The CRISPR/Cas9 platform holds promise for modifying fish traits of interest as a precise and versatile tool for genome manipulation. To reduce introgression of transgene and control reproduction, catfish species have been studied for upscaled disease resistance and intervening of reproduction to lower the potential environmental risks of introgression of escapees' as transgenic animals. Taking advantage of the CRISPR/Cas9-mediated system, we succeeded in integrating the cathelicidin gene from an alligator (Alligator sinensis; As-Cath) into the target luteinizing hormone (LH) locus of channel catfish (Ictalurus punctatus) using two delivery systems assisted by double-stranded DNA (dsDNA) and single-stranded oligodeoxynucleotides (ssODNs), respectively. In this study, high knock-in (KI) efficiency (22.38%, 64/286) but low on-target was achieved using the ssODN strategy, whereas adopting a dsDNA as the donor template led to an efficient on-target KI (10.80%, 23/213). On-target KI of As-Cath was instrumental in establishing the LH knockout (LH--As-Cath+) catfish line, which displayed heightened disease resistance and reduced fecundity compared to the wild-type sibling fish. Furthermore, implanting with HCG and LHRHa can restore the fecundity, spawnability and hatchability of the new transgenic fish line. Overall, we replaced the LH gene with an alligator cathelicidin transgene and then administered hormone therapy to gain complete reproductive control of disease-resistant transgenic catfish in an environmentally sound manner. This strategy not only effectively improves the consumer-valued traits, but also guards against genetic contamination. This is a breakthrough in aquaculture genetics to confine fish reproduction and prevent the establishment of transgenic or domestic genotypes in the natural environment.
Diverse seasonal characteristics of hydro-chemical parameters in the coastal zone are significantly related to aquaculture development. In this paper, general water quality condition derived from laboratory analysis from the coastal waters of Salimpur, Chittagong is presented. Samples were collected from onshore and offshore site of two adjacent coastal locations named as North Salimpur (experimental location) and South Kattoli (control) during a monsoon and a dry season spanning 2013-14. The spatio-temporal variability of studied parameters were found as air temperature 26.5-32.5 ˚C, water temperature 23-33 °C, pH 7.1-7.9, DO 4.29-7.11 mg/L, BOD 1.10-3.25 mg/L, salinity 1.6-21 ppt, EC 3.40-35.68 mS/cm, TDS 2.02-21.99 g/L, TSS 0.62-2.76 g/L, transparency 4.5-14 cm, precipitation 64-1992 mm, NO2-N 1.94-2.58 µg/L, PO4-P 0.45-1.84 µg/L, SiO3-Si 130.46-956.31 µg/L during investigation period. Average values of physicochemical parameters were found to be in compliance with standard guidelines. The ship breaking activities near experimental location possess negative impacts on local geomorphology, freshwater inputs, precipitation and aquatic environment as well. Moreover, wind driven forces, tidal action, wave characteristics and changes in monsoon pattern regulate the coastal processes. This research suggests the importance of regular monitoring to assess present status of water quality and future prospect of aquaculture in the Chittagong coastal zone.
Sediment organic matter regulates the coastal biogeochemical cycle which is influenced by rainfall pattern and has potential relationship with organic carbon as well as mud characteristics. The objective of present research was to identify the impact of precipitation on the spatiotemporal variation inorganic features of intertidal mudflats. Three locations were selected in freshwater zone; two were in brackish water region and last three locations situated in marine waters from Halda to Salimpur coast. Two sites from each location were designated as the highest high tide level (onshore) and lowest low tide level (offshore) of the intertidal zone during winter (December to February) and monsoon (June to August) spanning between 2013 and 2014. Sediment organic matter and carbon were measured by combustion and Walkey-Black wet oxidation in turn. Average organic matter content in monsoon were found 4.5±0.03, 2.3±0.01 and 2.4 in freshwater, brackish and marine locations whereas 5.2±0.6, 3.9±0.14 and 5.4±0.04% investigated during winter. Precipitation pattern varies from different seasons and locations that have impact on land runoff, freshwater inflow, mixing and circulation. Mud dominated coastal intertidal zones represented high organic matter content than sand dominated coast. Furthermore, depositional pattern, transport, erosion-accretion processes, tidal action, wave characteristics and seasonal inconstancy control the organic matter characteristics in the coastal sediment. This research suggests the necessity of periodic observation of rainfall pattern and organic matter distribution to assess the intertidal deposits which support the stabilization of local geomorphology and biogeochemistry in Chittagong coastal region.Res.
