A pirin-like protein from a marine denitrifying bacterium, Pseudomonas stutzeri Zobell has been heterologously expressed in E. coli and purified to homogeneity with metal-affinity and gel filtration chromatographies. The recombinant pirin-like protein has exhibited quercetinase activities upon the incorporation of a divalent metal ion, while its biological role remains unclear. In the case of Cu2+ the holo-protein demonstrated the highest activities and spectroscopic properties typical of type II Cu protein. A 3D-structual model constructed using the crystal structure of human pirin as temperate indicated that the metal biding site is constructed with 3His1Glu located in the consensus sequences in the N-terminal domain.
TK Wiji is a nursery school which was built at Jl. Untung Suropati no. 10 kelurahan Tegalrejo kota Pekalongan based on NPSN 69879295 of 2005 by Yayasan 99. TK Wiji is growing fast due to its education system which is in line to the 2013 national curriculum with the vision of toodler characteristic development. The most fundamental of character education is environment education. Considering to this situation, we have found that the most common litter of the nursery is disposable diaper. This kind of diaper is hardly degraded into its monomer. Colon’s research in 2013 has found a strategy to use this diaper as compost. Based on <strong>the problem of disposable diaper wastes</strong> in the nursery and also the result of the background research about <strong>exploiting the waste as compost</strong>, then the environment education for toodler of TK Wiji by using diaper waste as a material for hidroponic cultivation was done. In the beginning, guidances were given to the teacher in 2 phases which are making diaper waste composite into organic fertilizer and making hidrogel for interior plant cultivation by using the gel of the waste. In the next step, the teachers were asked to make improvisation regarding toodler environment education by using the skills. In spite of this, there was still an argument about the time management in the learning process by using diaper waste as the material. Thus an implementation of the skill for toodler extracuricular was stated.
Tanaman Obat Keluarga (TOGA) dikenal sebagai apotik hidup merupakan tanaman yang ditanam di halaman rumah. Penanaman TOGA di pekarangan warga RT02/RW05 Sukorejo, Gunungpati, Semarang selama ini belum terlaksana dengan baik. Oleh karena itu dilakukan pengabdian ini untuk meningkatkan pengetahuan dan keterampilan kader PKK dalam identifikasi dan pemanfaatan TOGA melalui penyuluhan, pelatihan dan pendampingan TOGA. Khalayak sasaran adalah kader PKK RT 02/RW05 dan ibu-ibu yang dipilih karena memiliki kemampuan lebih dalam menjalankan kegiatan PKK dan pandai mendesiminasikan hasil pengabdian pada khalayak yang lebih luas. Kegiatan berlangsung selama 6 bulan meliputi persiapan, pelaksanaan, monitoring dan evaluasi meliputi a) penyuluhan potensi tanaman rempah-rempah bagi kesehatan dan perlunya menanam rempah-rempah dalam bentuk Taman Tanaman Obat Keluarga (TOGA) di pekarangan, b). pelatihan bagi kader PKK RT02/RW 05 tentang cara menaman TOGA di lahan minimalis dan c) pelatihan mengolah hasil pasca panennya menjadi berbagai minuman yang berkhasiat bagi kesehatan. Hasil dari kegiatan ini adalah meningkatnya pengetahuan dan keterampilan kader PKK RT 02/ RW 05 tentang pengelolaan dan pemanfaatan taman TOGA, terwujudnya taman TOGA di masing-masing perkarangan halaman depan rumah, serta kemandirian dalam pemenuhan pengobatan tradisonal. Keberlanjutan program ini diharapkan dapat dilakukan dengan memanfaatkan pekarangan rumah sebagai lahan menanam TOGA, sehingga pemanfataan TOGA untuk kesehatan dapat dilakukan secara berkesinambungan dan berdampak pada sosial ekonomi masyarakat.
Elaeocarpus grandiflorus L. is an identity plant of Salatiga City. Wherein, the plant is difficult to naturally propagation. This study would obtain propagation technique of E. grandiflorus by tissue culture. It was experimentally conducted, consists of two sub-experiments. The first aimed to find the optimal sterilization agent was conducted with a completely randomized design, namely sterilizing compound. There were two compounds studied, i.e., NaOCl and Ca(OCl)2. The second sub-experiment aimed to determine the optimal medium type and antioxidant compounds for inducing in vitro shoots. It was conducted with a completely randomized design with two factors, namely the medium type [i.e. Murashige & Skoog (MS) and Wood Plant Medium (WPM)], and antioxidant agents (i.e. activated charcoal and polyvinyl pyrrolidone/PVP). Data were analyzed by descriptive percentage, analysis of variance, and least significant difference test. The results showed that Ca(OCl)2 and NaOCl were effective to obtain the sterile and alive explants. The MS media added by PVP resulted in the most explants forming shoots, and WPM supplemented with PVP or active charcoal caused most explants regenerating callus. The results could be used as a basis for the development of in vitro shoot multiplication to reach an efficient propagation of E. grandiflorus.
