated by protein immunoblotting and ELISA methods. pAbs inhibit eukaryotic SerRS aminoacylating activity and exert no effect on SerRS activity from prokaryotes. Исследовано распределение серил-тРНК синтетазы (CepPC) в органах и тканях различных представителей позвоночных с использованием иммунохимических методов исследования. Показано, что наибольшее относительное содержание данного фермента отмечено в печени всех исследуемых представителей позвоночных (от рыб до человека). Обнаружено наличие высокомолекулярных весьма устойчивых комплексов CepPC с другими белками, сохраняющихся в лизатах эукариотических тканей даже в денатурирующих условиях. Введение. Различные органы и ткани высших эукариот отличаются степенью метаболизма и соответственно уровнями активности аминоацил-тРНК синтетаз, что может быть связано как с интенсивностью белкового синтеза, так и с-аминокислотным составом синтезируемых белков. Одна из аминоацил-тРНК синтетаз высших эукариот-серил-тРНК синтетаза (CepPC) печени быка (КФ 6.1.1.4)-исследована нами ранее [1]. Установлено, что CepPC-фермент типа а 2 с молекулярной массой (м. м.) около 120 000. Получены моноспецифические поликлональные антитела к CepPC печени быка методом аффинной хроматографии и исследованы некоторые иммунохимические свойства данного белка [2]. Иммунохимический подход позволяет получить не только качественно новую информацию о некоторых структурно-функциональных свойствах белков, но и прямо определить относительное содержание исследуемого белка, которое не во всех случаях совпадает с уровнем ферментативной активности. Известно также, что аминоацил-тРНК синтетазы функционируют в составе высокомолекулярных мультиферментных комплексов (так называемых кодосом), и компартментализация метаболических процессов в клетке играет важную роль в функционировании белоксинтезирующего аппарата [3]. Изучение распределения CepPC в органах и тканях высших эукариот также важно и с точки зрения поиска наиболее богатого данным ферментом органа, так как выделение и очистка синтетаз эукариот до гомогенного состояния-весьма трудоемкий процесс, и нахождение «суперпродуцента» облегчило бы эту задачу. Материалы и методы. Материалами исследования служили: печень, почки, мозг и сердце мыши; печень, почки, мозг, поджелудочная железа кролика; печень, гипофиз и поджелудочная железа быка; печень рыбы, печень курицы; печень и сердце человека. Органы заморожены при-70 °С. CepPC печени быка выделяли по описанному методу [1]. Поликлональные антитела к CepPC печени быка получали по модифицированному методу Бейли, как описано ранее [2].
