Bovine pancreatic peptide (BPP) is a straight chain peptide containing 36 amino acid residues that has recently been isolated from pancreatic tissue. At a dose of 40 mug/kg-h intravenously, it stimulated gastric acid secretion when given alone but inhibited the submaximal secretion induced by the C-terminal pentapeptide of gastrin. Basal pancreatic secretion of dogs was inhibited by BPP (1-10 mug/kg-h) inhibited pancreatic protein secretion but often showed a biphasic action on water-bicarbonate response, an initial augmentation followed by reduction. BPP (2-5 mug/kg-h) inhibited pancreatic water-bicarbonate and protein secretions induced by an infusion of secretin plus cholecystokinin. Des-tyrosyl-NH2 BPP lacking the C-terminal tyrosyl amide, failed to inhibit gastric acid induced by C-terminal pentapeptide of gastrin or pancreatic secretion induced by secretin. BPP had no hyper- or hypoglycemic, hyperkalemic, or diuretic actions in the dog.

Many population-based epidemiological studies have unveiled an inverse correlation between intake of herbal plants and incidence of stroke. C. nutans is a traditional herbal medicine widely used for snake bite, viral infection and cancer in Asian countries. However, its role in protecting stroke damage remains to be studied. Despite of growing evidence to support epigenetic regulation in the pathogenesis and recovery of stroke, a clear understanding of the underlying molecular mechanisms is still lacking. In the present study, primary cortical neurons were subjected to in vitro oxygen-glucose deprivation (OGD)-reoxygenation and hypoxic neuronal death was used to investigate the interaction between C. nutans and histone deacetylases (HDACs). Using pharmacological agents (HDAC inhibitor/activator), loss-of-function (HDAC siRNA) and gain-of-function (HDAC plasmid) approaches, we demonstrated an early induction of HDAC1/2/3/8 and HDAC6 in neurons after OGD insult. C. nutans extract selectively inhibited HDAC1 and HDAC6 expression and attenuated neuronal death. Results of reporter analysis further revealed that C. nutans suppressed HDAC1 and HDAC6 transcription. Besides ameliorating neuronal death, C. nutans also protected astrocytes and endothelial cells from hypoxic-induced cell death. In summary, results support ability for C. nutans to suppress post-hypoxic HDACs activation and mitigate against OGD-induced neuronal death. This study further opens a new avenue for the use of herbal medicines to regulate epigenetic control of brain injury.

Lipids of Drosophila heads were extracted and separated by high-performance thin-layer chromatography. Fatty acid compositions of major phospholipids as well as of triglycerides were analyzed by gas-liquid chromatography. Proportions of the major fatty acids (14:0, 16:0, 16:1, 18:0, 18:1, 18:2, 18:3) varied depending on the lipid analyzed. Docosahexaenoic acid (22:6), common in vertebrate photoreceptors and brain, and arachidonic acid (20:4), a precursor of eicosanoids, were lacking. A comparison of the fatty acid composition of the diet vs. the head suggested that Drosophila can desaturate but may not be able to elongate fatty acid carbon chains. Fatty acid analyses were carried out after the following visual system alterations: i) the transduction mutant where no receptor potential results from a deficit in phospholipase C; ii) an allele of eyes absent; iii) the mutant outer rhabdomeres absent which lacks visual pigment and rhabdomeres in the predominant type of compound eye receptor, rhabdomeres 1 through 6; and iv) carotenoid deprivation which reduces opsin and rhabdomere size. We also evaluated aging by comparing newly-emerged vs. aged wild-type flies. Alterations in fatty acid composition based on some of these manipulations were found. Based on comparisons between flies reared on media differing in C16 and C18, there is an indication that diet readily affects tissue fatty acid composition.
